Highly Efficient Nitrogen Removal by Stutzerimonas stutzeri Strain MJ20: Metabolic Pathways and Potential for Biofloc Systems and Low C/N Ratio Aquaculture Wastewater

Although numerous studies have focused on the potential application of heterotrophic nitrification–aerobic denitrification (HNAD) bacteria in wastewater treatment, research exploring their potential in aquaculture biofloc systems remains limited. In this study, a promising HNAD strain, identified as Stutzerimonas stutzeri MJ20, was isolated from mature biofloc. This strain efficiently utilized low-cost carbon sources (e.g., glucose) and small-molecule carbon sources (e.g., sodium acetate and sodium succinate). Under conditions with glucose as the carbon source, a carbon-to-nitrogen (C/N) ratio of 15, pH 6–9, temperature 25–35 °C, salinity 0–35‰, and shaker speed of 0–150 rpm, it achieved removal rates of 95–100% for NH4+-N, NO2-N, and NO3-N at initial concentrations of 100 mg/L each. Even at higher concentrations (up to 200 mg/L NH4+-N and 500 mg/L for both NO2-N and NO3-N), removal rates exceeded 99%. Under mixed nitrogen sources, strain MJ20 demonstrated efficient nitrogen removal, preferentially utilizing NH4+-N, with only minimal and transient accumulation of nitrite and nitrate. Genomic analysis revealed that MJ20 carries key denitrification genes, including napA, nirS, norB and nosZ, and possesses complete pathways for nitrate reduction to nitrogen gas and ammonia assimilation, although typical autotrophic nitrification genes were not detected. Combined genomic data and autotrophic culture experiments indicated that, in addition to utilizing various organic carbon sources, the strain also exhibited certain autotrophic growth capabilities. Furthermore, MJ20 showed strong flocculation ability (flocculation rate > 96% within 16 h), sensitivity to multiple common antibiotics, and no toxicity to zebrafish, demonstrating favorable biosafety. In simulated seawater aquaculture wastewater with a C/N ratio of 5, it achieved a total nitrogen removal rate exceeding 94% within 72 h. These results indicate that strain MJ20 possesses comprehensive advantages, including efficient nitrogen removal, broad carbon source adaptability, strong environmental resilience, minimal accumulation of intermediate nitrogen products, excellent flocculation ability, and high biosafety. These traits highlight its potential for application in biofloc systems and in treating aquaculture tail water with a low C/N ratio. This study provides theoretical insights and practical guidance for screening HNAD bacteria suitable for biofloc systems. Full article

This article presents the design and development of an interactive architecture oriented toward the management of traditional vallenato, a musical genre recognized as an Intangible Cultural Heritage of Humanity by UNESCO. Architecture combines the principles of contextual awareness and the use of massive open online courses (MOOCs) to face the current challenges of preservation, dissemination, and teaching of this cultural expression, threatened by commercialization and the loss of its traditional roots. Through a modular structure, adaptive technological tools are integrated to capture, process, and use contextual information, personalizing learning experiences and strengthening the link between communities and their cultural heritage. The proposal consists of several functional layers, including context management, user profiles, educational resources, and a persistence unit, each designed to ensure the interoperability and sustainability of cultural data. In addition, the capacity of architecture to be used in other cultural contexts is highlighted, expanding its impact on different artistic manifestations and heritages worldwide. This article includes a comparative analysis with other existing models, highlighting the advantages of this solution in terms of customization and adaptability. Finally, opportunities for improvement and expansion are explored, as well as the pending challenges in the implementation of this technological tool in educational and cultural environments. The heritagisation of cultural landscapes is often understood as a state-led administrative process. At the same time, the discursive origins and adaptive mechanisms that precede formal designation remain underexplored, especially in relation to cultural sustainability. This study examines the pre-heritagisation of Suzhou’s classical gardens during China’s modern transformation by analysing periodical discourse published between 1870 and 1948. Using a mixed-methods approach that combines quantitative content analysis and Critical Discourse Analysis (CDA), it investigates 699 historical texts from the Index to Chinese Newspapers & Periodicals database. The findings reveal a dual discursive process. On the one hand, reports portrayed the gardens as accessible, multifunctional civic spaces through narratives of public use. On the other hand, literati discourse reinforced their classical value through historical memory and aesthetic preservation. Together, these tendencies show how the gardens were materially refunctioned and symbolically re-anchored under modern conditions. Rather than directly producing later heritage designation, this process helped create the socio-cultural conditions through which the gardens acquired broader public intelligibility, cultural legitimacy, and heritage-like meanings before formal institutional recognition. Ceratitis capitata (Diptera: Tephritidae), or medfly, is an invasive pest widespread in Argentina, where standardized management methods, such as cultural and chemical controls, are commonly implemented. The success in controlling medfly populations depends on implementing preventive, sustainable, long-term, and eco-friendly eradication/control strategies across all invaded environments. One strategy may involve augmentative biological control using parasitoids adapted to local conditions, such as Ganaspis pelleranoi (Brèthes) (Hymenoptera: Figitidae), a Neotropical-native parasitoid that mostly forages on tephritid larvae in a broad range of fallen fruit. Two hypotheses were tested in the current study: (1) G. pelleranoi females are more efficient in controlling medfly larvae infesting different fruits as the density of released females progressively increases, and (2) such parasitoid-induced host mortality capacity remains when host density is increased. Parasitism (reproductive effects) and additional host mortality (non-reproductive effects) were the indicator variables of parasitoid-induced host ability. Trials were performed in field cages (semi-field conditions) using two medfly-multiplier host fruit species, namely sour orange and peach, and with variations in both parasitoid release and host larval densities. Three major findings were highlighted: (1) G. pelleranoi females successfully parasitized host larvae on peach and sour orange, regardless of their strongly differing physical features, although medfly larvae in peaches were significantly more susceptible to the parasitoid; (2) medfly mortality significantly increased in both peach and sour orange relative to the gradual increase in released G. pelleranoi females, regardless of the increase in host density offered to parasitoids; and (3) G. pelleranoi females induced a substantially high host die-off rate when the additional mortality was added to the analysis, which was not revealed when parasitism alone was regarded as a medfly mortality variable. Such outcomes may provide relevant information for implementing an augmentative biological control against medfly using indigenous parasitoid species within an eco-friendly fruit fly pest management approach. Background: Haitian immigrants in the United States face health literacy challenges related to recent migration, language discordance, and unfamiliar healthcare systems, yet no general health literacy instrument has been psychometrically validated in Haitian Creole. This study translated, culturally adapted, and evaluated the Haitian Creole HLS19-Q12 (HLS19-Q12-HC). Methods: Haitian Creole-speaking adults without cancer diagnoses in South Florida (n = 168) completed the HLS19-Q12-HC and the Haitian Creole Brief Health Literacy Screen. Translation included forward–backward procedures, expert review, and cognitive interviews (n = 7). Psychometric evaluation used confirmatory factor analysis, reliability testing, and assessment of convergent and known-groups validity. Results: Cognitive interviews supported clarity and cultural appropriateness with minor refinements. Reliability was excellent (ω = 0.949; α = 0.944; AVE = 0.584). The unidimensional model showed good fit (CFI = 0.951; TLI = 0.944; RMSEA = 0.065; SRMR = 0.048), whereas multi-factor models showed limited discriminant validity. Convergent and known-groups validity were supported. Using provisional European-derived cutpoints, 70.2% of participants were classified as having inadequate or problematic health literacy. Conclusions: The HLS19-Q12-HC showed evidence of reliability and validity as a unidimensional measure of general health literacy and may support research, needs assessment, and culturally responsive interventions for Haitian Creole-speaking populations. Findings should be interpreted in light of the convenience sample from South Florida and the predominantly female composition of the cohort. Full article

Recently, polyhydroxyalkanoates (PHAs) have gained significant attention as a bioactive material for replacing petrochemical plastics. PHAs can be produced by microorganisms growing on sludge substrates. In this study, fish-processing wastewater was investigated as an alternative substrate for PHA production using Bacillus cereus. Wastewater dilution, carbon-to-nitrogen ratio modification, and the addition of fish oil as a lipidic substrate were examined, and bacterial growth and biopolymer production were optimized. First, wastewater was diluted (25–100%) and examined. The 50% dilution treatment was selected, yielding a CDM of 0.426 g/L and a PHA content of 6.69%. In subsequent steps, the effects of wastewater fermentation and bacterial adaptation prior to the main production processes were investigated. According to the results, the 50% and 100% fermented treatments exhibited higher CDM values (0.970–1.022 g/L) compared to the non-fermented treatments. Cultures inoculated with adapted bacteria showed superior performance (CDM: 1.455 g/L, PHA: 0.499 g/L, PHA content: 34.63%) relative to non-adapted treatments. The effect of the carbon-to-nitrogen (C/N) ratio was also optimized by supplementing two carbon sources: glucose and crude fish oil. The optimal treatment T1 (effluent + 0.6 g/L glucose) had a CDM of 1.32 g/L and a PHA content of 0.215 g/L. Treatment 1, which consisted solely of effluent and fish oil, exhibited higher values (CDM: 1.12 g/L, PHA: 0.65 g/L) and was therefore considered the cost-effective treatment. Subsequently, a scale-up process was conducted in a 4 L bioreactor over 300 h under semi-continuous, long-term cultivation. The optimal harvesting time for the biopolymer was achieved during the fourth cycle (180–240 h). The produced biopolymer was characterized using FTIR, NMR, TGA, DSC, SEM, and XRD analyses, confirming the production of a copolymer, specifically poly(3-hydroxybutyrate-co-3-hydroxyvalerate). This study used wastewater from the fish industry for the production of biodegradable polyhydroxyalkanoates. Full article

The digital transformation of healthcare requires stronger digital competencies among pharmacists, yet evidence on the effectiveness of structured training remains scarce. This study examines the impact of a blended digital health training programme delivered to practicing pharmacists in Bihor County, Romania, as part of the Romanian pilot of the EU-funded European Health Professionals’ and the DigitAl team SkillS (H-PASS) project. A single-group pre–post educational design was applied to pharmacists from Bihor County, Romania, participating in a modular digital health training programme delivered between May and July 2025. A total of 84 pharmacists completed both pre-training and post-training self-reported competency assessments comprising 18 items across three modules: digital innovation and change management, communication and collaboration, and data management and digital literacy. Paired samples t-tests, Cohen’s d effect sizes, Cronbach’s alpha, moderator analyses, and ceiling effect analyses were conducted using Python-based statistical workflows. Statistically significant improvements were observed across all three modules (all p < 0.0001), with large effect sizes (d = 1.04–1.30). Post-training internal consistency increased substantially, with overall Cronbach’s alpha reaching 0.74. The greatest item-level gains were recorded in adaptive communication, cultural adaptation of care, and data protection ethics. No significant moderation effects were found for age, gender, or years of experience. Course satisfaction showed a moderate positive correlation with competency gains (r = 0.528), while perceived improvement was not significantly associated with observed score change. A ceiling effect indicated greater gains among participants with lower baseline competencies. The Romanian implementation of the H-PASS training programme was associated with improved self-reported digital health competencies among practicing pharmacists, high-lighting its potential as a scalable model for digital upskilling in healthcare. Proteomic research in the genus Vitis has progressed from early biochemical studies of soluble proteins to high-resolution, quantitative analyses encompassing all major organs and derived products. This review provides a comprehensive synthesis of advances in grapevine and wine proteomics. In leaves, studies have revealed extensive remodeling of photosynthetic, antioxidant, and defense pathways under biotic (e.g., Plasmopara viticola, Erysiphe necator, Xylella fastidiosa, Candidatus Phytoplasma vitis) and abiotic stresses (drought, salinity, heat, light). Bud proteomics elucidated hormonal regulation and mechanisms of dormancy release, while root studies identified nitrate-dependent metabolic shifts and adaptive protein networks. Cell culture models enabled controlled investigation of elicitor responses, stilbene biosynthesis, and temperature-induced proteome changes. In berries, proteomics clarified developmental transitions from fruit set to ripening, emphasizing proteins related to secondary metabolism, vacuolar transport, and stress tolerance. Comparative analyses across cultivars and environments identified biomarkers linked to aroma, color, and texture. The wine proteome revealed selective persistence of grape-derived proteins (e.g., thaumatin-like proteins, chitinases) and yeast peptides influencing stability and sensory properties, while Botrytis cinerea infection significantly alters this balance by degrading PR proteins and introducing fungal enzymes. Altogether, the Vitis proteome emerges as a dynamic, multifunctional system crucial for understanding plant adaptation, enological quality, and biomarker discovery. Full article

Probiotics Exert Colonization Resistance Against F. nucleatum subsp. polymorphum: Disruption by Antibiotics and Underlying Molecular Mechanisms

Fusobacterium nucleatum (F. nucleatum), a key oral pathogen, promotes colorectal cancer (CRC) progression via gut translocation. Although gut probiotics provide colonization resistance against pathogens, antibiotic-induced dysbiosis may facilitate F. nucleatum integration and increase the risk of CRC. The mechanisms underlying probiotic–F. nucleatum antagonism and antibiotic modulation remain unclear. A 33-strain probiotic consortium and F. nucleatum subsp. Polymorphum (F. polymorphum) ATCC 10953 were co-cultured. The inhibitory effects of probiotics on F. nucleatum and the impacts of antibiotics (ABXs) on the microbial community structure in the co-culture system and on the probiotic-mediated inhibition of F. nucleatum were evaluated using spent medium assays, plate confrontation tests, growth curves, qRT-PCR, metagenomic sequencing, and transcriptomics. Hydrogen peroxide/pH/lysine assays and coaggregation models were performed to probe the associated mechanisms. Probiotics strongly inhibited the growth of F. nucleatum in a dose-dependent manner, primarily via organic acids, while F. nucleatum enriched amino acid/vitamin biosynthesis pathways without major growth suppression. Antibiotics weakened probiotic antagonism, shifted species abundance (↓ L. plantarum, ↑ L. paracasei), induced adaptive stress responses in F. nucleatum (↑ nucleotide metabolism, propanediol degradation, pdxS), and reduced lysine biosynthesis. Lysine supplementation restored probiotic abundance and disrupted F. nucleatum coaggregation. Multi-strain probiotics exert potent colonization resistance effects against F. nucleatum, mainly through organic acids and metabolic interference. Antibiotic-induced dysbiosis impairs this protective effect and may promote the persistence of F. nucleatum, which has been implicated in CRC risk. Targeted probiotic strategies may offer novel preventive approaches. Full article

South Africa’s food system reflects a growing imbalance between nutritionally rich indigenous foods and the increasing dominance of commercially processed alternatives. Despite well documented health, cultural, and environmental benefits, indigenous leafy vegetables and edible insects remain marginal within formal markets and everyday diets. This systematic review synthesised 141 empirical and theoretical studies to examine how household attitudes, consumption behaviours, and market structures interact to shape the role of indigenous foods in South Africa. The review identifies a consistent pattern in which positive perceptions of indigenous foods do not translate into regular consumption. Rural households continue to utilise these foods as part of seasonal and livelihood strategies, while uptake in urban areas remains uneven and context specific. Emerging interest among certain consumer segments highlights potential for product diversification and market development, particularly where indigenous foods are adapted to align with modern preferences. However, this potential is constrained by weak value chain integration, limited standardisation, and the absence of reliable consumption data. Global urban development has fully entered the stage of stock renewal, and the synergy between public health and historic heritage conservation has become a core issue of urban sustainable development in the post-pandemic era. As special spatial units carrying urban cultural memories, historic districts generally face problems such as chaotic traffic functions, a lack of slow traffic spaces, and insufficient public health support. Existing studies lack a public health-oriented special evaluation system and a sustainable renewal path adapted to their characteristics. This paper systematically sorts out eight core impact paths of street built environment elements on public health and constructs a healthy street evaluation system for historic districts, including six dimensions (transportation facilities, green squares, ancillary facilities, street-front commerce, urban furniture, and street network) and 30 core elements combined with the spatial and cultural characteristics of historic districts. Taking five typical streets in the Liuhe Area of Hankou Historic District as an empirical case, a comprehensive evaluation is carried out using a combination of quantitative surveys, questionnaire surveys, and spatial analyses. The results show that the overall health performance of street space in the study area is low, with extremely unbalanced development across dimensions. The core shortcomings are concentrated in incomplete slow traffic systems, lack of public spaces, prominent parking chaos, and fragmented historic styles, and the health problems of streets with different functional types show significant typological differentiation characteristics. Based on this, this paper proposes five systematic renewal strategies, transportation system optimization, public space improvement, landscape system perfection, historic style activation, and long-term mechanism construction, for achieving the synergistic goals of historic culture conservation, public health promotion, and urban sustainable development. This study not only enriches the theoretical system of research on healthy spaces in historic districts but also provides a referable evaluation framework and practical approach for modern historic districts in China and other similar historic districts with comparable spatial textures and functional characteristics. This study addresses the critical challenge of translating the profound social, spatial, and cultural dynamics of the traditional introverted Persian house into more tangible design metrics for contemporary Iranian housing. Relying on qualitative data from twenty-four diverse expert interviews across architecture, urban planning, and policy, the research demonstrates a broad consensus that the notion of replicating historical form is unsustainable. Instead, it indicates that the introverted configuration is likely a context-specific ontological imperative—viewed here as a fundamental socio-spatial requirement—rooted in measurable performance, serving simultaneous social, cultural, psychological, and environmental paradigms. The main findings show that preserving cultural continuity requires a shift from aesthetic conservation to prescriptive configuration. This logic is synthesised into a consolidated socio-spatial framework, whose originality lies in introducing three regulatory design instruments: (1) the sequenced depth and filtration protocol for spatial arrangement; (2) the controlled visual and environmental parameters for façade performance; and (3) the cultural adaptability and resilience requirement for functional programming. The framework’s prescriptive metrics, such as minimum space syntax values and the visual filtering coefficient, provide regulatory bodies with the precise technical tools necessary to enforce cultural protocols like privacy and dignity in high-density urban developments. While these metrics serve as an operationally promising model, they represent a theoretical framework that requires further empirical validation in diverse contemporary residential settings before mandatory regulatory adoption. This framework offers a pragmatic pathway for safeguarding Iranian housing’s cultural identity, ensuring future developments are certified not only for safety and structure, but for their adherence to the fundamental socio-spatial contract of the Persian dwelling. At the international level, heritage is widely recognised as a critical component of sustainable development. However, in South Asian countries such as Pakistan, India, and Bangladesh, historic cities continue to struggle to preserve and integrate built heritage amid rapid urbanisation, socio-economic transformation, and evolving contemporary urban demands. Heritage places in these contexts are shaped by complex interrelations between collective memory, the built environment, and socio-cultural identity. Yet, conservation practices have been mainly implemented through fragmented, building-by-building approaches that neglect urban-scale coherence and intangible cultural dimensions. This article addresses this gap by examining adaptive reuse as a value-based conservation strategy in historic urban quarters, where heritage serves as both a repository of cultural memory and a catalyst for sustainable, experience-based tourism. Drawing on qualitative fieldwork, heritage value assessment matrices, and doctoral research, this study uses the Saddar Bazaar Quarter in Karachi, Pakistan, as a case study to explore how tangible and intangible heritage values can be systematically integrated into conservation and regeneration processes. The findings demonstrate that heritage-led adaptive reuse, when embedded within a comprehensive urban-scale conservation framework, can sustain everyday socio-cultural practices, reinforce local identity, and enhance the legibility of historic urban environments. Rather than positioning tourism as a primary driver, the study shows that culturally sensitive and community-oriented tourism emerges as an outcome of successful heritage integration, grounded in lived urban experience rather than commodified representation. Based on these insights, the article proposes a value-based integration framework that aligns heritage conservation, adaptive reuse, and sustainable tourism within historic urban quarters. The framework offers transferable methodological guidance for revitalising heritage places and collective memories, while providing policy-relevant insights for heritage governance that support sustainability objectives, community resilience, and inclusive urban regeneration in post-colonial contexts. Background: Migration from low- and middle-income to high-income settings is often accompanied by dietary and lifestyle changes that may increase long-term risk of non-communicable diseases. African migrants represent a growing but under-studied population in Australia, with limited evidence on post-migration nutrition transitions and associated chronic disease risk. This study examined changes in diet and lifestyle among Nigerian-born adults before and after migration to Australia and explored any association with chronic diseases. Methods: A pilot cross-sectional study was conducted among adults who migrated from Nigeria to New South Wales, Australia, between 1992 and 2019. Data were collected via a culturally adapted, self-administered online questionnaire assessing socio-demographic characteristics, dietary intake, lifestyle behaviours, and self-reported chronic conditions in the 12 months immediately before and after migration. Descriptive statistics (frequencies and proportions) and inferential analyses (Chi-square tests, McNemar test, and the Bowker test) were used to compare pre- and post-migration behaviours and examine associations with chronic disease outcomes. Results: Ninety-three participants completed the survey (mean age 37.0 ± 7.2 years; 50.5% male). Post-migration, regular breakfast consumption declined (−24.3%), while irregular eating (low and moderate) patterns increased (+7.6% and +16.7%). Regular vegetable intake improved (+5.4%), whereas fruit intake remained low (13.0%). Regular consumption of Nigerian local foods decreased markedly (−53.7%), while regular intake of meat (+18.5%), dairy foods, fats (+14.3%), and non-alcoholic beverages increased (+22.8%). Salt use shifted away from the highest-risk category (−22.2%), and smoking and alcohol consumption remained low and stable. Self-reported chronic conditions were uncommon; hypertension (6.5%) and obesity (5.4%) were the most frequently reported. Conclusions: Nigerian migrants in Australia experience substantial post-migration dietary and lifestyle transitions that may elevate long-term chronic disease risk despite a currently low reported disease burden. Early, culturally responsive nutrition and lifestyle interventions are needed to support healthy adaptation and prevent the progression of cardiometabolic conditions in this growing migrant population. Antarctica’s extreme cryospheric conditions impose severe thermodynamic constraints on the natural attenuation of hydrocarbon pollutants. Despite the Antarctic Treaty System’s protections, the footprint of human logistics has left persistent reservoirs of petroleum hydrocarbons that threaten endemic biodiversity. This review critically synthesizes the state-of-the-art in Antarctic bioremediation, moving beyond traditional culture-dependent studies to integrate recent multi-omics breakthroughs (2020–2025). We analyze the molecular mechanisms limiting bioavailability in frozen soils and highlight the adaptive strategies of psychrophilic consortia, including the modification of membrane fluidity and the expression of cold-active enzymes (e.g., RHDs, AlkB). Notably, we discuss emerging findings on novel long-chain alkane degradation genes (almA, ladA) identified in 2025, which challenge previous assumptions about recalcitrance. Furthermore, the review evaluates the engineering bottlenecks of in situ versus ex situ strategies, emphasizing the synergistic potential of bacterial–fungal co-cultures and the ecological necessity of “climate-smart” remediation to mitigate methane emissions from thawing permafrost. By bridging the gap between fundamental microbial genetics and applied field engineering, we propose a roadmap for the next generation of biotechnological solutions in the warming polar environment.
