Round 1

Reviewer 1 Report

Ιt is a very well organized and written experimental work. It concludes both fermentation, fractionation and separation issues, using very modern instruments, with very attractive to scientists cosmetic issue, such us a powerful natural whitening agent.

Some minor remarks are the following:

Table 2. Clarify which exactly main component or total substances are determined in column Extract yield %

Line 302: It should be discussed a little why only ethyl acetate and butanol fractions had the best effect.

Line 416: I can not find the relation of absorbance enhancement in cosmetics in 13th reference. Please, clarify properly.

Reviewer 2 Report

Stachys sieboldii is a herbal medicine with astringent properties. The authors aimed to screen for the astringent effect of Stachys sieboldii during the process and found that the aqueous extract of Stachys sieboldii from the ferment of Hericium erinaceus mycelia showed the strongest tyrosinase inhibitory effect and the lowest melanin content. The single compound isolate, identified as acteoside, showed potential whitening activity and dose-dependent inhibition of melanin synthesis. In-depth pathway validation verified the molecular effects of acteoside's whitening activity at the molecular level. In conclusion, the study is a strong work of applied value. The study is well thought out and the WB experimental results are reliable and well written. Overall, it is a nice manuscript.

The following detail errors need careful revision.

On line 343, "[M-H]-" should read "[M-H]- (superscript)".

In line 489, "in vitro" and "in vivo" should be in italics.

All journal names in the references should be the prescribed abbreviations, please standardise the form.

Figure 5A, differences in statistical results should be labelled.

It is OK.
