This review covers several key points for the reader interested in psilocybin and its potential clinical uses, adopting a neutral stance based on information and with a very limited amount of speculation. In this sense, the review is informative and potentially useful as a reference.

I have some comments aiming to improve the quality of this article:

  1. Please include citations in the table where the effects of psilocybin are listed.
  2. The authors should mention and discuss the recent work comparing psilocybin with excitalopram for tratment resistant depression: https://www.nejm.org/doi/full/10.1056/NEJMoa2032994
  3. I think a deeper discussion of the blinding issue and how to overcome it could strengthen the paper. See for instance: https://www.tandfonline.com/doi/abs/10.1080/17512433.2021.1933434 for inspiration.

There are several typos/mistakes that should be corrected.

  1. Ernest Spath first synthesized mescaline (not Spaff) - line 29
  2. "attntion", line 59
  3. "the psilocybin" instead of "psilocybin" in line 59
  4. "suicide, and jumping out of the window" in line 224 (jumping out of the window counts as suicide in my estimation)
  5. "teratogeni city" in line 259
  6. "informaion" in line 337
  7. "partecipants" in line 348
