PTPN2 in the Immunity and Tumor Immunotherapy: A Concise Review
Round 1
Reviewer 1 Report
The authors make an exhaustive review of PTPN2 in the immunity and tumor immunotherapy. The article is very well written and well documented.
I recommend reviewing the paging of references that is not homogeneous: for example
[37] J Cell Mol Med 23(6) (2019) 4179-4195.
[42] J Autoimmun 53 (2014) 105-14.
[44] J Clin Invest 121(12) (2011) 4758-74.
[47] Nat Immunol 20(10) (2019) 1335-1347.
Besides, the manuscript needs a summary table of the PTPN family members. The authors should cite this article: Tang X, Qi C, Zhou H, Liu Y. Critical roles of PTPN family members regulated by non-coding RNAs in tumorigenesis and immunotherapy. Front Oncol. 2022 Jul 26;12:972906. doi: 10.3389/fonc.2022.972906 and build a summary table of the PTPN family. In this article Tang X et al begin with a summary paragraph:' Protein tyrosine phosphatases (PTPs) are a wide class of enzymes that oppose protein tyrosine kinases (PTKs). PTPs can be categorized into four families based on the amino acid sequence of its catalytic structural domain, each with different substrate specificities. Of these, 17 intracellular non-receptor PTPs, belonging to the largest class I cysteine PTP family, are designated as PTPNs'
Thus, instead of going directly to talk about PTPN2, they should talk about its belonging to this family of intracellular non-receptor PTPs (PTPNs), which belong to the largest class I cysteine PTP family, are essential for the regulation of a variety of biological processes.

Reviewer 2 Report
In this review, the authors have described-/summarized the roles of PTPN2 for immunotherapy. In my opinion, this manuscript (ijms-1875849) provides a good overview in this field, and could be published after considering the following comments:
1. The roles and mechanisms of tumor exososmes containing PTPN2 in the immunoregulation. What will be difference between the exosomal PTPN2 and secreted PTPN2 in tumor microenvironment? A detailed description is required.
2. The roles and mechanisms of PTPN2 in cold and hot tumors? How PTPN2 expression can improve the immunotherapy of cold tumors? A detailed description is required.
