This review focuses on enhancers during development and diseases. The author summarizes knowledge on enhancers with a focus on transcription factor and histone modification mediated generation of enhancer signatures during development and differentiation. Overall, this is a comprehensive review and the author brings up interesting points with respect to enhancer signatures and lineage priming. However, I do not recommend publication before the following issues are addressed:

  1. The review should be subjected to editing by a native English speaker. It seems that the current version has not even been subjected to spelling checks.
  2. The table is not helpful. It simply lists the title of publications that are presented in the review and that are already listed in the reference list. A graphic representation summarizing the major points of this review would be much more helpful.

2) I am extremely sorry that the table is not helpful, table is just collection of recent studies in this new area to help the researcher to get information here at a single platform. As per suggestion, I also added a schematic diagram about Enhancer biology. Again, I am very thankful for your valuable comments.

This review presents a useful summary of the relationship of enhancers to regulation of expression of controlling genes in development and disease. Much editing for English is needed.

It has some limitations in scope. It does not consider, in detail, the important roles of DNA methylation, open chromatin, and transcriptionally associating domains (TADs). Some of the listed references do mention these additional factors afftecting enhancer function.

2) I do agree with the reviewer that this review has limitations as we are not focusing on methylation status, open and closed chromatin and Topological associated domains. Because this short review only focuses on the role of enhancer that’s why we only stick around Enhancers. Our team will be planning to write a full complete review. Your suggestion is really helpful, and we will definitely consider this.
