The manuscript article by Wang et al. entitled "Genome-wide Characterization of NLRs in Saccharum spontaneum L. and Their Responses to Leaf Blight in Saccharum" is very scientifically sound research work based on the genome-wide analysis of nucleotide-binding site-leucine-rich repeat (NLR) receptor through genomics, phylogenetic and transcriptomic analysis with the help of Saccharum spontaneum L. genomic sequence and got a link that how NLRs expression can regulate the Stagonospora tainanensis mediated sugarcane leaf blight (SLB) disease.

The manuscript is written very well with huge molecular research works. The author pointed out the major molecular strategy of NLRs to combat with sugarcane leaf blight disease.

I have some specific points to the authors for corrections:

Line 11: Is 724 the total of allelic and non-allelic NLRs? Please check.

Line 22-23: 'The difference of genetic background' is not clear. It can be write as 'The different genetic background'.

Line 127: Please write the full scientific names with their common names, if possible.

Line 141: Did the author used also other algorithms than ClustalW? Because it is so obsolete now.

Figure 4: The works cannot be read properly. Please increase the size.

Figure 5 and 6: Can be put in the supplementary file.

The article by Wang et al. “Genome-wide characterization of NLRs in Saccharum spontaneum L. and Their Responses to Leaf Blight in Saccharum" describes the use of genome-wide analysis of nucleotide-binding site-leucine-rich repeat (NLR) receptor to disease-resistant breeding in sugarcane. This paper is written very well with a large quantity of data and elaborations. The work is interesting and appropriate to be published in the special issue of Agronomy “Genetic diversity of disease resistance in crops”.

 

I suggest some additions and minor corrections to improve the manuscript:

Line 74: Please, put “S. officinarum” in italics

Line 183: it is not clear if S. tainanensis infections are natural or artificial, please, explain

Line 201: There is no information about RNA extraction method. Please add a description how samples were prepared.

Line 346: Please, improve the quality and size of Figure 4 to be readable.
