Dear authors

1. Novelty of this manuscript lies in the tested algae strain which is Parachlorella kessleri  

2. How authors can claim that growth in TAP  media  is autotrophic growth? Tris-acetate media has organic carbon in its composition. Autotrophic growth is only possible if algae growth only on CO2 and light. Therefore, authors are encouraged to run an experiment with TAP media without adding acetic acid or any other organic in the media. 

3. Authors must measure the glucose uptake by the microalgae under mixotrophic and heterotrophic mode of cultivation and assess the increase in the lipids  and biomass contents per unit amount of glucose consumed.

4. Though its choice of the authors to cite the relevant studies, but due credit must be given to authors and publishers. I have not seen any citation of articles from Fermentation journal.

Introduction:

·        Cite and discuss more relevant past literature on the effects of trophic modes on microalgal growth and composition. The introduction currently only cites 9 references, which is quite low.

·        More information could be provided on why glucose was selected as the organic carbon source.

·        Provide more context about Parachlorella research. The authors could cite studies examining this genus specifically.

·        The innovation of this study as compared with the existing literature, should be clearly stated in the introduction. I listed some references that may help you

Materials and methods:

·       The authors should clarify if they isolated the microalgal strain by themselves. Because if so they need to clarify the enrichment and isolation methods used and screening criteria and tests conducted to identify and select this strain

·       Providing glucose concentrations tested in initial screening experiments in the results would aid interpretation.

·       In Line #117 "After 24 h, it was removed," I kindly request clarification regarding what exactly was removed after the specified time period.

·       In some instances, "mg.L-1" is used, while in others, "mg L-1" is employed. To ensure consistency and clarity throughout your work, I kindly suggest unifying the formatting for this unit.

·       Line# 127 “In order to further study the effect of nitrogen deficiency on the photosynthetic” It appears that there might be a mismatch between the statement and the actual content of your study, as you did not investigate the effect of nitrogen deficiency as mentioned in this sentence. Please consider revising this statement to accurately reflect the scope and findings of your research.

·       I've noticed a repetition in your manuscript between Lines 137-140 and Lines 134-146. The content appears to be duplicated in these sections.

·       Another repetition in your manuscript between Lines 179-184 and Lines 191-197. The content seems to be duplicated in these sections.

·       Please replace algal mud by algal cell or pellet and algal solution by alga culture throughout the manuscript

Results

·       Line # 205: Parachlorella kessleri in subtitle should be in italic and should be appreciated to P. kessleri after the first mention.

·       Line# 207: correct figure 5.1 to figure 1

·       I suggest ensuring that the y-axis maximum value is uniform across all subplots in Figure 1.

·       I've noticed that there are citations within the results section of your manuscript, specifically in Line 247, 259, 275, and possibly others, that appear to be more appropriate for inclusion in the discussion section.

·       I would like to request clarification regarding the glucose concentration used for the reset experiment after the initial experiments with different concentrations.

·       I recommend adding a time course reference to the legend for Figures 1, 2, 3, 4, 5 to provide a more comprehensive description of the data.

·       Figure 2 repeated the data of figure 1

·       I recommended moving carbohydrate and protein section before the lipid section.

Discussion

·       The discussion could be strengthened by more thoroughly explaining mechanisms behind observed effects.

In my opinion, the manuscript ID fermentation-2591074 is very up-to-date and valuable. It presents interesting research on the potential for biomass production and lipid accumulation by Parachlorella kessleri TY. I believe that the research was very well thought out and planned, and the method of presenting the results meets all standards set for scientific work. The manuscript was very well prepared both in terms of introduction, research methodology and presentation of the results obtained. I do not make any comments to the descriptive and graphic part. The literature used is up-to-date and directly related to the research topic. In my opinion, the manuscript can be published in its present form.

I am satisfied with your reply. However, in future work, you should not only measure the consumption of glucose but also monitor the organic carbon released by algae during its growth.
