PGRMC1 Promotes the Development of Cervical Intraepithelial Neoplasia in HPV-Positive Patients

Background/Objectives: Persistent human papillomavirus (HPV) infection is the leading cause of cervical intraepithelial neoplasia (CIN), a known precursor to cervical squamous carcinoma. While progesterone receptor membrane component 1 (PGRMC1) has been implicated in various cancers, its specific role in cervical carcinogenesis has remained uncertain. This study aimed to elucidate the function of PGRMC1 in the progression of CIN. Methods: Bioinformatics techniques were employed to assess the expression levels of PGRMC1 in cervical cancer tissues and to investigate its correlation with patient prognosis. To explore the functional role of PGRMC1, we manipulated its expression in the cervical cancer cell line HeLa using siRNA. Subsequently, we evaluated cell migration via the scratch assay, and invasion through the Transwell assay. We employed mass spectrometry to identify proteins interacting with PGRMC1 and confirmed these interactions using co-immunoprecipitation (co-IP). Further co-IP experiments were conducted to pinpoint the specific binding sites of these protein interactions, and immunofluorescence staining was utilized to observe the spatial distribution of interacting proteins within the cells. The phosphorylation status of VIM was further confirmed by WB. At the clinical level, we collected cervical biopsy specimens from HPV-positive patients and verified the expression patterns of PGRMC1 and VIM using immunohistochemical staining in cervical squamous cell carcinoma (CSCC) tissues. Results: We discovered a correlation between progressively increasing PGRMC1 expression and the severity of CIN as well as a poor prognosis. Knockdown of PGRMC1 resulted in the inhibition of migration and invasion capabilities in cervical cancer cells. Furthermore, PGRMC1 was found to physically interact and colocalize with Vimentin (VIM). Notably, PGRMC1 knockdown specifically increased phosphorylation at the Ser-39 residue of VIM. Conclusions: Our findings suggest that PGRMC1 facilitates CIN progression by binding to VIM and suppressing Ser-39 phosphorylation, thereby promoting the migration and invasion of cervical carcinoma cells. This study enhances our understanding of PGRMC1’s role in CIN progression and lays an experimental foundation for targeted therapeutic approaches to cervical squamous carcinoma. Full article

Human papillomavirus (HPV) infection is a primary driver of cervical cancer. Integration of HPV into the human genome causes persistent expression of viral oncogenes E6 and E7, which promote carcinogenesis and disrupt host genomic function. However, the impact of integration on host gene expression remains incompletely understood. We used multimodal RNA sequencing, combining total RNA-seq and Cap Analysis of Gene Expression (CAGE), to clarify virus–host interactions after HPV integration. HPV-derived transcripts were detected in 17 of 20 clinical samples. In most specimens, transcriptional start sites (TSSs) showed predominant early promoter usage, and transcript patterns differed with detectable E4 RNA region. Notably, the high RNA expressions of E4 region and viral-human chimeric RNAs were mutually exclusive. Chimeric RNAs were identified in 13 of 17 samples, revealing 16 viral integration sites (ISs). CAGE data revealed two patterns of TSS upregulation centered on the ISs: a two-sided pattern (43.8%) and a one-sided pattern (31.3%). Total RNA-seq showed upregulation of 12 putative cancer-related genes near ISs, including MAGI1-AS1, HAS3, CASC8, BIRC2, and MMP12. These findings indicate that HPV integration drives transcriptional activation near ISs, enhancing expression of adjacent oncogenes. Our study deepens understanding of HPV-induced carcinogenesis and informs precision medicine strategies for cervical cancer. Full article

Advanced composite materials and manufacturing technologies are critical to sustain human presence in space. Mechanical testing and analysis are needed to elucidate the effect of processing parameters on composites’ material properties. In this study, test specimens are 3D printed via a fused-filament fabrication (FFF) approach from a basalt moon dust-polylactic acid (BMD-PLA) composite filament and from pure PLA filament. Compression and tensile testing were conducted to determine the yield strength, ultimate strength, and Young’s modulus of specimens fabricated under several processing conditions. The maximum compressive yield strength for the BMD-reinforced samples is 27.68 MPa with print parameters of 100% infill, one shell, and 90° print orientation. The maximum compressive yield strength for the PLA samples is 63.05 MPa with print parameters of 100% infill, three shells, and 0° print orientation. The composite samples exhibit an increase in strength when layer lines are aligned with loading axis, whereas the PLA samples decreased in strength. This indicates a fundamental difference in how the composite behaves in comparison to the pure matrix material. In tension, test specimens have unpredictable failure modes and often broke outside the gauge length. A portion of the tension test data is included to help guide future work. Full article

We investigated respiratory virus epidemiology in older adults across pre-pandemic (2007–2019), pandemic (2020–2022), and post-pandemic (2023–2024) periods, focusing on how public health interventions shaped surveillance, prevalence, and sex-specific trends. We conducted a retrospective cross-sectional study at a 1000-bed tertiary hospital in the Republic of Korea during 2007–2024, analyzing 4692 nasopharyngeal swab specimens collected from adults aged ≥ 65 years with suspected respiratory infections during 2007–2024. The specimens were tested for 15 respiratory viruses using multiplex real-time polymerase chain reaction. The outcomes included virus-specific detection rates and seasonal, sex-based and temporal trends before and after the COVID-19 pandemic. During the pre-pandemic period, older adults accounted for 13.2% of the tested individuals, which significantly increased to 52.0% in the later periods. Influenza A was the most frequently detected virus, followed by rhinovirus and human metapneumovirus. Influenza, RSV A/B, and coronaviruses 229E and OC43 showed peak positivity in winter, parainfluenza virus type 3 peaked in summer, and rhinovirus circulated year-round. Virus circulation was markedly suppressed during 2020–2022 and partially rebounded during 2023–2024. This study highlights the shift in diagnostic access and epidemiologic patterns of respiratory virus infections in older adults following the COVID-19 pandemic. Full article

The proper selection of bioactive root-end material is one of the main prognostic factors for the successful healing outcome of apical microsurgery (AMS). The aim of the present in vitro study was to evaluate and compare the marginal adaptability of a novel calcium silicate cement (CSC), Harvard MTA Universal, and Biodentine used as root-end filling materials. The endodontic treatment of 20 extracted human maxillary central incisors was performed. The apicoectomy was simulated, and root-end cavities were prepared ultrasonically using universal retrotips. Teeth were randomly assigned into two equal groups (n = 10) according to the retrofilling cement used: Group 1—Harvard MTA Universal and Group 2—Biodentine. The specimens were stored in relative humidity for 48 h and sectioned longitudinally. The data were processed and analyzed statistically. Harvard MTA exhibited a significantly lower mean gap width (1.16 ± 0.37 µm) than Biodentine (2.48 ± 0.38 µm) (p < 0.05), indicating a more intimate interfacial adaptation. Additionally, the phenomenon of material penetration into the dentinal tubules was observed only in the Harvard MTA group. Within the limitations of this in vitro study, Harvard MTA Universal demonstrated better interfacial properties than Biodentine when applied as a root-end filling material. This novel biomaterial could be regarded as a promising alternative for earlier calcium silicate cements in the context of AMS goals. Clinical relevance: The quality of marginal adaptation is a determinative feature for the clinical performance of CSCs and the long-term prognosis of AMS. Full article

Background: Primary hepatocellular carcinoma (HCC) and liver metastases differ in origin, progression, and therapeutic response, yet a direct high-resolution spatial comparison of their tumor microenvironments (TMEs) within the liver has not previously been performed. Methods: We applied high-definition spatial transcriptomics to fresh-frozen specimens of one HCC and one liver metastasis (>16,000 genes per sample, >97% mapping rates) as a proof-of-principle two-specimen study, cross-validated in human proteomics and patients’ survival datasets. Transcriptional clustering revealed spatially distinct compartments, rare cell states, and pathway alterations, which were further compared against an independent systemic dataset. Results: HCC displayed an ordered lineage architecture, with transformed hepatocyte-like tumor cells broadly dispersed across the tissue and more differentiated hepatocyte-derived cells restricted to localized zones. By contrast, liver metastases showed two sharply compartmentalized domains: an invasion zone, where proliferative stem-like tumor cells occupied TAM-rich boundaries adjacent to hypoxia-adapted tumor-core cells, and a plasticity zone, which formed a heterogeneous niche of cancer–testis antigen–positive germline-like cells. Across both tumor types, we detected a conserved metabolic program of “porphyrin overdrive,” defined by reduced cytochrome P450 expression, enhanced oxidative phosphorylation gene expression, and upregulation of FLVCR1 and ALOX5, reflecting coordinated rewiring of heme and lipid metabolism. Conclusions: In this pilot study, HCC and liver metastases demonstrated fundamentally different spatial architectures, with metastases uniquely harboring a germline/neural-like plasticity hub. Despite these organizational contrasts, both tumor types converged on a shared program of metabolic rewiring, highlighting potential therapeutic targets that link local tumor niches to systemic host–tumor interactions. Microplastic pollution is pervasive in marine ecosystems and poses a growing threat to marine organisms and human health. This study simultaneously investigates microplastic ingestion and phthalate exposure in Parapenaeus longirostris, a commercially valuable and ecologically relevant Mediterranean crustacean occupying an intermediate trophic position. Specimens were collected from three coastal areas in the central Tyrrhenian Sea (Western Mediterranean): near the Tiber River mouth, one of the most polluted rivers in Italy, and two additional sites to the north and south. The frequency of individuals with ingested microplastics varied among locations: 78% near the Tiber River, 64% at site S, and 38% at site N, reflecting anthropogenic pressure gradients. Analyses confirmed the lower occurrence at site N, indicating higher ingestion near land-based pollution sources. Ingested microplastic polymer types varied among sites, reflecting location-specific contamination. Phthalates were present in shrimp muscle at all sites (5–1122 ng/g w.w.) with the highest average concentration (68.26 ± 55.74 ng/g) at the site with the highest microplastic ingestion. Although no statistical correlation was found, the similar spatial distribution of microplastics and phthalates suggests a potential link influenced by local pollution and individual variability. These findings provide novel evidence of microplastic and phthalate contamination in P. longirostris, highlighting its role as a trophic connector mediating contaminant transfer through the food web. While current levels suggest no potential risk to human health, continued monitoring and further studies on exposure along trophic pathways are recommended. Full article

Funerary archaeoentomology is the discipline that studies insects and other arthropods in archaeological contexts, with a particular focus on the funerary domain. The presence of specific species, such as necrophagous beetles or saprophagous flies, can provide crucial evidence regarding post-mortem conditions—whether bodies were left exposed to the air or buried suddenly after death—and whether they underwent particular preservation practices, such as desiccation or embalming. This study concentrates on entomological specimens collected from three mummified bodies at the Sanctuary of Madonna della Corona in the province of Verona (northeast Italy), aiming to reconstruct aspects of funerary practices, especially the season of death and the authenticity of the garments worn by the mummified individuals. Insects were manually collected from bodies belonging to three hermits living between the 17th and 19th centuries. A complex entomofauna consisting of Diptera, Coleoptera, Lepidoptera, and minor taxa was collected and analyzed. Diptera puparia, primarily from the families Calliphoridae, Muscidae, and Fanniidae, were the most abundant entomological elements recovered. Their presence suggests potential exposure of the bodies before burial and indicates that death likely occurred during a mild period of the year (end of spring/beginning of autumn). The co-occurrence of holes caused by maggots on the hermits’ skin and their garments allows us to speculate about the authenticity of the clothing used during the funerary rituals. By combining entomological evidence with textile analysis, this research offers a more precise understanding of historical funerary practices within this devotional context. It sheds light on methods of managing human remains, burial traditions, and preservation techniques, particularly regarding the clothing of the deceased. Full article

Background: Oncogenic fusions of MAML2 with CRTC1, CRTC3, YAP1, and NR1D1 retain the MAML2 transactivating domain (TAD) and are believed to drive aberrant gene transcription. While the oncogenic roles of these known fusions have been established, we aimed to identify novel MAML2 fusions across a range of human malignancies. Methods: DNA and RNA sequencing were performed on tumor samples submitted to Caris Life Sciences. MAML2 fusions were identified from RNA transcripts and filtered to include only known pathogenic fusions or recurrent, in-frame fusions containing a C-terminal MAML2 TAD. Fusion burden was defined as the number of unique fusion isoforms per sample. Results: Among 180,124 tumor samples, 143 specimens harbored MAML2 fusions with a MAML2 TAD: >50% of specimens harbored known fusions, but novel fusions with MTMR2 (31/143), SESN3 (11/143), CCDC82 (6/143), FAM76B (4/143), and ATXN3 (3/143) were also identified. Compared to the known fusions, the novel fusions generally had lower expressions (median: 8 vs. 13 junction reads/sample, p = 0.0064), higher fusion burdens (median: 6 vs. 2 unique fusion isoforms/sample, p < 0.0001), more frequent TP53 co-mutations (80% vs. 11.5%, p < 0.0001), and no clear association with the tissue of origin. Excluding ATXN3::MAML2, the novel fusion partners were located near MAML2 in the genome, likely arose from duplications or deletions, and occurred in samples harboring concurrent mutations. In contrast, ATXN3::MAML2 arose via interchromosomal translocation, occurred in samples with a low fusion burden, and was not associated with TP53 mutations. Conclusions: We identified novel MAML2 fusion partners, most of which likely represent passenger alterations, possibly arising from genomic instability or impaired p53 function. However, ATXN3::MAML2 fusions, previously reported in a pre-cancerous pancreatic disease case, may represent a pathogenic alteration warranting further investigation. Full article

Background: To overcome the esthetic limitations of dental monolithic zirconia restorations, multichromatic systems were developed to combine improved structural integrity with a natural shade gradient that mimics the optical properties of natural teeth. In response to the clinical demand for time-efficient, i.e., chairside fabrication of zirconia restorations, rapid sintering protocols have become necessary to adjust clinical efficiency along with material performance. This study addresses the challenges of a rapid sintering protocol related to optical performance and phase transformation of the final restoration and the zirconia–cell interaction. Methods: The influence of a rapid sintering protocol on the color stability of the final dental restoration was evaluated by the CIE L*a*b* color space. Phase transformation was assessed through X-ray diffraction analysis. Cellular behavior was evaluated by measuring wettability, the material’s surface energy, and a cell mitochondrial activity assay on human gingival fibroblasts. Results: Optical measurements demonstrated that the total color change in all layers after rapid sintering was above the perceptibility threshold (ΔE* > 1.2), while only the polished enamel layer (ΔE* = 3.01) exceeded the acceptability threshold (ΔE* > 2.7), resulting in a clinically perceptible mismatch. Results of X-ray diffraction analysis, performed for fixed occupancy at Z0.935Y0.065O0.984, revealed that rapid sintering caused a decrease in the cubic (C-) phase and an increase in the total amount of tetragonal (T-) phases. Conventionally sintered zirconia consists of 54% tetragonal (T-) and 46% cubic (C-) phase, whereas in the speed-sintered specimens, an additional T1 phase was detected (T = 49%; T1 = 27%), along with a reduced cubic fraction (C = 24%). Additionally, a small amount of the monoclinic (M) phase is noticed. Although glazing as a surface finishing procedure resulted in increased hydrophilicity, both polished and glazed surface-treated specimens showed statistically comparable cell adhesion and proliferation (p > 0.05). Conclusions: Rapid sintering induced perceptible color changes only in the enamel layer of multichromatic zirconia, suggesting that even layer-specific alterations may have an impact on the overall esthetic outcome of the final prosthetic restoration. Five times higher heating and cooling rates caused difficulty in reaching equilibrium, leading to changes in lattice parameters and the formation of the metastable T1 phase. Full article

Objective: To investigate the influence of chitosan-based nano-silver fluoride (CNSF) treatment of tooth tissues on shear bond strength (SBS) of resin composite (RC) and resin-modified glass ionomer (RMGI) restorations. Methods: 90 extracted human molars were collected. Specimens were randomly assigned to three groups (n = 30): non-pretreated (NPT) pretreated with either CNSF or silver diamine fluoride (SDF). Each group was subdivided into two restorative subgroups (n = 15): RC and RMGI. Specimens in CNSF and SDF groups were pretreated with CNSF or SDF per their manufacturer’s instructions. Then specimens in RC subgroups were etched, treated with chlorhexidine cleanser, followed by adhesive application. Specimens in RMGI subgroups were treated with cavity conditioner only. A cylindrical restoration (2.38 mm θ × 2 mm height) of RC or RMGI restoration was fabricated with a standardized mold and light-cured on all specimens. After 5000 times of thermocycling between 5 °C and 55 °C with dwell times of 30 s intervals, SBS was measured using the Ultradent UltraTester. Data was analyzed statistically (α = 0.05) using ANOVA/Tukey’s comparisons. Results: No statistically significant difference in SBS among RC restorations in the three treatment groups: NPT (17.48 ± 3.96), CNSF (18.38 ± 5.59), and SDF (14.03 ± 6.56). For RMGI restorations, SBS was significantly (p < 0.05) higher in NPT (15.99 ± 3.59) compared to CNSF-treated (11.45 ± 5.48), but there was no significant difference between NPT and SDF-treated (14.27 ± 2.17) or between SDF- and CNSF-treated groups. Conclusions: No difference in SBS of resin composite restorations when the dentin tissue is pretreated with either chitosan-based nano-silver fluoride or silver diamine fluoride. However, SBS of RMGI was lowered when the dentin tissue was pretreated with CNSF. Full article

Keel bone lesions (KBLs) represent a relevant welfare concern in laying hens, arising from complex interactions among genetics, housing systems, and management practices. This study presents the development of an image analysis system for the automated detection and classification of KBLs in slaughterhouse videos, enabling scalable and retrospective welfare assessment. In addition to lesion classification, the system can track and count individual carcasses, providing estimates of the total number of specimens with and without significant lesions. Videos of brown laying hens from a commercial slaughterhouse in northeastern Italy were recorded on the processing line using a smartphone. Six hundred frames were extracted and annotated by three independent observers using a three-scale scoring system. A dataset was constructed by combining the original frames with crops centered on the keel area. To address class imbalance, samples of class 1 (damaged keel bones) were augmented by a factor of nine, compared to a factor of three for class 0 (no or mild lesion). A YOLO-based model was trained for both detection and classification tasks. The model achieved an F1 score of 0.85 and a mAP@0.5 of 0.892. A BoT-SORT tracker was evaluated against human annotations on a 5 min video, achieving an F1 score of 0.882 for the classification task. Potential improvements include increasing the number and variability of annotated images, refining annotation protocols, and enhancing model performance under varying slaughterhouse lighting and positioning conditions. The model could be applied in routine slaughter inspections to support welfare assessment in large populations of animals. Full article

Purpose: Cadaveric dissection is a cornerstone of neurosurgical education, providing trainees with a realistic 3D understanding of anatomy and a safe environment to practice surgical approaches. A preservation technique was developed that merges the advantages of fresh-frozen and embalmed cadavers, maintaining tissue realism while enhancing durability. This approach preserves flexibility and natural color, improves anatomical detail, and creates a safe, long-lasting model ideal for neurosurgical training. Methods: Four specimens were thawed, cannulated, and irrigated before implementing a protocol consisting of low concentration formaldehyde with glycerol and ethanol for extended preservation. The specimens were prepared for both neurosurgery training and educational purposes, and their condition was evaluated with a semi-quantitative scale. Each specimen was evaluated independently by two raters, blinded to the time-point, using a semi-quantitative scale anchored to predefined criteria (0–3 per domain). Inter-rater reliability was calculated using the intraclass correlation coefficient (ICC [2,k]) for continuous scores and Cohen’s κ for categorical agreement. Results: Over nine years of intermittent use, the specimens remained in good condition: tissues retained sufficient softness for dissection, injected vessels stayed vivid in color, and no foul odor or microbial growth was observed. The evaluation employed a semi-quantitative scale, with results ranging from 11/14 to 14/14. The mean values demonstrate stable tissue quality over time, with only minor variations in color and perfusion. The inter-rater reliability was high (ICC = 0.91; κ = 0.88). Conclusions: The preservation method leverages the strengths of both fresh-frozen and embalmed models. The results suggest feasibility of long-term reuse, although further quantitative validation is needed. Full article

Background/Objectives: This study explores the potential of the inducible G protein-coupled kinin B1 receptor (B1R) as a target for the diagnosis and treatment of prostate cancer (PCa) and aims to develop the first theranostic agent targeting hB1R for both molecular imaging and radionuclide therapy. Methods: B1R expression was analyzed via qPCR and immunohistochemistry in human PCa cells and tissues specimens. A novel 64Cu/NOTA-conjugated peptide analog of the potent B1R antagonist R954 was synthetized and evaluated in vitro and in vivo. Results: B1R was confirmed to be expressed (RNA, protein) by varying degrees in all PCa cell lines and tissues investigated, with protein level significantly correlating with tumor grades. This finding was supported by similar analyses from the TCGA and MSKCC databases. In vitro, the 64Cu/NOTA-βAla-R954 conjugate showed nanomolar affinity/potency at hB1R, complete plasma stability over 24 h, significant cellular uptake (up to 33% of ID at 24 h), and dose-dependent anti-clonal growth effects. In vivo, the radioconjugate remained stable in circulation for up to 90 min and was primarily excreted intact via the kidneys following IV administration. Intravenous 64Cu/NOTA-βAla-R954 (7.5 MBq) effectively detected subcutaneous PCa xenografts via µPET imaging in male athymic nude mice. At a single higher dose (65 MBq; 50 µg/kg), it significantly reduced tumor growth without observable toxicity. This antitumor effect was associated with increased apoptosis (active caspase-3) and reduced proliferation (Ki67), as shown by immunohistochemistry. In contrast, the nonradioactive NatCu/NOTA-βAla-R954 had no therapeutic effect at the same dose. Conclusions: Our findings provide proof-of-concept for the potential theranostic use of 64Cu/NOTA-R954 in PCa, and potentially other types of B1R-positive solid cancers. Full article

Rickettsia parkeri strain Atlantic rainforest is an emerging pathogen in Brazil, but human infections remain rarely reported. We report the first confirmed case in the municipality of Caraguatatuba on the northern coast of São Paulo State, Brazil. A 37-year-old man was bitten by an Amblyomma ovale tick while visiting a forested area. Six days later, he developed a papular skin lesion with a necrotic center at the tick bite site, accompanied by regional lymphadenopathy, myalgia, and flu-like symptoms. Ticks parasitizing both the patient and his dog were collected, along with the eschar. Two ticks were analyzed for Rickettsia isolation, and R. parkeri was successfully isolated from one infected specimen. The patient’s infection was confirmed by molecular testing through the PCR amplification of the gltA and ompA genes from an eschar inoculation sample. This represents the sixth confirmed case of rickettsiosis caused by R. parkeri in Brazil, and it reinforces the need for increased epidemiological surveillance in endemic regions for spotted fever caused by this pathogen. With the increasing recognition of R. parkeri in South America, further research is needed to better understand its transmission dynamics, clinical manifestations, and potential public health impact.
