Comment on Lai et al. Dipeptidyl Peptidase 4 Stimulation Induces Adipogenesis-Related Gene Expression of Adipose Stromal Cells. Int. J. Mol. Sci. 2023, 24, 16101
Round 1

Comments and Suggestions for AuthorsChen’s group reported that DPP4 stimulation with MCP-1 increases adipogenesis-related gene expression. While, Cordero et al. commented that these data were nor reflected the soluble-type DPP-4. However, Chen et al. did not commented about the involvement of soluble DPP-4 in this comment in their Reply. It should be commented about this involvement and possibility in their manuscript.

Comments and Suggestions for AuthorsThe commentary critically evaluates the methodologies, results, and conclusions of the original study in order to clarify, and expand on, the results presented.
The commentary is well structured and presents a clear and concise critique of the original article "Dipeptidyl Peptidase 4 Stimulation Induces Adipogenesis-Related Gene Expression of Adipose Stromal Cells".
The authors of the comment seems to have a good knowledge of the topic and raises several pertinent points.

Comments and Suggestions for AuthorsThis response by Hsiao-Chi Lai to Oscar J Cordero's comment is mostly a summary of their findings, but not an answer. Authors need to address the issues raised in the comment.
“Adipose tissue expands by either the hypertrophy of adipocytes from adipocyte precursor cells (APCs) or the differentiation of new adipocytes (adipogenesis).“ should be “
Adipose tissue expands by either the hypertrophy of adipocytes or the differentiation of new adipocytes (adipogenesis) from adipocyte precursor cells (APCs)”.
type 2 DM, please explain DM
“Monocyte chemoattractant protein-1 (MCP-1), known as chemokine (CC-motif) ligand 2 (CCL2), treatment induced macrophage cell division in adipose tissue, whereas MCP-1 deficiency in vivo decreased adipose tissue proliferation” this is unclear proliferation of macrophages, adipocytes?
“AdipoQ“ please use lower case letter. If authors refer to mRNA names have to be in italic.
“in DPP4 cells“ please write: DPP4-positive interstitial progenitor cells
“the increase of insulin-stimulated glucose in muscles and adipose tissue in mice” please include uptake, please indicate that this is only one of the multiple beneficial effects of adiponectin.
“This suggests that injection of MCP-1-supplemented plasma into adipose tissue induces adipogenesis-related gene expression in SVFs through the increase of PDGFRα mRNA expression in adipose tissue macrophages” please rewrite for clarity.
“is associated with and weight gain and Mice lacking the DPP4 gene were difficult to develop obesity after a high-fat diet” please correct
“We demonstrated that injection of MCP-1-supplemented plasma into adipose tissue of Leprdb/db mice increases PDGFRα mRNA expression in adipose tissue macrophages and most PDGFRα+ cells are macrophages.” And “We further demonstrated that the injection of MCP-1-supplemented plasma into adipose tissue of Leprdb/db mice increased PDGFRα mRNA expression in adipose tissue macrophages” give identical information. The last paragraph of this Comment is mostly a repetition.
