Round 1

Reviewer 1 Report

1. In the introduction section, how would cfDNA lipid biopsy help to solve the remaining challenges in the management of gastric and gastroesophageal junction cancers? Please indicate.
2. In section 2, which of the genetic alterations are related to cfDNA lipid biopsy? Please focus on the topics.
3. A list of abbreviations is necessary.
4. Figure 1 needs to be explained. What is >1% mean? Sensitivity over 1%?
5. How specific is the cfDNA lipid biopsy in the determination of tumor origin in gastric and gastroesophageal junction cancers?

Reviewer 2 Report

This is a well-written review article.

Major critiques:

  1. Authors should include a section on the biology of ctDNA and a discussion on tumor-informed vs tumor agnostic assay methods for MRD assessment.
  2. Authors should include a section titled 'Future directions'.
  3. A table summarizing the studies on tumor genomic profiling with liquid biopsy vs tissue biopsy should be included.

Minor comments-

  1. Fig1 legend should be included.
  2. A sentence should not start with the word 'Also'.  'Furthermore' may be the most appropriate word for such sentences.
