Identification and Characterization of the Tomato UGT Gene Family and Effects of GAME 17 Overexpression on Plants and Growth and Development under High-CO2 Conditions
Round 1
Reviewer 1 Report
In this manuscript, the authors identify UDP-glycosyltrasferase (UGT) in tomato through sequence analysis, and find that constitutive expression of GAME17 results in significant differences in parameters such as plant height, leaf length, etc) under high CO2 conditions. In particular, they find overexpression to results in significantly higher contents of glucose, fructose, and soluble sugar.
The bioinformatics and experiments seem to be performed adequately, and the results presented support the claims made. However, the English language of the manuscript is very poor and distracting. The text would require extensive editing before it could be seriously considered.

- The manuscript deals with the identification and characterization of tomato UGTS gene family and the effects of GAME17 overexpression on plants. In particular, the effects of the induced overexpression of GAME17 and its effects on the soluble sugars' content in vegetative and generative parts of tomato plants are studied. As such it is of significant interest to both tomato breeders and producers.
-- We thank the reviewer for this observation and suggestion.
- While comprehensively presenting the methodology and results from the research undertaken authors somewhat underdelivered on the important part of the Discussion. Their results indicate that further implications of the diversion of metabolites by the overexpression of GAME17 gene could be inferred. Therefore, a more extensive discussion in that respect would greatly improve the value of the manuscript.
-- We thank the reviewer for this observation and suggestion.
-- According to the reviewer’s suggestion, we have extensive discussion in that respect.
- I also have some technical remarks, as described below, that need to be corrected before the manuscript could be considered ready for publication:
The last sentence of 2.1. (on Page 2) states inaccurately that "... the identified candidate protein sequences containing the UDPGT domain were considered as the SlUGT genes ...". However protein sequences are NOT genes, but rather products thereof. Therefore, the sentence should be re-formulated to correctly reflect the actual situation. The name of bacterium used for genetic transformation is misspelled in 2.5. as Agrobacterium tumefactions, which should be replaced by the correct one.The 3-rd sentence in 2.7. ("For real-timequantitative PCR (RT-PCR) of , SYBR Premix Ex Tag was used ...") is incomplete and should be edited for clarity.References to Figure 4c and 4d in section 3.5 need to be replaced to references to Figure 5c and 5d.
-- We thank the reviewer for this observation and suggestion.
-- According to the reviewer’s suggestion, We have proofread in this manuscript .
