The article "Antitumor Strategies Targeting Peptidergic Systems" provides a concise synopsis of the ongoing debate regarding peptides and their various medical applications.

The published works provide strong support for this manuscript. All things considered in this review/point of view convey a coherent worldwide message. The topic covered in Rodriguez and Coveñas' article is relevant to the journal audience, much like Cancer is. The manuscript is well-written, and the author's interpretations appear well-founded.

This review addresses an interesting aspect of the therapeutic potential of peptide receptor antagonists as broad antitumor agents. It summarizes and discusses the amazing finding that there are enough experimental in vitro and in vivo data to carry out a clinical trial and clear up doubts about the broad antitumor effect exerted by peptide receptor antagonists in preclinical studies. While the reported findings are recurrent and complete, the text lacks focus and clarity. In the introduction section discuss more about cell penetrating peptide types and antitumor action mechanisms. Besides schematic illustrations, more figures, tables, and data should be added to the manuscript. I think the manuscript should pay more attention to focus on the in vivo studies and applications. In addition, the titles of Sections 2 and 3 should be edited and show the main aspects of their sections. Some of the references are too old and should be updated. The caption of Fig 1 should be improved.

English is modest. Therefore, the authors need to improve their writing style. In addition, the whole manuscript needs to be checked by native English speakers.

The English text has undergone a complete revision to enhance clarity. Some sentences have been rephrased to improve readability. A new bibliographic reference (number 8) related to cell-penetrating peptides has been included. Also, the figure legend for figure number 1 has been modified. The text has been adjusted to fit the format of an entry for the Encyclopedia.
