Title:  Development of Sakon Nakhon Rice Variety for Blast Resistance through Marker Assisted Backcross Breeding.

The work presented here is interesting, falls within the scope of the journal and is well worth investigating. In the present study, the authors report breeding of resistant Sakon Nakhon using marker-assisted backcrossing, together with a phenotypic selection of resistant strains to Pyricularia oryzea.  Material and methods are not described in detailed regarding on how SSR Markers was made. The discussion is described clearing but could be improved by explaining why the phenotypic selection of resistant strains is better than genotype.

The manuscript is entitled to the development of important rice variety, Sakon Nakhon against blast disease via Marker assisted Backcross breeding. The major objective of this study is to develop blast resistance but also retain desirable aromatic traits of sakon Nakhon rice. The manuscript was written well with adequate supporting information and results, however, there are few specific issues, the authors can clarify or modifying in their response.

1.     Gel picture showing molecular marker analysis for the foreground selection with the SSR markers for blast resistant alleles

2.      Authors are recommended to include field validation picture of  BC2F4 progenies, which shows blast resistance

3.     Including graphical representation of QTL on the chromosomes, may help readers to understand better on different combination of QTL in introgression lines

In this paper authors present the genetic improvement of a high quality rice variety by increasing its resistance to blast disease. This approach is useful for rice genetic improvement and possible use of the new variety in diferent environments (upland and lowland). Language in clear and requires minor revisions. For example, all scientific names must go on italics and once a species is first cited there is no need to write the full name next time, abbreviations must be used. Also, it is not clear the number of BC2F3 progenies obtained, since 13 were selected for greenhouse validation, but Figure 1 specifies that 7 is the number obtained. 

Finally, it is no clear the method employed for the QTL analysis in the lines selected. A table or graphic with QTL representation per chromosomes would be usefull for clarifying this issue.
