The majority of the information presented in this review paper is unrelated to the topic of bone marrow. It would be beneficial for the authors to conduct a more comprehensive analysis of the literature and focus on summarizing current research studies. Additionally, a section dedicated to providing details on the bone marrow environment and its components, as well as the challenges associated with developing bone marrow organoid models, would be valuable.

Author response: We thank our reviewer for their time and valuable feedback. As per your suggestion, we have now added more details on the bone marrow (section 2, lines 131-155). We recently published our article titled, ‘Aging, Bone Marrow and Next-Generation Sequencing (NGS): Recent Advances and Future Perspectives’ in which we have described the components and details of the bone marrow environment literature summary in great detail (https://www.mdpi.com/1422-0067/22/22/12225). To avoid the repetition of our previous paper - as both these papers discuss the bone marrow and NGS, we focussed our current manuscript on 3D models of the bone marrow instead.

Considering our article is titled focussing on the potential of the combination of 3D models and next generation sequencing, we have now highlighted recent research in the field as per your suggestion (lines 440-454, lines 520-523) as well as the challenges associated with developing bone marrow organoid models (lines 538-557, 561-565).

 

 

Additionally, we have added a sentence in the abstract (line 22), introduction (lines 98-100) and three references towards the future directions of our current article with the potential of organoids as a platform for precision medicine (line 565).

 

All the changes made are have been indicated in red in the revised version.

The authors nicely describe the current knowledge surrounding bone marrow culture conditions in 2D and 3D as well as advantages and disadvantages, respectively.

The only small concerns I have is the following:

Figure 1: 2D doesn´t reflect physiology in vivo…. is not correct. It does maybe reflect only very few aspects, but not none.

While microfluidic devices for bone marrow are mentioned in figures, no discussion or references are made. A paragraph discussing this extremely important field needs to be added! A broad variety of publications are available.
