Mechanistic and Small Molecule Discoveries for Therapeutic Advances in Hypertrophic and Dilated Cardiomyopathies

Special Issue Information

Dear Colleagues,

The heart is a remarkable organ that, in response to different stimuli, undergoes compensatory changes to manage constant cardiac output. However, these changes can become pathogenic in response to not only genetic mutations and electrical dysfunction but also external factors (e.g., hemodynamic stress) that manifest into a maladaptive phenotype called cardiomyopathy (CM). There are several forms of genetic CM, which often ends in heart failure despite current drug therapies. Here, we focus on hypertrophic (1/500) and dilated (1/250) CM (HCM and DCM, respectively), which are caused by mutations and/or deletions in sarcomeric proteins.

For this Special Issue, we invite papers focused on the following areas:

  • Mechanistic characterization of mutations in sarcomeric proteins that cause HCM and/or DCM;
  • Therapeutic advances (small molecule discovery, development, and testing) for HCM and/or DCM;
  • The mechanism of action of small molecules on HCM and/or DCM.

Suggested methodologies include, but are not limited to, biochemistry, spectroscopy, kinetics, structural, molecular dynamics, physiological, mechanical, high-throughput screening, hit-to-lead optimization, SAR analysis, ADMET, animal models, and translational studies of HCM and/or DCM.

We encourage and welcome submissions of original research and review articles that are focused on or related to elucidating the molecular mechanisms of HCM and DCM and discovering, developing, and characterizing potential small molecules for therapeutic management of this disease.
