In this extensive review, Lejman et al., have reviewed most of the known genomic abnormalities related to acute lymphoblastic leukemia. The literature and references are fine and it seems to me that the authors have reviewed and prepared a really nice and informative manuscript. In my opinion, they have included the most common translocations/mutations that serve as biomarkers in clinics for ALL, the molecular pathways and proteins downstream these biomarkers, and current and potential therapies based on them. 

I have to thank and congratulate the authors for bringing together this information and I recommend this manuscript for publication in IJMS.

Lejman et al have written a timely and extensive review on ALL. The manuscript is well written with clear organization. Here are some of my comments.

  1. It would be helpful if the readers add schematic to section 2 to make it easier for the readers
  2. It would also be helpful if the authors mention some of the current treatment methods in the introduction section and better emphasize the need to study these genetic biomarkers

Overall, the authors have done a substantial job in writing this manuscript.
