Special Issue Information

Dear Colleagues,

This Special Issue on "Molecular Mechanisms and Biological Regulation in Membrane Transport" in the International Journal of Molecular Sciences focuses on the conformational dynamics of membrane proteins, particularly transporters. These proteins facilitate the movement of small molecules across membranes through an alternating access mechanism. This process involves the initial binding of substrates on one side of the membrane in an outward-facing state, followed by a transition to an inward-facing state on the opposite side, where the cargo is released. Understanding the range of metastable conformations, their energetic barriers, and driving forces is essential for deciphering the structural transformations in membrane transporters. This knowledge enables for the exploration of intermediate states, underlying pathways, and dynamics, ultimately revealing transporter functions at a molecular level and aiding the development of targeted therapeutics. However, the conformational dynamics of transporters within the lipid bilayer have proven challenging to elucidate, limiting our ability to decipher their mechanisms and develop effective therapeutics.

This issue emphasizes the importance of understanding how membrane proteins are modulated at the mechanistic level as well as the complex interplay between membrane proteins and lipids for future advances in membrane protein research and targeted therapeutics.
