Molecular Alterations in Intraductal Carcinoma of the Prostate
Round 1
Reviewer 1 Report
Comments and Suggestions for AuthorsThe article entitled “Molecular alterations in Intraductal Carcinoma of the Prostate” from Marit Bernhardt and Glen Kristiansen represents an effort to review the molecular alterations associated with intraductal prostatic neoplasias (IDC-P), extensively mentioned at the literature but poorly studied. Loss of PTEN, TP53, BRCA, ATM, FOXA1 mutations (among others), TMPRSS2::ERG fusion or the upregulation of some lncRNAs are frequently associated with IDC-P. A table with the results would be helpful.
Along the article, there are some minor mistakes, for example:
Pag. 1, line 9: “IDC - P can be present either in association with a high grade prostate cancer or independent of any high grade or even invasive tumor at all. IDC - P in association with a high grade cancer. In this Review we….” Incomplete, repeated phrase
Pag. 2, line 92: “…are frequentlyasuwah evaluated concomitantly….”
I recommend its publication with minor changes.

Comments and Suggestions for AuthorsBernhardt and Christiansen provide a comprehensive review on the molecular alterations reported in IDC-P, emphasizing the dissimilarities between isolated forms and those (much more common) associated with invasive adenocarcinoma.
The paper is very well organized, it reads well and constitutes a very nice addition to the field, providing a quite useful overview.
Some suggestions for further improving the quality of the work:
1. There are a few studies (not many, indeed) addressing epigenetic alterations, and more specifically, DNA methylation in IDC-P. The addition of these data would complete the "molecular" overview of the entity.
2. A table summarizing the main molecular alterations and, eventually, an illustration depicting the main pathways would further enhance the readibility of the paper and facilitate its dissemination.
3. Some minor issues requiring correction:
a. Simple summary, lines 9-10: Please clarify the sentence "IDC-P in association with..."
b. Line 92: please correct the typo "...frequentlyasuwah..."

Some suggestions for further improving the quality of the work:
- There are a few studies (not many, indeed) addressing epigenetic alterations, and more specifically, DNA methylation in IDC-P. The addition of these data would complete the "molecular" overview of the entity.
Answer: We added a paragraph to explain and include epigenetic alterations.
- A table summarizing the main molecular alterations and, eventually, an illustration depicting the main pathways would further enhance the readibility of the paper and facilitate its dissemination.
Answer: We added a Table with common molecular alterations.
- Some minor issues requiring correction:
- Simple summary, lines 9-10: Please clarify the sentence "IDC-P in association with..."
- Line 92: please correct the typo "...frequentlyasuwah..."
Answer: We have carefully revised the paper and corrected the aforementioned mistakes.
