Background: Nephrolepis exaltata (sword fern) possesses a considerable amount of phytochemicals and different biological activities. The current study investigates the anti-biofilm potential of greenly synthesized bimetallic nanoparticles of Nephrolepis exaltata leaf methanol extract (NEME-MnO2-MgO BNPs). Methods: The NEME was subjected to UPLC/MS analysis, followed by characterization of its NPs by size, zeta potential, FTIR, entrapment efficiency, and release. Then, antioxidant, antimicrobial and antibiofilm assays were employed, followed by in silico studies. Results: The UPLC/MS analysis of NEME led to the tentative identification of 27 metabolites, mostly phenolics. The MnO2-MgO BNPs presented a uniform size and distribution and exhibited IC50 values of 350 and 215.6 μg/mL, in the DPPH and ABTS assays, respectively. Moreover, the NPs exhibited antimicrobial and anti-biofilm efficacies against Pseudomonas aeruginosa, Klebsiella pneumonia (ATCC-9633), Staphylococcus aureus (ATCC-6538), Escherichia coli, Bacillus cereus, and C. albicans, with MIC values of 250–500 μg/mL. The MnO2-MgO BNPs inhibited Candida albicans biofilms with a % inhibition of 66.83 ± 2.45% at 1/2 MIC. The network pharmacology highlighted epigallocatechin and hyperoside to be the major compounds responsible for the anti-biofilm potential. The ASKCOS facilitated the prediction of the redox transformations that occurred in the green synthesis, while the docking analysis revealed enhanced binding affinities of the oxidized forms of both compounds towards the outer membrane porin OprD of P. aeruginosa, with binding scores of −4.6547 and −5.7701 kcal/mol., respectively. Conclusions: The greenly synthesized Nephrolepis exaltata bimetallic nanoparticles may provide a promising, eco-friendly, and sustainable source for antimicrobial agents of natural origin with potential biofilm inhibition. Illyrian iris (Iris pallida subsp. illyrica (Tomm. ex Vis.) K.Richt.) is a rhizomatous geophyte, an endemic species (subspecies), occurring within a limited range along the eastern coast of the Adriatic Sea. The study presents the first in-depth chemical and functional investigation of its rhizome extracts using both conventional and greener solvents, as well as essential oil (EO) via hydrodistillation, employing gas chromatography-mass spectrometry (GC/MS) and ultra-high-performance liquid chromatography-diode array detector-tandem mass spectrometry (UHPLC-DAD-MS/MS) for metabolic fingerprinting, which was further interpreted through a chemosensory lens. High-performance thin-layer chromatography (HPTLC) bioautography (HPTLC-DPPH/ HPTLC-Tyrosinase) was applied for the first time to this species, revealing zones of bioactivity. HaCaT cell viability and spectrophotometric assays were employed to further evaluate the cosmetic potential. Results showed a distinctive volatile profile of EO, including, to the best of our knowledge, the first identification of a silphiperfol-type sesquiterpenoid in the Illyrian iris rhizome. UHPLC-DAD-MS/MS and HPTLC fingerprinting further supported solvent-dependent differences in metabolite composition. Notably, acetone, ethyl acetate, and ethanol extracts exhibited similar chemical profiles, while greener extracts showed more divergent patterns. The results provide a foundation for the future exploration of Illyrian iris in sustainable cosmetic applications, emphasizing the need for further in vitro and in vivo validation. Full article

Following the trend of food waste valorization to produce innovative bio-based materials, this study proposes the conversion of brewer’s spent grain (BSG) into added value edible, high oxygen barrier, flexible, active packaging films via an extrusion molding compression method. Gelatin (Gel) was used as both a reinforcement and barrier agent and glycerol (Gl) as a plasticizer. Eugenol was nanoencapsulated on natural zeolite (EG@NZ), and pure clove powder (ClP) was used as an active agent to obtain BSG/Gel/Gl/xEG@NZ and BSG/Gel/Gl/xClP (x = 5, 10, and 15 %wt.) active films. Both BSG/Gel/Gl/xEG@NZ and BSG/Gel/Gl/xClP films show enhanced tensile, oxygen barrier, antioxidant, and antibacterial properties, and low toxicity and genotoxicity values. All BSG/Gel/Gl/xEG@NZ films presented a higher oxygen barrier, higher total phenolic content (TPC) values, higher antioxidant activity according to a 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, higher inhibition zones against Staphylococcus aureus and Escherichia coli, and lower toxicity and genotoxicity than all BSG/Gel/Gl/xClP films. Thus, the superiority of the nanoencapsulated EG in NZ as compared to the physical encapsulated EG in ClP is proved. Briefly, BSG/Gel/Gl/15EG@NZ active film exhibited ~218% higher tensile strength, ~93% higher TPC value, and ~90% lower effective concentration for a 60% antioxidant activity value (EC60) as compared to the pure BSG/Gel/Gl film. The zones against S. aureus and E. coli were 45 and 30 mm, respectively, and the oxygen barrier was zero. The use of this film extended the shelf life of fresh minced meat by two days and exhibited the high potential to be used as active packaging material. Full article

Erythrina caffra is a traditional plant used to treat cancer and inflammation. The study aimed to assess and isolate anticancer compounds from E. caffra bark. The plant material was extracted sequentially in n-hexane, dichloromethane, ethyl acetate and methanol. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and 3-(4,5-di methyl thiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays were used to evaluate the crude extracts’ antioxidant and anticancer activities, respectively. Column chromatography was used to purify the potent extracts of the stem bark in order to isolate the bioactive compounds. The crude extracts of the E. caffra bark demonstrated antioxidant and anticancer activity, with the dichloromethane (DCM) extract producing the most favorable activity. Three compounds, namely Hexacosanyl isoferulate, Tetradecyl isoferulate, and 1-Heneicosanol, were detected in fractions from the DCM extract. All the isolated compounds showed significant anticancer potential, with the hydroxycinnamic acid compounds showing better anticancer effects in the cervical (HeLa) and breast cancer (MCF-7) cells. The compounds showed greater activity than even the standard drug, 5-fluorouracil, in the MCF-7 cells, with the tetradecyl isoferulate and hexacosanyl isoferulate fractions having IC50 values of 123.62 and 58.84 µg/mL, respectively. The compounds were observed to be capable of triggering caspase cascade events, leading to apoptotic cell death. Overall, E. caffra extracts contained important bioactive compounds that induced apoptotic cell death in HeLa and MCF-7 tumor cells, warranting further investigations in vitro and in vivo. Apigenin (AP) is a natural flavonoid with senomorphic potential and neuroprotective action; however, poor aqueous solubility (<1 μg/mL) limits its bioavailability and therapeutic use. Therefore, the aim of this study was to obtain an amorphous dispersion of AP and evaluate its biological properties. Screening of AP solubilization capabilities under supercritical carbon dioxide processing conditions showed that the system with Soluplus (SOL) achieved the greatest improvement in AP dissolution (6455.4 ± 27.2 μg/mL). Using optimized process parameters (50 °C, 6500 PSI), the AP solubility increased to 8050.2 ± 35.1 μg/mL. X-ray powder diffraction (XRPD) confirmed amorphization, aligning with improved dissolution of AP in both acidic and neutral pH media. As a result, using the PAMPA model, an improvement in AP penetration through membranes simulating gastrointestinal and blood–brain barriers was demonstrated. The significant stability of the obtained amorphous AP dispersion (12 months at room conditions) was associated with stabilizing AP–solubilizer intermolecular interactions, mainly expressed as the shifts in the bands of AP in the range of 1018–1269 cm−1 observed in ATR-FT-IR spectra. Chromatographic analysis confirmed the lack of AP decomposition immediately after the preparation of the amorphous dispersion, as well as after 12 months. As expected, the improvement of AP solubility is correlated with better biological activity assessed in selected in vitro tests such as antioxidant properties (2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and cupric ion reducing antioxidant capacity (CUPRAC) assays) and anticholinesterase inhibition capabilities (AChE and BChE assays). The effect of the studies on improving AP solubility under supercritical carbon dioxide processing conditions is obtaining a stable amorphous AP dispersion (up to 12 months). Regardless of the pH of the media, an improvement in AP dissolution and penetration, conditioned by the passive diffusion process, through biological membranes was noted. Moreover, a more efficient antioxidant and neuroprotective effect of AP in the developed amorphous dispersion can also be suggested. Full article

Honeybee brood biopeptides (HBb-Bps) are a novel source of bioactive compounds with potential health benefits. In this study, HBb-Bps were conjugated with honey via a Maillard reaction and their physicochemical properties, digestive stability, antioxidant capacity, and anti-obesogenic effects were evaluated. Simulated gastrointestinal digestion revealed significantly enhanced resistance after conjugation, with the residual content increasing from 46.99% for native HBb-Bps to 86.12% for the honey-conjugated forms; furthermore, antioxidant activity was largely preserved according to the DPPH and ABTS assays. In the in vivo experiments, 30 male BrlHan: WIST@Jcl (GALAS) (Wistar) rats were fed a high-fat diet (HFD) to induce obesity and orally administered honey-conjugated HBb-Bps at doses of 200, 500, or 1000 mg/kg body weight for 16 weeks. The highest dose led to significant reductions in body weight gain, the Lee index, and body mass index. The serum lipid profiles markedly improved, with decreases in the total cholesterol, triglyceride, and LDL levels, as well as cardiovascular risk indices. Furthermore, fecal analysis showed increased levels of short-chain fatty acids, particularly butyrate. These changes suggest enhanced gut microbial activity; however, the prebiotic effects were inferred from the SCFA profiles, as the gut microbiota composition was not directly analyzed. In conclusion, honey-conjugated HBb-Bps improve gastrointestinal stability and exhibit antioxidant, hypolipidemic, and gut-modulating effects, supporting their potential use as functional ingredients for managing diet-induced metabolic disorders. The Australian native foods, despite high phytochemical composition, are severely underutilized in research and on the commercial market. One of these plants is the Davidson plum (Davidsonia jerseyana), a nutrient-dense and sustainable food ingredient. The study aimed to develop functional fruit sorbets incorporating freeze-dried Davidson plum powder (0–20% w/w) and evaluate their physicochemical, antioxidant, and sensory properties. Sorbets were created using strawberry, raspberry, pomegranate, and Davidson plum bases and analyzed for nutritional content, color, melting rate, texture, and antioxidant capacity (Total Phenolic Content (TPC), Total Flavonoid Content (TFC), Ferric Reducing Antioxidant Power (FRAP), Cupric Reducing Antioxidant Capacity (CUPRAC), 2,2-Diphenyl-1-picrylhydrazyl (Radical Scavenging Assay (DPPH)), total proanthocyanin and anthocyanin content. Sensory evaluation was also conducted using a semi-trained panel. The results showed that increasing Davidson plum concentration led to higher antioxidant activity and slower melting rates. Sorbets containing 10% and 15% Davidson plum demonstrated the highest levels of phenolic and flavonoid compounds. However, sensory analysis indicated that sorbets with 5% and 10% Davidson plum, particularly those made with a strawberry base were the most acceptable in terms of flavour, texture, and overall appeal. These findings suggest that incorporating Davidson plum into frozen desserts, especially at lower concentrations, can enhance both the functional and sensory qualities of sorbets while offering potential health benefits. This study investigates the phenolic composition and antioxidant potential of root extracts from three Opuntia ficus-indica varieties (green, red, and orange) using ultra-high-performance liquid chromatography coupled with diode array detection and electrospray ionization–tandem mass spectrometry (UHPLC-DAD-ESI-MS/MS). Phenolic compounds were extracted with a hydromethanolic solvent and quantified by spectrophotometric assays, while antioxidant activity was assessed through DPPH, ABTS, iron III reduction, hydroxyl radical, and nitric oxide scavenging methods. A total of 26 compounds were identified, including piscidic acid, epicatechin-3-O-gallate, and isovitexin, with several phenolics newly reported for O. ficus-indica roots. The green and red varieties showed the highest phenolic contents (up to 147.82 mg/g extract) and strong antioxidant capacity, particularly in ABTS (IC50 = 29.38 μg/mL) and hydroxyl radical inhibition (>90%). Relative Antioxidant Capacity Index (RACI) analysis confirmed a consistent correlation between phenolic/flavonoid content and antioxidant efficacy. These findings highlight the analytical relevance of UHPLC-DAD-ESI-MS/MS for profiling underutilized plant matrices and support the potential use of O. ficus-indica root extracts as natural sources of bioactive compounds for pharmaceutical and biomedical applications. The native fruits of the Cerrado have an interesting composition of bioactive compounds responsible for antioxidant, anti-inflammatory, and antimicrobial activities, with technological potential for functional industries. This study investigated the extraction of bioactive compounds in cagaita and mamacadela fruits, under different conditions, using eutectic solvents based on choline chloride (CC) with citric acid (CA) or tartaric acid (TA), plus ethanol as reference. For a better understanding of the extracts, their antioxidant capacity was assessed by the DPPH, FRAP, ABTS•+, and total phenolic compounds and flavonoids assays, as well as for color, water activity, and identification of bioactive compounds by mass spectrometry. Additionally, the carotenoid contents were evaluated in the ethanolic extracts. The results showed that ethanol was efficient for the extraction of flavonoids and presented advantages demonstrated in the antioxidant analyses of ABTS•+ and FRAP. However, eutectic solvents stood out in the extraction of phenolic compounds, with yields 14.0 and 4.5 times higher than ethanol for mamacadela and cagaita, respectively. In addition, when compared to cagaita, mamacadela had twice the carotenoid content. Furthermore, the CC:TA solvent was the most efficient, demonstrating, by DI-ESI-MS, 29 phenolic compounds in mamacadela and 27 in cagaita. Therefore, the extracts obtained present potential for use as natural pigments, adding value to the fruits and encouraging their exploration by industries. Full article

This study explores the antioxidant potential and delivery performance of five structurally distinct cannabinoids, with a particular focus on cannabinol (CBN). Comprehensive structural characterization using mass spectrometry (MS) and nuclear magnetic resonance (NMR) revealed key molecular features relevant to antioxidant function. Among the tested compounds, CBN exhibited the most potent and balanced radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl, and superoxide radicals. Based on these findings, CBN was selected for formulation into soy lecithin liposomes. The resulting CBN-loaded liposomes displayed favorable colloidal properties, with an average size of approximately 122.9 ± 0.4 nm. Results indicating increased membrane order upon CBN incorporation suggest enhanced stability of the liposomal bilayer. Antioxidant activity assays showed that CBN-loaded liposomes retain significant radical scavenging capacity, though with a moderate reduction compared to free CBN. EPR imaging further demonstrated superior diffusion of liposomal CBN through a gelatin-based semi-solid model compared to the control solution. While the current model does not replicate skin architecture, it provides a cost-effective and reproducible platform for early-stage screening of formulation mobility. These results position CBN-loaded liposomes as a promising candidate for dermal antioxidant applications, combining favorable physicochemical properties with enhanced diffusion behavior. Full article

Functional beverages are increasingly sought as components of a healthy diet, and goat milk offers a nutritious base with unique sensory attributes. This study aimed to develop a novel fermented goat milk beverage enriched with spent coffee grounds (SCG) extract, utilizing SCG’s high natural antioxidant content to improve nutritional and functional properties. SCG was extracted via aqueous solid–liquid extraction and lyophilized; its extract was incorporated into goat milk–fructose blends at 0%, 1%, and 2% (w/v). Analyses included physicochemical characterization (pH, acidity, fat, and protein), total phenolic content, and antioxidant capacity via DPPH assay, alongside consumer sensory evaluation for acceptance and purchase intent. Results demonstrated that higher SCG extract levels significantly increased pH, phenolic concentrations, and radical scavenging activity while reducing titratable acidity. The 2% SCG formulation achieved the highest overall, taste, and aftertaste acceptance and purchase intention. These findings suggest that SCG-enriched goat dairy beverages are feasible functional foods with enhanced antioxidant properties and consumer appeal, promoting valorization of coffee by-products. Background and Objectives: The cultivation of Humulus lupulus has been encouraged in Brazil, despite the country’s climate generally being unfavorable for its development. This study aimed to evaluate the chemical composition, antioxidant activity, and photoprotective potential of four H. lupulus varieties, Cascade, Columbus, Comet, and Nugget, cultivated in the northwestern region of Paraná State, Brazil. Methods: The varieties were grown in experimental plots. Crude extracts (CEs) of cones and leaves were obtained through dynamic maceration with solvent renewal (96% ethanol), followed by concentration in a rotary evaporator. Assays for sun protection factor (SPF), total phenolic content (TPC), total flavonoid content (TFC), ferric reducing antioxidant power (FRAP), 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and 2.2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS⁺) were performed to assess antioxidant activity. Chemical profiling was performed using UHPLC-MS/MS. Results: UHPLC-MS/MS analysis revealed the presence of phenolic and organic acids, flavonoids, phenolic aldehydes, alkaloids, and α-benzopyrone-type lactones, with high concentrations of rutin (>500 µg/g) in both cones and leaves. Total phenolic content ranged from 69.70 to 95.95 µg gallic acid equivalent/mg CE; flavonoids from 170.53 to 696.67 µg quercetin equivalent/mg CE; DPPH EC50 values ranged from 1.34 to 3.13 mg/mL; FRAP from 1.19 to 2.52 µM ferrous sulfate/mg; ABTS⁺ from 5.11 to 22.60 mM Trolox/mg CE; and SPF ranged from 16.02 to 39.48 in the CE of H. lupulus cones and leaves. Conclusions: These findings demonstrate that the cultivated varieties possess antioxidant and photoprotective properties, encouraging further studies to explore their potential applications. This study aimed to optimize the total phenolic content (TPC) and antioxidant activity (AA) of acerola (Malpighia emarginata) by-product extracts obtained by probe-type ultrasound-assisted extraction and assess the composition and antibacterial activity of the extract obtained under optimized conditions. A Box–Behnken experimental design was applied to evaluate the effects of ultrasonic power (350 to 650 W), ethanol concentration (20% to 80% v v−1), and extraction time (20 to 60 min) on TPC and AA. The optimal extraction conditions were 650 W, 50% (v v−1) ethanol, and 20 min, which yielded the highest values of TPC (3.36 g gallic acid equivalent 100 g−1) and AA through the DPPH radical scavenging method (4.97 mM Trolox equivalents 100 g−1) and a ferric reducing antioxidant power assay (11.35 mM Trolox equivalents 100 g−1). Organic acids, phenolic acids, flavonoids, and alkaloids were identified in the optimized extract, including malic acid, protocatechuic acid, resorcylic acid, and rutin. The optimized extract (2.89–11.32 mg mL−1) inhibited the growth of Listeria monocytogenes, Shigella sonnei, Salmonella enterica subsp. enterica Typhi, Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. Acerola by-products represent a promising source of extracts with the potential to replace synthetic additives, contributing to the circular economy of agroindustries. Full article

Equisetum telmateia Ehrh. (great horsetail) belongs to the Equisetaceae family and its aerial parts have been traditionally used for skin conditions and to achieve healthy and resilient skin, nails, and hair. This study aimed to evaluate the inhibition of skin-related enzymes by, the antioxidant capacity of, and the phytochemical composition of E. telmateia. Additionally, a novel emulgel was formulated from the main methanolic extract and characterized in terms of pH, viscosity, determination of content quantification, textural profile analysis, and spreadability. After the characterization studies, in vitro release and ex vivo permeation and penetration studies were performed. Firstly, the dried aerial parts of E. telmateia were macerated in methanol, followed by partitioning with solvents of increasing polarity: n-hexane, chloroform, ethyl acetate, and n-butanol. Antioxidant activity was assessed using DPPH, FRAP, CUPRAC, and TOAC assays, while enzyme inhibition was analyzed for collagenase, elastase, hyaluronidase, and tyrosinase. LC-MS/MS analysis identified 53 phytochemical compounds. Protocatechuic acid, the main phenolic compound, was quantitatively analyzed in each subfraction by HPTLC. The in vitro release studies showed sustained release of the reference substance (protocatechuic acid) and the kinetic modeling of the release was fitted to the Higuchi model. The ex vivo permeation and penetration studies showed that the formulation exhibited a retention of 3.06 ± 0.21 µg.cm−2 after 24 h, whereas the suspended extract demonstrated a skin retention of 1.28 ± 0.47 µg.cm−2. Both the extracts and the formulated emulgel exhibited inhibitory effects on skin-related enzymes. Our finding suggested that E. telmateia might be a valuable ingredient for wrinkle care and skin-regenerating cosmetics. Full article

Using ultrasound-assisted extraction, we obtained a chlorophyll-rich extract from Opuntia ficus-indica cladodes (OFI) characterized through thin-layer chromatography (TLC), Fourier-transform infrared spectroscopy (FTIR), and spectrophotometric absorption analysis. The dye exhibited a strong fluorescence response in the visible range (400–800 nm) with a pronounced red emission when excited with a UV source. Antioxidant ability was evaluated via DPPH assay, showing an IC50 of 185 µg/mL, highlighting its potential for reactive oxygen species scavenging. The extract was incorporated into polymethyl methacrylate (PMMA), polyvinylpyrrolidone (PVP), and polyvinyl alcohol (PVA), leading to fluorescence intensity enhancements of up to 40 times compared to the dye alone depending on matrix polarity, consistent with aggregation and polarity effects. Stability tests confirmed the dye’s resistance to CO2 exposure, pH variations, and prolonged storage, positioning it as a viable alternative to synthetic fluorophores. These findings suggest that the OFI extract provides a functionally relevant, bio-derived dye platform promoting the valorization of agricultural by-products in high-value technological applications, highlighting a circular and scalable approach to developing ecofriendly fluorescent materials, aligning with sustainability and green technology goals.
