This manuscript describes how CD80/86 triggers the tilapia NCC functions. The study is interesting and important to ascertain fish NCC functions.

The manuscript really needs to be improved in the writing and edition. A native speaker or revisor is encouraged.

 

Comments:

-          In section 2.4. it is not explained why you have to stimulate with 5C6 before the addition of the rOn-CD80/86. No control or nonstimulated leucocytes are included as proper controls.

-          Section 2.5. The NCC killing assay was performed with 1 NCC:1 CIK cells. This is very low and uncommon for fish NCCs. Additionally, the method used to determine the NCC killing activity is “rare” and uncited. Please, explain them.

-          Line 196-198. “….Nile tilapia head kidney lymphocytes (HKLs) can be divided into four subgroups, including nonspecific NCC cells, T cells and macrophage/monocyte (Mo/Mø)”. You indicate 4 populations and then describe 3, and macrophage/monocyte are not lymphocytes. Please, revise this.

-          The number of fish used is not indicated. Are NCCs from individual fish or pooled?

This paper is interesting and addresses a new important topic, however, minor revision is required:

In the title: Please add a dash in-between non and specific (Non-specific) instead of Nonspecific

Please be consistent in using either Nile tilapia or Oreochromis niloticus, I observed that you used mostly Nile tilapia so please replace Oreochromis niloticus in lines 24, 29, 376 by Nile tilapia.

Line 67: Please change the words Streptococcus agalactiae and Aeromonas hydrophila to italic.

Line 73: Please change the word Escherichia coli to italic.

The manuscript with ID (fishes-2035098) by Huang and coauthors has evaluated the costimulatory Molecules CD80/86 that trigger the non-specific cytotoxic cell of Nile tilapia to kill CIK Cells. The manuscript is interesting; however, there are several major issues to be revised by the authors prior the manuscript considered for publication in Fishes.

Ø  The manuscript in general needs to be sent to a native English Speaker. MDPI has an English Editing and proof-reading system. This is because there are several grammar mistakes throughout the whole manuscript.

Ø  All my revisions have been included in the text of the PDF file with Track changes and highlights. Authors have to revise them very carefully and respond to all of them in a detailed response letter.
