The authors have presented a significant review article deciphering the mechanism associated with COVID-19 which can help future researchers to develop lead candidates for its efficient management. The coronavirus disease 2019 (COVID-19) became a worldwide concern at the beginning 13 of 2020, and since then it has affected millions of people. In addition, this review also proposes the potential of TNF-α 25 as a biomarker for the prognosis of severe COVID-19 and its related complications, and as a molecular target for anti-TNF-α therapy. All the sections are well explained and correlated with each other. The flow of the review article is very good. The review is very well framed with good references. Hence, this review article can be accepted after reframing the title because the title is not very strong.

Dear Authors

 

The article titled Association of “Tumor Necrosis Factor-Alpha (TNF-α) with 2 Morbidity and Mortality of COVID-19” by Z.M. Zawawi et al., reviewed the role of TNF-a in promoting inflammation, and how this influences COVID-19 morbidity and mortality. The authors clearly introduced the topic and the issues that relate to TNF-a. The authors were able to clearly discuss the importance of this work in a clear and concise manner by dividing the article into easy-to-follow section, clearly explaining pathways, relating each explanation to prepared figures and referencing important findings in the field. I congratulate the authors on preparing a well written and topic review article which I enjoyed reading!

This review provides an in-depth overview of the roles of TNF-a in the pathogenesis of COVID-19. The manuscript is well-organized and offers valuable information about the application of TNF-a in monitoring and treating COVID-19. However, several issues are needed to be addressed.

 

1.      On page 5, lane 176, references 25 and 40 are inappropriate for this sentence. Please choose other papers to support this opinion.

2.      On page 6, lane 233, reference 54 is not suitable.

3.      Please check the sentence “The association of hyperactivation of NF-κB via the pathogen pattern recognition-the myeloid differentiation primary response 88 (PPR-MyD88) pathway and SARS and MERS patients has been documented”. (page 6, lanes 261-263)
