Establishment of DNA Molecular Fingerprint of Caladium Core Collections
Round 1
Reviewer 1 Report
The study aimed at molecular fingerprinting for the establishment of core collection in Caladium, is well perceived and conducted resulting in identification of core collection retaining most of the diversity of the collection. The set of 16 EST-SSR markers selected were highly polymorphic with large number of alleles that allowed identification of each of the members of core collection.
However, the collection was probably limited to commercially grown genotypes/varieties limiting the scope of core collection derived from it. Moreover the discussion is centered on molecular markers where as it would be desirable if Caladium is given the importance. The results might be discussed towards distribution, origin, migration and parentage and breeding/propagation method of the genotypes/species.

I have gone through the manuscript entitled "Establishment of DNA molecular fingerprint of Caladium core collections". In this article the authors have performed diversity analysis by 16 EST-SSR markers to screen 144 Caladium accessions. Cluster analysis divided the core collections into four populations. Finally they have done DNA molecular fingerprint of 44 core accessions .These genomic resource will be useful for dentifying, preserving, and utilizing Caladium germplasm resources. I advised the authors to improve the introduction section and conclusion section for more clarity.

The manuscript ”Establishment of DNA molecular fingerprint of Caladium core collections” is dealing with analysis of genetic variability of Caladium accessions. Authors used 16 EST-SSR markers to screen 144 Caladium accessions. They indentified
final core of 44 accessions and established DNA molecular fingerprint which include QR code-type DNA molecular IDs. This DNA based IDs established for the Caladium core collections in this study can play a significant role in identifying and protecting germplasms.
The manuscript has some merit and falls in a scope of the journal. It can be accepted for publication with minor, mostly grammar and spelling errors. All errors are highlighted in text.
