Round 1

Reviewer 1 Report

Comments and Suggestions for Authors

The review is interesting, but other then the english revision it will be useful to add a paragraph describing possible therapeutic strategies addressing inflammation in the better way to support healing.

An additional paragraph should include possible intervention strategies based on the use of biomaterials/stem cells.

Comments on the Quality of English Language

Moderate language revision could ameliorate the review.

Author Response

  2. An additional paragraph should include possible intervention strategies based on the use of biomaterials/stem cells.

We thank the reviewer for the comment. We have revised the whole manuscript and enhanced the scientific English quality. We have also added sections discussing inflammation in the context of a potential therapeutic target and support and promote healing. The newly added sections are on page 8, lines 284-326 (highlighted in red text).

Reviewer 2 Report

Comments and Suggestions for Authors

In this review, the authors have discussed the fracture healing process and the role of immune cells and their cytokines. Here, the authors have also discussed the role of acute and chronic inflammation in fracture healing. Both acute and chronic inflammation negatively affect the fracture healing process. They discussed the pivotal role of Hematoma in providing early fracture stability. Below are the comments that I would like to highlight and need authors to address them appropriately:

1.     The authors should shed more light on the adaptive immune cells with viz. i.e., T regulatory cells (mentioned in Fig. 3 but not included in theory), Th17 and B cell subtypes such as Bregs, etc..

2.     Figure 2 could be more elaborate with details matching the text.
