1. Is the methodology designed by the Authors? If not, please provide appropriate references.

2. Could the Authors provide correlation analyses for the performed test?

3. The provided references are not up to date. Only 4 out 25 of the refs cited are of the last 5 years (2019-2024) and many refs are old or very old: this is strange because we should give the readers the last updates and we should discuss in the view of latest findings.

1.     This manuscript presents an in vitro study on the anticancer agent Cacalol Acetate. The anticancer effects were investigated across various cellular physiological processes, but only the HeLa cell line was used. It seems necessary to conduct key experiments on other cancer cell lines. Alternatively, the title of the paper could specify that the effects were observed in the HeLa cell line.

2.     The legend mentions three repeated experiments and provides explanations about p-values, but Figures themselves lack p-value indications marked by asterisks (*). It is necessary to include p-value indications for key comparisons in Figures that support the main conclusions.

 

3.     The comparative experiments between CA and I3C were well conducted, but the specific gene expression changes induced remain unclear. It would be beneficial to conduct supplementary experiments to observe the differences in various intracellular stress signaling pathways (MAPK, ERK, JNK) or immediate early genes (IEGs; e.g., jun, fos) shortly after CA and I3C treatment.
