11-Hydroxy-7-Methoxy-2,8-Dimethyltetracene-5,12-Dione
Round 1
Reviewer 1 Report
Comments and Suggestions for AuthorsThe paper describes the isolation, characterization and the microbiological activity evaluation of a new reduced anthracycline.
The paper is well structured, the results are well presented and the conclusions are firmly based on the experimental data.
Despite that, there are some aspects to be discussed.
Lines 77-79: I would suggest to place the entire sentence about the IUPAC numbering before the 1H NMR section, in order to make the section clearer.
Line 66: The carbonyl carbon signals are at 188 and 182 ppm. Is it possible to explain the chemical shift?
Line 72: What does "chelated 12-OH" refer to? Is there any coordination bond?
Anthracyclines are largely used as anticancer drugs. Why was 4 not tested as a possible anticancer drug?
2.3 section: Why were more concentrated tests not carried out?
Line 183: I would suggest to add the information related to the experimental deuterated solvent and the sample concentration.
DEPT experiments are a very useful tool for organic compound structural elucidation. Were they carried out?
Comments on the Quality of English LanguageLine 44: I would suggest to write "Compound 4" instead of just 4 when a sentence begin.
