Traditional fermented dairy products represent an important source of autochthonous microorganisms with potential applications in food biotechnology. This study aimed to isolate and characterize microorganisms from the traditional Kazakh fermented product kurt collected from different regions of the Abai area (Kazakhstan) and to evaluate their suitability for biotechnological applications in meat processing. Microbial isolation was performed using MRS medium under anaerobic conditions, followed by morphological and physiological characterization. Accurate identification was carried out using MALDI-TOF MS and 16S rRNA gene sequencing. The results showed that microbial counts ranged from 106 to 108 CFU/g, confirming high microbial diversity of kurt. MALDI-TOF MS analysis revealed the presence of Pichia fermentans, Enterococcus faecalis, Leuconostoc mesenteroides, and Lactobacillus helveticus, indicating that MRS medium supports the growth of both lactic acid bacteria and accompanying microbiota. Subsequent molecular analysis confirmed Leuconostoc mesenteroides and Lactobacillus helveticus as the most promising strains. These isolates demonstrated tolerance to salt, acidic conditions, and mesophilic temperatures, which are essential for meat fermentation processes. In contrast, Enterococcus faecalis was excluded from further application due to potential safety concerns. Overall, the study demonstrates that kurt is a valuable source of technologically important microorganisms and that the identified strains (Leuconostoc mesenteroides and Lactobacillus helveticus) are promising candidates for the development of starter cultures for fermented goat meat processing. This study aimed to evaluate the chemical composition of milk from Kazakh mares kept under free-grazing conditions on natural pasture without supplementary feeding. Fifty Kazakh mares were aged 3 to 13 years, kept in herds, and categorized by age, number of foalings, and lactation stage for statistical analysis. Milk samples were collected, chilled, and analyzed on the same day using a Milkoscan FT2. The composition of fat, protein, lactose, total solids, casein, and other components was examined. Statistical analyses, including ANOVA, PCA, CDA, and Pearson correlation analysis, were performed using JMP 17 Pro to assess the effects of lactation stage, mare age, and foaling number and to identify key relationships. The results showed that milk composition was significantly influenced by these factors. Milk from the late-lactation stage contained higher protein, lactose, and total nutrients, while milk from the early-lactation stage had a higher sugar content and acidity. Older mares and those with higher foalings produced milk with higher concentrations of glucose and galactose, while younger mares and those with fewer foalings produced milk with higher fat. A key aspect of this experiment was that all mares grazed exclusively on natural pastures without supplementation, allowing for an objective assessment of their inherent productive potential. These findings deepen the understanding of mare milk composition dynamics in Kazakh breed horses under extensive management, supporting its potential for organic dairy production. Full article

The emergence of antibiotic-resistant pathogens in clinical settings has intensified the search for new probiotic strains with both health benefits and technological utility. This study aims to identify and characterize promising antimicrobial cultures derived from milk and dairy products, capable of inhibiting opportunistic pathogens. The samples of dairy products were collected from various markets across Kazakhstan. Microorganisms isolated from these samples underwent identification through 16S rRNA and ITS gene sequencing, using the BLAST algorithm. Their antimicrobial activity was assessed using the delayed antagonism method against pathogenic microorganisms including E. coli, S. aureus, Pseudomonas sp., Candida sp., and B. subtilis. Additionally, the isolates were evaluated for resistance to environmental stress factors such as temperature, pH, salt, ethanol, glucose, and peroxide. From 24 distinct samples, 33 isolates were purified, with 15 demonstrating high viability (108–109 CFU/mL) and stress resistance. Notably, Lacticaseibacillus casei AK and Enterococcus faecium KS exhibited resistance to all tested stress conditions. Antimicrobial screening revealed strong activity by strains LP, LB, and S-2 against multiple pathogens. Genotyping and carbohydrate fermentation tests identified these effective isolates as belonging to the genera Lactobacillus, Lactococcus, Enterococcus, Lactiplantibacillus, Streptococcus, and the yeast genus Pichia. This study underscores the industrial and health potential of the identified microorganisms. Prominent among the strains, Lactiplantibacillus pentosus LP, Lacticaseibacillus casei AK, Lactiplantibacillus argentoratensis LB, Lactiplantibacillus plantarum S-2, and Enterococcus faecium KS have been recognized as potent probiotics. These strains exhibit broad-spectrum antimicrobial activity coupled with robust stress tolerance, making them suitable candidates for probiotic applications. Full article

The Asian Central Steppe, consisting of current-day Kazakhstan and Russia, has acted as a highway for major migrations throughout history. Therefore, describing the genetic composition of past populations in Central Asia holds value to understanding human mobility in this pivotal region. In this study, we analyse paleogenomic data generated from five humans from Kuygenzhar, Kazakhstan. These individuals date to the early to mid-18th century, shortly after the Kazakh Khanate was founded, a union of nomadic tribes of Mongol Golden Horde and Turkic origins. Genomic analysis identifies that these individuals are admixed with varying proportions of East Asian ancestry, indicating a recent admixture event from East Asia. The high amounts of DNA from the anaerobic Gram-negative bacteria Tannerella forsythia, a periodontal pathogen, recovered from their teeth suggest they may have suffered from periodontitis disease. Genomic analysis of this bacterium identified recently evolved virulence and glycosylation genes including the presence of antibiotic resistance genes predating the antibiotic era. This study provides an integrated analysis of individuals with a diet mostly based on meat (mainly horse and lamb), milk, and dairy products and their oral microbiome. Full article
