Based on morphological and molecular data, Brotulella n. gen. is proposed to accommodate the dactylogyrid monogeneans Brotulella laurafernandae n. sp. (type species) and Brotulella luisahelenae n. sp. on the gill filaments of the Pacific bearded brotula Brotula clarkae Hubbs, 1944 (Ophidiiformes: Ophidiidae) from Puerto Pizarro in the Tumbes region (northern Peru). Species of the new genus are distinguished from all other dactylogyrids by the combination of the following features: (1) anchors with a stocking-shaped sheath associated with the distal end of superficial and deep roots, (2) tandem gonads, (3) a vas deferens looping left intestinal caecum, (4) a distally twisted male copulatory organ (MCO) with a delicate membranous accessory piece articulated to the shaft of the MCO, (5) a U-shaped ovary, (6) an almost sigmoid seminal vesicle, and (7) two prostatic reservoirs with thick muscular walls. A 28S ribosomal DNA-based phylogenetic analysis (Maximum likelihood and Bayesian inference) of sequences of two new species of Brotulella n. gen. from the Southeastern Pacific Ocean, along with sequences from closely related genera of the marine Dactylogyridae, supports the establishment of the new genus for the dactylogyrid parasites on the gills of the Pacific bearded brotula. The back cusk eel, Genypterus maculatus (Tschudi, 1846), (Ophiidiformes: Ophiididae) is one of the benthic-demersal fish usually consumed in northern Peru. Here, we identified the third stage (L3) Anisakidae sampled from 29 specimens of G. maculatus captured off the south American Pacific coast, Lambayeque Region, Peru. A total of 20 anisakid nematode larvae were collected on the visceral surface and divided morphologically into three types (Type I–III). These larvae were identified by mtDNA Cox2 sequences analysis, which indicated that corresponded to Anisakis pegreffii Campana-Rouget and Biocca, 1955, Skrjabinisakis physeteris (Baylis, 1923) and S. brevispiculata (Dollfus, 1966) Safonova, Voronova, and Vainutis, 2021, respectively. This is the first record of S. brevispiculata in Peru. The results obtained in this study provide knowledge on the diversity and distribution of Anisakis Dujardin, 1845 and Skrjabinisakis Mozgovoi, 1951, species in the south American Pacific waters and their relevance for public health. In addition, we suggest that combined use of molecular and morphological approaches is needed to characterize L3 anisakid larvae. Full article

Environmental stressors, such as temperature, are relevant factors that could generate a negative effect on several tissues in fish. A key fish species for Chilean aquaculture diversification is the red cusk-eel (Genypterus chilensis), a native fish for which knowledge on environmental stressors effects is limited. This study evaluated the effects of high-temperature stress on the liver of red cusk-eel in control (14 °C) and high-temperature (19 °C) groups using multiple approaches: determination of plasmatic hepatic enzymes (ALT, AST, and AP), oxidative damage evaluation (AP sites, lipid peroxidation, and carbonylated proteins), and RNA-seq analysis. High-temperature stress generated a significant increase in hepatic enzyme activity in plasma. In the liver, a transcriptional regulation was observed, with 1239 down-regulated and 1339 up-regulated transcripts. Additionally, high-temperature stress generated oxidative stress in the liver, with oxidative damage and transcriptional modulation of the antioxidant response. Furthermore, an unfolded protein response was observed, with several pathways enriched, as well as a heat shock response, with several heat shock proteins up regulated, suggesting candidate biomarkers (i.e., serpinh1) for thermal stress evaluation in this species. The present study shows that high-temperature stress generated a major effect on the liver of red cusk-eel, knowledge to consider for the aquaculture and fisheries of this species. Full article

Spatio-temporal variability of marine soundscapes reflects environmental dynamics and local habitat health. This study characterizes the coastal soundscape of the Cres-Lošinj Natura 2000 Site of Community Importance, encompassing the non-tourist (11–15 March 2020) and the tourist (26–30 July 2020) season. A total of 240 h of continuous recordings was manually analyzed and the abundance of animal vocalizations and boat noise was obtained; sound pressure levels were calculated for the low (63–2000 Hz) and high (2000–20,000 Hz) frequency range. Two fish sound types were drivers of both seasonal and diel variability of the low-frequency soundscape. The first is emitted by the cryptic Roche’s snake blenny (Ophidion rochei), while the second, whose emitter remains unknown, was previously only described in canyons and coralligenous habitats of the Western Mediterranean Sea. The high-frequency bands were characterized by bottlenose dolphin (Tursiops truncatus) vocalizations, indicating dolphins’ use of area for various purposes. Boat noise, however, dominated the local soundscape along the whole considered periods and higher sound pressure levels were found during the Tourist season. Human-generated noise pollution, which has been previously found 10 years ago, is still present in the area and this urges management actions. Full article

Chile has promoted the diversification of aquaculture and red cusk-eel (Genypterus chilensis) is one of the prioritized species. However, many aspects of the biology of the species are unknown or have little information available. These include intestinal microbiota, an element that may play an important role in the nutrition and defense of cultured animals for meat production. This study compares the microbiota composition of the intestinal contents of wild and aquaculture fish to explore the microbial communities present and their potential contribution to the host. DNA was extracted from the intestinal content samples and the V4 region of the 16S rRNA gene was amplified and sequenced using the Ion Torrent platform. After the examination of the sequences, strong differences were found in the composition at the level of phylum, being Firmicutes and Tenericutes the most abundant in aquaculture and wild condition, respectively. At the genus level, the Vagococcus (54%) and Mycoplasma (97%) were the most prevalent in the microbial community of aquaculture and wild condition, respectively. The evaluation of predicted metabolic pathways in these metagenomes showed that in wild condition there is an important presence of lipid metabolism belonging to the unsaturated fatty acid synthesis. In the aquaculture condition, the metabolism of terpenoids and polyketides were relevant. To our knowledge, this is the first study to characterize and compare the intestinal microbiota of red cusk-eel (Genypterus chilensis) of wild and aquaculture origin using high-throughput sequencing. The culture of red cusk eel Genypterus chilensis is currently considered a priority for Chilean aquaculture but low larval survival rates have prompted the need for the continuous use of antibacterials. The main aim of this study was to evaluate the role of live feed as a source of antibacterial-resistant bacteria in a commercial culture of G. chilensis. Samples of rotifer and Artemia cultures used as live feed were collected during the larval growth period and culturable bacterial counts were performed using a spread plate method. Rotifer and Artemia cultures exhibited high levels of resistant bacteria (8.03 × 104 to 1.79 × 107 CFU/g and 1.47 × 106 to 3.50 × 108 CFU/g, respectively). Sixty-five florfenicol-resistant isolates were identified as Vibrio (81.5%) and Pseudoalteromonas (15.4%) using 16S rRNA gene sequence analysis. A high incidence of resistance to streptomycin (93.8%), oxytetracycline (89.2%), co-trimoxazole (84.6%), and kanamycin (73.8%) was exhibited by resistant isolates. A high proportion of isolates (76.9%) carried the florfenicol-resistance encoding genes floR and fexA, as well as plasmid DNA (75.0%). The high prevalence of multiresistant bacteria in live feed increases the incidence of the resistant microbiota in reared fish larvae, thus proper monitoring and management strategies for live feed cultures appear to be a priority for preventing future therapy failures in fish larval cultures. Fatty acid composition and distribution in edible species of fish and shellfish captured in the South Pacific were studied, with a focus on n-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA). Fatty acids were quantified using gas-chromatography coupled with flame ionization detection (GC-FID), and the distribution of different fatty acids within lipid classes (neutral and polar lipids) was achieved after oil extraction using the Folch method and separation of lipid classes via solid-phase extraction for further GC-FID analysis. Red cusk-eel was the fish species with the lowest content of both EPA and DHA (40.8 and 74.4 mg/100 g, respectively) whereas mackerel contained the highest amount (414.7 and 956.0 mg/100 g for EPA and DHA, respectively). Sea squirt was the shellfish species with the highest content of EPA and DHA (375.0 and 165.7 mg/100 g, respectively) whereas the lowest amount of EPA + DHA was found in Chilean abalone (63.6 mg/100 g). PUFA were mostly found in neutral or polar lipids depending on the studied species. Indexes used to discuss the nutritional quality of lipids (PUFA/SFA, n-6/n-3 ratio and the hypocholesterolemic/hypercholesterolemic fatty acid index) were calculated and reported in the manuscript. This information provides a novel nutritional insight which may be useful to help nutritionists and other health professionals give more accurate counseling for the population to reach the recommended daily intakes of EPA and DHA.
