Community Structure Diversity of Endophytic Fungi in Cissampelos pareira from Different Habitats and Their α-Glucosidase Inhibitory Activity

Cissampelos pareira is a medicinal plant with the potential effect of treating diabetes, commonly used by the Dai people in southern Yunnan Province. However, the wild resources of C. pareira are currently scarce. Endophytic fungi are a natural component of medicinal plants, while also serving as important repositories for discovering active natural products. In this study, we focused on 2-year-old C. pareira plants cultivated in potted and non-potted conditions. The community structure of endophytic fungi in the roots, stems, leaves, and flowers of two cultivation methods of C. pareira was investigated by using high-throughput sequencing (HTS) and traditional culture methods. Through HTS, we discover that the richness and diversity of endophytic fungi in C. pareira are associated with its growth environment and plant tissues. The endophytic fungi richness of C. pareira showed significant differences between the two habitats. And significant differences existed in the diversity of root endophytic fungi of C. pareira compared to those in the stems, leaves, and flowers. Additionally, the richness of endophytic fungi in the stems showed significant differences from that in the roots, leaves, and flowers. The results obtained using traditional culture methods revealed 69 endophytic fungi strains, classified into 2 phylum, 4 classes, 11 orders, 23 families, and 69 genera. The fermentation products of the obtained strains were evaluated for in vitro α-glucosidase inhibitory activity, and the results demonstrated that 11 endophytic fungi strains exhibited an inhibition rate exceeding 80%. The above-mentioned study can provide a theoretical basis for a comprehensive understanding of the community composition and diversity of endophytic fungi in C. pareira. Full article

Cissampelos pareira is a plant known for its edible and medicinal properties, with bioactive potential in various applications. This study aimed to isolate and characterize pectin from C. pareira (PCP) and assess its safety, as well as its efficacy in skin moisturizing and anti-aging effects on human volunteers. The highest yield of PCP extraction was achieved after a 45 min extraction period. Chemical composition analysis using Fourier transform infrared (FT-IR) spectroscopy confirmed the presence of a characteristic pectin structure. Optimized PCP samples revealed high methoxy pectin content, degree of esterification, and galacturonic acid concentration, along with consistent physicochemical properties. PCP exhibited antioxidant activity and showed no cytotoxicity at 1 mg/mL, and effectively reduced hydrogen peroxide-induced oxidative damage in NIH/3T3 cells. Stability assessment indicated that PCP maintains its physical appearance and total polysaccharide content after heating–cooling periods. None of the PCP formulations caused skin irritation among volunteers. In an 8-week clinical trial, subjects who used serum formulations containing 0.5% w/w and 1.0% w/w PCP showed significant improvements in skin hydration and elasticity compared to the placebo serum group. While skin smoothness, scaliness, and wrinkling also improved, these anti-aging results were not statistically significant. The results indicated PCP is a promising candidate for skin care applications due to its safety, stability, antioxidant activity, and demonstrated efficacy in enhancing skin hydration and elasticity. Dengue fever, a mosquito-borne viral infectious disease caused by the dengue virus, is a significant global health concern, especially in tropical and subtropical regions. Despite preventive efforts, Sri Lanka faces recurring dengue outbreaks, with the Western province being the most affected. Current treatments primarily focus on supportive care, as specific antiviral therapies remain elusive. This review presents an overview of dengue, its clinical presentations, the dengue burden in Sri Lanka, and the potential of Sri Lankan medicinal plants used in traditional medicine for treating dengue. Several plants, such as Munronia pinnata, Azardirachta indica, Cissampelos pareira L., Carica papaya, Zingiber officinale, Curcuma longa, and Bambusa vulgaris, show antiviral properties against dengue. The utilization of these medicinal plants in dengue treatment could offer a promising avenue for further research and drug development. Full article

Three new polyhydroxylated oleanane triterpenoids, cissatriterpenoid A−C (13), along with one known analogue (4), were isolated from the whole plant of Cissampelos pareira var. hirsuta. Their chemical structures were elucidated by extensive spectroscopic data (IR, HR-ESI-MS, 1H-NMR, 13C-NMR, DEPT, 1H-1H COSY, HSQC, HMBC, NOESY) and the microhydrolysis method. The isolation of compounds 14 represents the first report of polyhydroxylated oleanane triterpenoids from the family Menispermaceae. All isolated compounds were evaluated for their cytotoxicity against five human cancer cell lines, and the inhibitory activity against NO release in LPS-induced RAW 264.7 cells. Compound 3 showed the most potent cytotoxic activities against the A549, SMMC-7721, MCF-7, and SW480 cell lines, with IC50 values of 17.55, 34.74, 19.77, and 30.39 μM, respectively, whereas three remaining ones were found to be inactive. The preliminary structure–activity relationship analysis indicated that the γ-lactone ring at C-22 and C-29, and the olefinic bond at C-12 and C-13 were structurally required for the cytotoxicity of polyhydroxylated oleanane triterpenoids against these four cell lines. Based on lipid-water partition coefficients, compound 3 is less lipophilic than 1 and 4, which agrees with their cytotoxic activities. This confirms the potential of C. pareira var. hirsuta in the tumor treatment. Full article

Aphis craccivora Koch is a polyphagous and major pest of leguminous crops causing significant damage by reducing the yield. Repeated application of synthetic insecticides for the control of aphids has led to development of resistance. Therefore, the present study aimed to screen the insecticidal activity of root/stem extracts/fractions, and pure molecules from Cissampelos pareira Linnaeus against A. craccivora for identification of lead(s). Among root extract/fractions, the n-hexane fraction was found most effective (LC50 = 1828.19 mg/L) against A. craccivora, followed by parent extract (LC50 = 2211.54 mg/L). Among stem extract/fractions, the n-hexane fraction (LC50 = 1246.92 mg/L) was more effective than the water and n-butanol fractions. Based on GC and GC-MS analysis, among different compounds identified in the n-hexane fraction of root and stem, ethyl palmitate (known to possess insecticidal activity) was present in the highest concentration (24.94 to 52.95%) in both the fractions. Among pure molecules, pareirarineformate was found most effective (LC50 = 1491.93 mg/L) against A. craccivora, followed by cissamine (LC50 = 1556.31 mg/L). Parent extract and fractions of C. pareira possess promising activity against aphid. Further, field bio-efficacy studies are necessary to validate the current findings for the development of botanical formulation. Full article

The alkaloidal fraction (AFCP) of roots of Cissampelos pareira Linn. was screened for in-vitro antioxidant activity and immunomodulatory activity in mice. The HPTLC finger print profile was also established for the identification of AFCP which was found to contain 0.176 % of berberine. AFCP possess strong antioxidant activity which was revealed by its ability to scavenge the stable free radical DPPH, superoxide ion and to inhibit lipid peroxidation in rat liver homogenate induced by iron/ADP/Ascorbate complex. AFCP was found to have significant immunosuppressive activity at lower doses (25 and 50 mg/kg) while no activity was observed at higher doses (75 and 100 mg/kg). Humoral antibody titre was significantly (p<0.01) lowered by AFCP at the doses of 25 and 50 mg/kg. Delayed type hypersensitivity response was also significantly (p<0.01) suppressed by the AFCP at the dose of 75 mg/kg. Thus the present study revealed the immunosuppressive and antioxidant activities of the alkaloidal fraction of C. pareira roots. Full article
