Round 1

Reviewer 1 Report

The authors found KIF23 as a significant gene related to IPAH and pyroptosis, being reportedly its expression increased in pulmonary arterial smooth muscle cells 15 (PASMCs). I think this paper is interesting and well written, with interesting translational applications and implications, but I think methods should be clearly devided from results.

Dear Editors and Reviewers:

Thank you for your letter and for the reviewers’ comments concerning our manuscript entitled “Inhibition of KIF23 alleviates PAH by targeting pyroptosis and proliferation of PASMCs” (ijms-1668343). Those comments are all valuable and very helpful for revising and improving our paper, as well as the important guiding significance to our researches. We have studied comments carefully and have made correction which we hope meet with approval.  The main corrections in the paper and the responds to the reviewer’s comments are as following:

Response: Thank you very much for your suggestion. We have removed method-related content from the results, all changes have been annotated.

Reviewer 2 Report

-Extremelly novel research, highly qualitative.

-Very detailed analysis of all methods and matherials used to confirm the first hypothesis.

-IPAH is a very rare disorder but severe too with no cure so all therapies alleviates this condition raises our interest.

-Extremely promising the suggestion that KIF23 may be a new therapeutic target for IPAH by inhibiting the pyroptosis and proliferation of PASMCs.

-Very understandable, accurate scientific data and resources and detailed study.

-Very interesting topic for the scientific community.

-Very detailed analysis in an understandable way even for  those who are not familiar with genes therapies, cell proliferation and cell death, apoptosis or pyroptosis.
