The manuscript on Metabolite profile of Athrixia phylicoides DC. (Bush tea) and determination of inhibitory mechanism against mushroom tyrosinase is well written and sound. Methods used are appropriate and results are well discussed.

Methods: Why was methanol the only solvent used for extraction? Further study should include the use of more solvents for extraction.

Do the authors intend to check and identify the exact metabolite responsible for the activity of the plant under study? It would be valuable to do this.

Comment: Methods: Why was methanol the only solvent used for extraction? Further study should include the use of more solvents for extraction.

Response: Literature (Dhawan and Gupta et al., 2017) has reported on methanol as an effective extraction solvent for various phytochemicals hence the solvent was chosen for the present study. Various solvents of varying polarities are been investigated for their inhibitory effect on the enzyme.

 

Comment: Do the authors intend to check and identify the exact metabolite responsible for the activity of the plant under study? It would be valuable to do this.

Response: The tyrosinase-based TLC autography was an effective assay for highlighting those metabolites and hence the on-going studies are on isolating and purifying the active metabolite and incorporation in a cosmeceutical product.

The manuscript entitled " Metabolite profile of Athrixia phylicoides DC. (Bush tea) and determination of inhibitory mechanism against mushroom tyrosinase" mainly focuses on use of plant extract enzyme inhibition and cytotoxic effects. From the contents of the manuscript to estimate, this research is good in the qualitative and quantitative analysis. In my judgement, it needs to be revised for publication in this journal.

  Title: is tyrosinase is a mushroom? “mechanism against mushroom tyrosinase” could be revise as “against an enzyme tyrosinase from mushroom”  

Abstract

Abstract is well written. However, results should be further specified specifically in quantitative values.

Also add which mushroom was used for extraction of tyrosinase

Introduction

Intro is very short; it needs modification for example the author should provide general background of the topic.

Intro should be start from the potential of medicinal plants in against various disease and specifically against skin diseases. The following studies could be helpful and cited.
