This review by Halasa and colleagues faced the issue of target therapy in cancer, that is a very important issue. Debating the deacetylation of transcription factors and, in particular, the activity of deacetylation inhibitors as a potential therapy for cancer onset and progression is a very original contribution to the field.
The work is well-structured, clear and detailed in the explanation of the molecular pathways, with a huge and appropriate literature to support the central idea of the paper.
The images are understandable. Only in Figure 5, panel A and B are switched (see the legend).
In my opinion, this review paper is suitable for publication.

The review on the role of HDAC on transcription factor and their effect on caner progression is well written. The references are up to date and the authors have summarize them beautifully. I support the publication of the manuscript.
