Novel Insights on the Corpus Luteum Function: Role of Vaspin on Porcine Luteal Cell Angiogenesis, Proliferation and Apoptosis by Activation of GRP78 Receptor and MAP3/1 Kinase Pathways
Round 1
Reviewer 1 Report
The authors have examined the role of Vaspin on the angiogenesis in the Porcine luteal cells. The study design is good and the results are supporting for the conclusion. There are 2 minor points needing to be addressed.
- A vehicle-trearted control group is needed
- As Vaspin could promote cell proliferation, the VEGF-A sscretion level should be adjusted or the authors can add discussions on this aspect

Comments on Kurowska et al.
This manuscript by Kurowska et al. is identical to recently published article (PMID: 31752432) by the same author. They just changed the cell type. This manuscript is well structured and nicely written.
Minor comments:
Some better images of western blots are expected. For example, Fig. 1C, and 3C.
Lines 380-381; Please replace ‘ml” with µl”. …………….….performed with 100 ng cDNA, 1 mL TaqMan GeneExpression primers, and 10 mL TaqMan PCR master mix (Applied Biosystems) in a final reaction volume of 20 mL.
Line 429, Please correct the sentence by adding “s” to “stimulate” and “inhibit”……………….Vaspin stimulate angiogenesis and proliferation, while inhibit apoptosis by activation…………

We thank Reviewer for his/her thorough review and comments. In previous article (PMID: 31752432) we showed vaspin effect on porcine follicular: granulosa cells proliferation, cell cycle progression and apoptosis, as well as molecular mechanism of it action. In present article we focused on vaspin action on luteal cells: angiogenesis process, which is essential for CL formation and function establishment. Moreover, as confirmation of vaspin function on CL formation we examined it effect on luteal proliferation and apoptosis.
Please find below our response to minor concerns regarding the present work.
Minor comments:
Some better images of western blots are expected. For example, Fig. 1C, and 3C.
Better images of fig. 1C and 3C were provided and some new densitometry analysis was performed.
