Round 1

Reviewer 1 Report

Dear authors,

Discussion section should be improved. Authors should begin with a sentence that introduces what the paragraph is about, link their findings with those in the literature, and finish with what are the main points. The discussion section in parts is weak and rather the results are repeated and compared with previous studies. I suggest to the authors a more in-depth analysis of your own results. What are the mechanisms behind your findings?  The novelty and implication of your study should also be highlighted throughout this section.

The conclusions section should illustrate the mechanistic links of findings obtained under applied treatments. The conclusions should answer the hypothesis of your study and should focus on the implication of your findings. This section should highlight the novelty and implication of your study also.

Reviewer 2 Report

The research investigates a method for rapid regeneration and ex-situ conservation of Juglans regia L., an endangered species in Albania. The method involves the use of synthetic seeds containing shoot tips of four native walnut varieties. I have following suggestions that will be helpful in revising the MS.

1. As per the author claim, the status of the plant is endangered, if it is so, author needs to provide the link from the IUCN Red List of Threatened Species.

2. Please provide the reference for the claim in line 37-38.

3. Please provide the reference for the claim in line 48.

4. Figure 2 e&f; please provide clear photographs for planet regeneration from the synseed. It is hard to identify that these are arising from synthetic seed or not.

5. Figures are not satisfactory and need to be replaced (Figure 2 e-h).

6. Results are presented in well manner

6. Overall discussion section is good.

Reviewer 3 Report

Dear Authors

I think that your study will make useful contributions to the literature. However, in order for the manuscript to be published, some improvements need to be made. In this context, I have marked both my suggestions and some of my corrections on the manuscript, as well as stating some critical points below;

# The current study aimed to develop a short-term preservation protocol of Juglans regia L. using syhthetic seed technology. From the current title, it is perceived as if only synthetic seeds were produced and plants developed in vitro. My title suggestion for this study is as follows;

"Short-term conservation of Juglans regia L. via synthetic seed technology", this title is more suitable for the manuscript.

# There are many repetitions in the text. The text needs to be revised and checked from start to finish. In addition, a follow-up scheme should be created in the narration.

# Manuscript should be revised according to the writing rules of the journal. Particular attention should be paid to units.

# The image quality of the figures is very poor. If there are better ones, they should be used.

# All references should be reviewed in accordance with the journal's writing rules.

Kind regards

Round 2

Reviewer 1 Report

Dear Authors, please write the geographic coordinates where the seeds of walnut were collected in 2.1 of the Materials and Methods.

Specify the duration of  plantlets propagation in in vitro conditions  for 2.2.

Author Response

Dear reviewer,

We are very thankful for the valuable suggestions on the manuscript.

  1. The coordinates of natural habitats are added In section 2.1 (Materials and Methods).
  2. The duration of plantlets propagation on in vitro conditions In section 2.2. is specified

All the text has been revised and some improvements in the English language are done.

Thank you for your support!
