The review article discusses caged protein, problems and advances in their applications.

The issues that should be corrected are the following:

  1. The article title is too broad, and should be restrict to the topics that are discussed in the manuscript. The same is about the abstract. It should be a short summary of a review, rather than a set of very general sentences.
  2. Is the manuscript focused on different caged techniques with respect to study of mechanisms or functions or the primary interest is genetic techniques? The focus should be more pronounced in the manuscript.
  3. Figures 3-5 contain experimental data. The references should be explicitly mentioned on figure caption. Does the author have permission for their reprinting?
  4. I wonder whether it is ok to acknowledge so many source of funding for one author.

This review is reflects the current progress in the field of protein photoactivation. But I have one point, and I hope the authors could add something about the application of protein photoactivation and discuss its potential applications in the future. Although it has mentioned sporadically in the text, it should be a sufficient supplement to attract more readers from different fields, especially for readers who are interested in applying protein photoactivation technology, which will be of great interest.

The review article summarizes the techniques involved in photo-activation of proteins. The paper can be published after incorporating a few additional details.

(1) There needs to be a section on photocleavable proteins used by the optogenetic community 

(2) Other light based proteins including the LOV domain could make the review article more comprehensive
