The Communication entitled: “3a-(4-Chlorophenyl)-1-methyl-3a,4-dihydroimidazo[1,5-a] quinazolin-5(3H)-one: Synthesis and in silico Evaluation as Ligand in the µ-Opioid Receptor”
by Andrea Defant, Nicole Innocenti, and Ines Mancini
reports the synthesis of the title compound with a noticeable 60 % overall yield owing the high yield and purity of the intermediates involved, and the consequent no need of their purification. Thus, the chemistry is fine, and the intermediates and the final compound are fully characterized. Moreover, the authors carried out an ‘in silico’ study of the novel compound as ligand in the µ-opioid receptor in comparison with the opioids morphine, fentanyl and BU72, that displayed favourable predicted blood-brain barrier permeation and drug-likeness.
Overall, I think this manuscript is well-written and appropriate for publication in Molbank, however, a minor revision is needed.
· At row 9 of ‘Results and discussion’ section: please correct the typo ‘transamidation-cyclizaion’ into ‘transamidation-cyclization’
· In both ‘Abstract’ and ‘Conclusion’ sections the synthesis is indicated to occur in 3 steps, but the overall steps are four, please correct.
