The paper is very well written and extremely comprehensive. The authors have spoken about the disadvantages and the shortcomings of using DDR based drugs for solving CRC drug resistance problems.

-       Major concern

-       1. The manuscript lack of new ideas or perspective/future direction /. It is only provide the summary of previously published information that minimized the enthusiasm.

-       2. The manuscript also presents the idea overcoming treatment resistance via targeting DNA damage response. The author should provide a clear picture how the IR in combination with DDR/or repair inhibitor such as PARPi induces better treatment response in CRC.

Minor comments

The Simple summary section needs to be clear and concise. At the current form it is confusing with many abbreviation

Table 1 may be included as supplement information.

The review article entitled Targeting the DNA Damage Response and DNA Repair Pathways to Enhance Radiosensitivity in Colorectal Cancer is relevent in this field of research. However, I don't think the quality of this review article is high enough to be published in this journal.

Despite providing just one image, the authors should have provided more graphical representations in this article.

 

It does not appear that there is any redundancy in the abbreviation used in the abstract section. The authors elaborated on CSC, CRC, etc., but didn't provide the same for ATM/ATR, CHK1/CHK2, WEE1 and PARP1.

 

Reference 3 is more than a decade old to mention the 5-year survival rate. Update with latest citations. 

The manuscript contains a number of old references. It would be appropriate for authors to update their references with recent publications in this area of research.
