Round 1

Reviewer 1 Report

Dear Authors,

The topic of the article is great of interest. However your manuscript presents lots of criticalities:

Abstract: Abstract should be revised, rewritten and make it more fluent and appealing.

Keywords: To ensure a properly research in medical databases, use MeSH terms to find  appealing keywords that could be helpful in finding your article.

“hese inflammatory cells release lyso- 58 somal enzymes that cause tissue destruction leading to permanent, irreversible damage 59 or necrosis [5], requiring endodontic treatment with/without placement of intracanal me- 60 dicament for complete disinfection and prevention of post-endodontic pain [6,7]. Various 61 signalling events in the pulp stimulate various mediators, which play a vital role in the 62 progression of inflammation/or abrupt regression of the inflammatory process” More Citations are needed and authors should be more specific regarding this topic.

“LL-37 antimicrobial peptide is regulated by the CAMP gene. The known inducer of 73 CAMP gene expression is 1,25 dihydroxyvitamin D through vitamin D receptor binding 74 to the vitamin D responsive element at 500 bp upstream of the camp gene promoter [16]. 75 The first experiment conducted to prove that LL-37 peptide expression up-regulates to 76 fight against bacterial infection was using a cystic fibrosis xenograft model, where a 77 planned overexpression of LL-37 using an adenovirus vector was done [17]. The promis- 78 ing candidate for inducing LL-37 is 1,25-dihydroxyvitamin D3 through toll-like receptor 79 via TLR2/1- Vitamin D Cathelicidins (LL-37) pathway.” I would like to suggest the authors to insert a figure that could illustrate the entire pathway described in these sentences.

The present manuscript should follow the PRISMA guidelines as described into the author instruction.  Reviews: These provide concise and precise updates on the latest progress made in a given area of research. Systematic reviews should follow the PRISMA guidelines."

Please revise the introduction section make it more fluent.

Materials and Methods and Results sections are missing.

Figures are well presented.

Conclusion section must be entirely revised making it more appealing.

English spell revision is necessary.

References needs to be revised according to the author instruction. In some references it is presented not only the year and volume but also the issue. Please check if it is necessary.

Best Regards.

This study  assessed the role of cathelicidins (LL-37) peptides in dental pulp, its release and role in immuno-defense mechanism and reparative dentin formation. The review is well structured and brings interesting insights towards the theme. I suggest that, since one of the studies included in the search involves VEGF related factors, the authors should mention the mechanism of VEGF action in the dental pulp and how Cathelicidins help the control of VEGF. Otherwise the article reads well and deserves publication.
