Phytophthora Species and Their Associations with Chaparral and Oak Woodland Vegetation in Southern California

Evidence of unintended introductions of Phytophthora species into native habitats has become increasingly prevalent in California. If not managed adequately, Phytophthora species can become devastating agricultural and forest plant pathogens. Additionally, California’s natural areas, characterized by a Mediterranean climate and dominated by chaparral (evergreen, drought-tolerant shrubs) and oak woodlands, lack sufficient baseline knowledge on Phytophthora biology and ecology, hindering effective management efforts. From 2018 to 2021, soil samples were collected from Angeles National Forest lands (Los Angeles County) with the objective of better understanding the diversity and distribution of Phytophthora species in Southern California. Forty sites were surveyed, and soil samples were taken from plant rhizospheres, riverbeds, and off-road vehicle tracks in chaparral and oak woodland areas. From these surveys, fourteen species of Phytophthora were detected, including P. cactorum (subclade 1a), P. multivora (subclade 2c), P. sp. cadmea (subclade 7a), P. taxon ‘oakpath’ (subclade 8e, first reported in this study), and several clade-6 species, including P. crassamura. Phytophthora species detected in rhizosphere soil were found underneath both symptomatic and asymptomatic plants and were most frequently associated with Salvia mellifera, Quercus agrifolia, and Salix sp. Phytophthora species were present in both chaparral and oak woodland areas and primarily in riparian areas, including detections in off-road tracks, trails, and riverbeds. Although these Mediterranean ecosystems are among the driest and most fire-prone areas in the United States, they harbor a large diversity of Phytophthora species, indicating a potential risk for disease for native Californian vegetation. The most significant ectoparasitic mite of honeybees, Varroa destructor, has a detrimental effect on bee health and honey output. The principal strategy used by the control programs is the application of synthetic acaricides. All of this has resulted in drug resistance, which is now a major worry for beekeeping. As a result, research on alternate products and techniques for mite management is now required. The aim of this study was to determine whether essential oils (EOs) extracted from botanical species of Lamiacae, typical of the Calabria region of Southern Italy, could reduce the population of the mite V. destructor. Among the best-known genera of the Lamiaceae family are oregano, rosemary and thyme, whose EOs were employed in this study. By steam distillation, the EOs were extracted from Origanum vulgare subsp. viridulum (Martrin-Donos) Nyman, Thymus capitatus Hoffmanns. and Link, Thymus longicaulis C.Presl and Salvia rosmarinus Schleid. plant species harvested directly on the Calabrian territory in their balsamic time. Each EO went to the test in vitro (contact toxicity) against V. destructor. Fifty adult female mites, five for each EO and the positive and negative control, were used in each experimental replicate. The positive controls comprised five individuals treated to Amitraz dilute in acetone, and the negative controls included five individuals exposed to acetone alone. To create the working solution to be tested (50 μL/tube), the EOs were diluted (0.5 mg/mL, 1 mg/mL, 2 mg/mL and 4 mg/mL) in HPLC-grade acetone. After 1 h of exposure, mite mortality was manually assessed. Origanum vulgare subsp. viridulum, Thymus capitatus and Thymus longicaulis were the EOs with the highest levels of efficiency at 2 mg/mL, neutralizing (dead + inactivated), 94%, 92% and 94% of parasites, respectively. Salvia rosmarinus EO gave a lower efficacy, resulting in a percentage of 38%. Interestingly, no adverse effects were highlighted in toxicity tests on honeybees. These results show that these OEs of the Lamiaceae family have antiparasitic action on V. destructor. Therefore, they could be used, individually or combined, to exploit the synergistic effect for a more sustainable control of this parasite mite in honeybee farms. Full article

Members of the genus Salvia are used as culinary herbs and are prized for their purported medicinal attributes. Since physiological effects can vary widely between species of Salvia, it is of great importance to accurately identify botanical material to ensure safety for consumers. In the present study, an in-depth chemical investigation is performed utilizing GC/Q-ToF combined with chemometrics. Twenty-four authentic plant samples representing five commonly used Salvia species, viz. S. apiana, S. divinorum, S. mellifera, S. miltiorrhiza, and S. officinalis, are analyzed using a GC/Q-ToF technique. High-resolution spectral data are employed to construct a sample class prediction (SCP) model followed by principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA). This model demonstrates 100% accuracy for both prediction and recognition abilities. Additionally, the marker compounds present in each species are identified. Furthermore, to reduce the time required and increase the confidence level for compound identification and the classification of different Salvia species, a personal compound database and library (PCDL) containing marker and characteristic compounds is constructed. By combining GC/Q-ToF, chemometrics, and PCDL, the unambiguous identification of Salvia botanicals is achieved. This high-throughput method can be utilized for species specificity and to probe the overall quality of various Salvia-based products. Full article

Sage species belong to the family of Labiatae/Lamiaceae and are diffused worldwide. More than 900 species of sage have been identified, and many of them are used for different purposes, i.e., culinary uses, traditional medicines and natural remedies and cosmetic applications. Another use of sage is the application of non-distilled sage extracts and essential oils to control phytopathogenic bacteria and fungi, for a sustainable, environmentally friendly agriculture. Biocidal propriety of non-distilled extracts and essential oils of sage are w documented. Antimicrobial effects of these sage extracts/essential oils depend on both sage species and bacteria and fungi species to control. In general, it is possible to choose some specific extracts/essential oils to control specific phytopathogenic bacteria or fungi. In this context, the use of nanotechnology techniques applied to essential oil from salvia could represent a future direction for improving the performance of eco-compatible and sustainable plant defence and represents a great challenge for the future. The monoterpenoids and diterpenoids in the preparation penetrate the skin of the feet and stop the pain chemokine cycle, which may be the basis of chronic pain. The present work is conducted as part of the development and the valorization of bioactive natural substances from Algerian medicinal and aromatic spontaneous plants, a clean alternative method in biological control. For this purpose, the bio-acaricidal activity of Salvia officinalis (sage)essential oil (EO)was evaluated against the Varroa destructor, a major threat to the honey bee Apis mellifera ssp. intermissa. The aerial parts of S. officinalis L., 1753 were collected from the Chrea mountainous area in Northern Algeria. They were subjected to hydro distillation by a Clevenger apparatus type to obtain the EO, and screened for bio-acaricidal activity against Varroa destructor by the method of strips impregnated with the mixture EO and twin according to three doses. Pre-treatment results revealed infestation rates in the experimental site ranging from 3.76% to 21.22%. This showed the heterogeneity of infestations in hives according to the density of bees. This constituted a difficulty in monitoring the population dynamics of this parasite. After treatment, a difference in the acaricidal effect of Sage essential oil is noticed. It gives a mortality rate of 6.09% by the dose D1: 5%, 2.32% by the dose D2: 15%, and a low mortality rate of 0.9% by the dose D3: 20%. The chemical treatment carried out by Bayvarol gives a result close to that of the essential oil of Sage (9.97%).These results point to the fact that Sage essential oil treatments have a significant effect and good biological activity with regard to harmful species. The chemical composition of the essential oils of Salvia africana L., Salvia elegans Vahl, Salvia greggii A. Gray, Salvia mellifera Green and Salvia munzii Epling, cultivated in Eboli (Salerno, Southern Italy), was studied by means of GC and GC-MS analyses. In all, 88 compounds were identified, 54 for S. africana, accounting for 95.4% of the total oil, 55 for S. elegans (92.9%), 50 for S. greggii (96.9%), 54 for S. mellifera (90.4%) and 47 for S. munzii (97.5%), respectively. In S. africana,the amount of monoterpenoids and sesquiterpenoids is very similar. For other species, the monoterpenoid percentage is greater than the amount of sesquiterpenoids. The oils of S. elegans, S. greggii and S. munzii were active inhibitors of germination and radical elongation of Raphanus sativus L. and Lepidium sativum L. Full article
