Gamma-aminobutyric acid (GABA) is a non-protein amino acid playing a significant role in the central nervous system and the gut–brain axis. This study investigated the potential to produce GABA by lactic acid bacteria (LAB) isolated from different varieties of organic tomatoes. The isolated LAB were taxonomically identified by 16S rRNA gene sequencing, the presence of the gadB gene (glutamate decarboxylase) was detected, and GABA production was quantified using HPLC. Levilactobacillus brevis CRAI showed the highest GABA production under optimised fermentation conditions with 4% monosodium glutamate (MSG). The genome sequencing of L. brevis CRAI revealed the presence of gadA and gadB isoforms and assessed the strain’s safety profile. The gene expression analysis revealed that the gadA and gadB genes were upregulated in the presence of 4% MSG. The probiotic potential of L. brevis CRAI was also assessed by functional assays. The strain showed strong antimicrobial activity against representative enteropathogens, i.e., Escherichia coli ETEC, Salmonella choleraesuis, and Yersinia enterocolitica, and anti-inflammatory effect, reducing nitric oxide production in LPS-stimulated RAW264.7 macrophages. In addition, its ability to adhere to intestinal epithelial Caco-2 cells was demonstrated. These results highlight L. brevis CRAI as a promising candidate for the development of GABA-enriched functional foods or probiotic supplements with the perspective to modulate the gut-brain axis. Background: The early detection of type 1 diabetes (T1D) through screening for major islet autoantibodies is receiving increasing attention as a public health strategy, exemplified by the recent implementation of a pilot pediatric screening program in Italy. The transition from research-based screening to large-scale population initiatives needs automated and standardized assays that are capable of processing extensive sample volumes. Hence, this study aimed to evaluate the analytical performance and comparability of a fully automated chemiluminescence immunoassay (CLIA) compared to a conventional enzyme-linked immunosorbent assay (ELISA) for the detection of three classes of major islet antibodies—anti-GAD (GADA), anti-IA-2 (IA-2A), and anti-ZnT8 (ZnT8A). Methods: A total of 104 serum specimens were analyzed for each autoantibody using both ELISA (RSR and Medyzim, DYNES, DSX) and CLIA (MAGLUMI 800). Assay precision and linearity were assessed through intra-assay variability studies and dilution protocols. Methods agreement was evaluated with Passing–Bablok regression, Spearman’s correlation, Bland–Altman analysis, and Cohen’s kappa statistics. Results: The CLIA showed good precision and excellent linearity across clinically relevant concentration ranges of all islet antibodies. Correlation coefficients and categorical agreement between CLIA and ELISA were high (r > 0.96 and Cohen’s kappa >0.8 for all), with ZnT8A exhibiting the highest concordance. However, proportional biases were found, as CLIA systematically underestimated GADA and ZnT8A levels, while overestimated IA-2A compared to the ELISA. Conclusions: The CLIA displayed satisfactory precision and agreement with ELISA for GADA, IA-2A, and ZnT8A detection. Our findings support the use of these automated immunoassays in large-scale population initiatives for diagnosing T1D, but we also highlight the need for further efforts to achieve better inter-assay harmonization. Herein, a sensitive electrochemiluminescence (ECL) immunosensor is designed by immobilizing ruthenium-tagged immune complexes at flexible poly-ethylene-glycol (PEG) chains on the electrode surface, which offers more freedom for the collision of the ruthenium complex at the electrode during the initial ECL reaction. The electrochemical characterizations confirm the loose structure of the assembled layer with the immune complex, providing an increase in the current and the resultant enhanced ECL emissions. Comparing the sensors with the rigid structure, a 34-fold increase in the maximal ECL emission is recorded when PEG3400 is used as a linker. Using the optimized protocol, the prepared immunosensor exhibits a wide-ranging linear response to the model antibody (glutamate decarboxylase antibody) ranging from 10 pg/mL to 10 ng/mL. The detection limit is almost two orders lower than the value using the classic enzyme-linked immunosorbent assay, which offers a new design to enhance ECL emissions and the resultant analytical performance. The root bark extract of the Maerua oblongifolia plant in the green synthesis of titanium dioxide nanoparticles (TiO2 NPs) for photocatalytic degradation of toxic pollutants and antibacterial activities was implemented in this study. The root bark extract served as a novel capping and reducing agent for the first time. Characterization of the TiO2 NPs was conducted by using visual observation, ultraviolet visible spectrometry (UV-Vis), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM) techniques, confirming their successful synthesis. The TiO2 NPs exhibited maximum absorbance at 323 nm and an average particle size of 19.58 nm; the conjugations and existences of Ti-O and OH vibrational bands were revealed by the FTIR spectrum. The photocatalytic activities of the TiO2 NPs were investigated by using solar irradiation as an energy source for aqueous solutions of methyl orange (MO) and methylene blue (MB) dyes. The TiO2 NPs showed strong photocatalytic activities by degrading 97.23% MB and 91.8% MO under optimized conditions. Degradation behavior was investigated by isotherms and kinetics models, with the Langmuir isotherms (R2: 0.996, 0.979) and Langmuir–Hinshelwood (R2: 0.998, 0.997) highest correlation coefficients for MB and MO, respectively. Moreover, the antibacterial efficacy of the green-synthesized TiO2 NPs and the results indicated higher antibacterial activities on Gram-negative bacteria (27 ± 0.52). Full article

Latent autoimmune diabetes of adults (LADA) is the most prevalent form of autoimmune diabetes (AI-D) in adulthood; however, its accurate diagnosis and optimal treatment remain challenging. Vitamin D deficiency (VDD) is commonly observed in LADA patients, while increased vitamin D exposure through supplementation and dietary intake is associated with a reduced incidence of LADA. Although limited, case reports, case-control studies, and randomized clinical trials have examined the effects of vitamin D supplementation—alone or combined with dipeptidyl peptidase-4 inhibitors (DPP4-is)—on glucose regulation, residual β-cell function, and glutamic acid decarboxylase antibody (GADA65) levels. Findings, while preliminary, indicate that vitamin D supplementation may enhance glycemic control, preserve β-cell function, and reduce autoimmune activity. Given its accessibility, affordability, and relative safety, vitamin D supplementation presents an attractive adjunct treatment option for LADA patients. This narrative review discusses current evidence on the potential therapeutic benefits of vitamin D supplementation in patients with AI-D, including LADA, who are also vitamin D deficient. Beginning with an exploration of the epidemiological patterns, clinical presentation, and diagnostic framework essential for understanding and identifying LADA, this review then examines the proposed mechanisms through which vitamin D may influence autoimmune modulation of pancreatic β-cells, integrating recent data pertinent to LADA pathology. By distilling and consolidating existing research, we aim to provide a platform for advancing targeted investigations within this distinct patient population. Full article

Latent autoimmune diabetes in adults (LADA) is characterized by the presence of glutamate decarboxylase autoantibodies (GADA). LADA has intermediate features between type 1 diabetes and type 2 diabetes. In addition, genetic risk factors for both types of diabetes are present in LADA. Nonetheless, evidence about the genetics of LADA in non-European populations is scarce. This study aims to perform a genome-wide association study with a phenome-wide association study of LADA in a southeastern Mexican population. We included 59 patients diagnosed with LADA from a previous study and 3121 individuals without diabetes from the MxGDAR/ENCODAT database. We utilized the GENESIS package in R to perform the genome-wide association study (GWAS) of LADA and PLINK for the phenome-wide association study (PheWAS) of LADA features. Nine polymorphisms reach the nominal association level (1 × 10−5) in the GWAS. The PheWAS showed that rs7305229 is genome-wide and associated with serum GADA levels in our sample (p = 1.84 × 10−8). rs7305229 is located downstream of the FAIM2 gene; previous reports associate FAIM2 variants with childhood obesity, body mass index, body adiposity measures, lymphocyte CD8+ activity, and anti-thyroid peroxidase antibodies. Our findings reveal that rs7305229 affects the GADA levels in patients with LADA from southeastern Mexico. More studies are needed to determine if this risk genotype exists in other populations with LADA. Full article

Takotsubo syndrome (otherwise known as broken-heart syndrome or left ventricular apical ballooning) is a rare cause of reversible heart failure that predominantly affects postmenopausal women. It was first described by Japanese researchers in the 1990s and has become established as a differential for heart failure following a physically or psychologically stressful event. This was popularised by a spike in cases following natural disasters in Japan. As the recognition of takotsubo syndrome in the differential diagnosis for sudden, onset heart failure in a previously healthy individual has grown, so has the discussion concerning takotsubo in the peripartum period. Peripartum cardiomyopathy is a rare cause of reversible heart failure in the latter weeks of pregnancy and the postpartum period. Morbidity and mortality for both cardiomyopathies can be highly variable, ranging from complete recovery of cardiac function to life threatening arrhythmias and even death. This rapid review highlights the similarities between both cardiomyopathies and challenges the hitherto assumption that both takotsubo and peripartum cardiomyopathies are distinct entities that can easily be distinguished from one another. The implications of this are significant within the context of the behavioural aspects of diagnosis, treatment, and outcome. Full article

The 3 Screen ICA ELISA is a novel assay capable of simultaneously measuring autoantibodies to glutamic acid decarboxylase (GADA), insulinoma-associated antigen-2 (IA-2A), and zinc transporter 8 (ZnT8A), making it a valuable tool for screening type 1 diabetes. Despite its advantages, it cannot specify which individual autoantibodies are positive or negative. This study aimed to estimate individual positive autoantibodies based on the 3 Screen ICA titer. Six hundred seventeen patients with type 1 diabetes, simultaneously measured for 3 Screen ICA and three individual autoantibodies, were divided into five groups based on their 3 Screen ICA titer. The sensitivities and contribution rates of the individual autoantibodies were then examined. The study had a cross-sectional design. Sixty-nine percent (424 of 617) of patients with type 1 diabetes had 3 Screen ICA titers exceeding the 99th percentile cut-off level (20 index). The prevalence of GADA ranged from 80% to 100% in patients with a 3 Screen ICA over 30 index and 97% of patients with a 3 Screen ICA ≥300 index. Furthermore, the prevalence of all individual autoantibodies being positive was 0% for ≤80 index and as high as 92% for ≥300 index. Significant associations were observed in specific titer groups: the 20–29.9 index group when all the individual autoantibodies were negative, the 30–79.9 index group when positive for GADA alone or IA-2A alone, the 30–299.9 index group when positive for ZnT8A alone, the 80–299.9 index group when positive for both IA-2A and ZnT8A, the 300–499.9 index group when positive for both GADA and ZnT8A, and the ≥300 index group when positive for all individual autoantibodies. These results suggest that the 3 Screen ICA titer may be helpful in estimating individual positive autoantibodies. Full article

The expansion of forested areas through afforestation and reforestation is widely recognized as a highly effective natural solution for mitigating climate change. Accurately assessing the potential carbon uptake capacity of newly afforested areas requires modelling tools to estimate biomass stocks, including site index curves and biomass models. Given the unique conditions in terms of tree size, uniform spacing, and tree allometries observed in young afforestations compared to natural stands, specific tools are necessary. In Spain, over 800,000 ha has been afforested with native forest species since 1992, but specific modelling tools for these plantations are lacking. Using data from 370 stem analyses collected across an extensive network of plots in young afforestations, we developed dynamic height growth and site index models for the main native species (five pines and five oaks) commonly used in afforestation in Spain. We compared various nonlinear models, such as ADA (algebraic difference approach) and GADA (generalized algebraic difference approach) expansions. The developed site index models were then used to predict the total biomass stored in the afforestation. Our results underscore the necessity for specific site index models tailored to afforestations, as well as the potential of the established site index in predicting biomass and carbon fixation capacity in these young forests. To search for a novel thermostable esterase for optimized industrial applications, esterase from a thermophilic eubacterium species, Thermoanaerobacter tengcongensis MB4, was purified and characterized in this work. Sequence analysis of T. tengcongensis esterase with other homologous esterases of the same family revealed an apparent tail at the C-terminal that is not conserved across the esterase family. Hence, it was hypothesized that the tail is unlikely to have an essential structural or catalytic role. However, there is no documented report of any role for this tail region. We probed the role of the C-terminal domain on the catalytic activity and substrate preference of T. tengcongensis esterase EstA3 with a view to see how it could be engineered for enhanced properties. To achieve this, we cloned, expressed, and purified the wild-type and the truncated versions of the enzyme. In addition, a naturally occurring member of the family (from Brevibacillus brevis) that lacks the C-terminal tail was also made. In vitro characterization of the purified enzymes showed that the C-terminal domain contributes significantly to the catalytic activity and distinct substrate preference of T. tengcongensis esterase EstA3. All three recombinant enzymes showed the highest preference for paranitrophenyl butyrate (pNPC4), which suggests they are true esterases, not lipases. Kinetic data revealed that truncation had a slight effect on the substrate-binding affinity. Thus, the drop in preference towards long-chain substrates might not be a result of substrate binding affinity alone. The findings from this work could form the basis for future protein engineering allowing the modification of esterase catalytic properties through domain swapping or by attaching a modular protein domain. Stand dominant height has a close relationship with stand productivity and is not much affected by stand density and thinning within a reasonable density range, making it an excellent indicator for estimating stand site quality. Topographic factors (altitude, aspect, slope, etc.) have a significant influence on the growth process of stand level, and the combination of different site factors increases the randomness of the evaluation of forest productivity. In this paper, with one-way ANOVA, it was determined that the effects of density and management mode on the Masson pine stand dominant height were not significant. The data on the Masson pine stand dominant height in the southern subtropics in Guangxi, China, were analyzed, and the GADA model was established using the nonlinear least squares method, the Bayesian approach, and the one-level nonlinear mixed-effects model with the topographic factor as the random effect, respectively. The results indicated that the nonlinear mixed-effects model had the best fitting performance and the highest prediction accuracy for stand site quality (a 0.27% improvement in R² compared to the least squares method and a 1.30% improvement in R² compared to the Bayesian approach), while the model obtained by the Bayesian approach had more elasticity and biological significance. In summary, when the data distribution is uniform and comprehensive, introducing terrain factors into the establishment of site index models can provide a more scientific basis for estimating the productivity of southern subtropical Masson pine stands under different site conditions. When the data distribution is uneven, applying the Bayesian approach can make the site index model more biologically meaningful. The stand site quality model can predict the potential production capacity of forests, which is an important basis and can support forest management and harvest prediction. The results of this study provide a theoretical and practical basis for the establishment of a reasonable site index model for the Masson pine stand. Full article

One of the most important areas of modern genome research is the search for meaningful relationships between genetic variants and phenotypes. In the livestock field, there has been research demonstrating the influence of copy number variants (CNVs) on phenotypic variation. Despite the wide range in the number and size of detected CNVs, a significant proportion differ between breeds and their functional effects are underestimated in the pig industry. In this work, we focused on the problem of leg defects in pigs (lumps/growths in the area of the hock joint on the hind legs) and focused on searching for molecular genetic predictors associated with this trait for the selection of breeding stock. The study was conducted on Large White pigs using three CNV calling tools (PennCNV, QuantiSNP and R-GADA) and the CNVRanger association analysis tool (CNV-GWAS). As a result, the analysis identified three candidate CNVRs associated with the formation of limb defects. Subsequent functional analysis suggested that all identified CNVs may act as potential predictors of the hock joint phenotype of pigs. It should be noted that the results obtained indicate that all significant regions are localized in genes (CTH, SRSF11, MAN1A1 and LPIN1) responsible for the metabolism of amino acids, fatty acids, glycerolipids and glycerophospholipids, thereby related to the immune response, liver functions, content intramuscular fat and animal fatness. These results are consistent with previously published studies, according to which a predisposition to the formation of leg defects can be realized through genetic variants associated with the functions of the liver, kidneys and hematological characteristics. Relationship between β-Cell Autoantibodies and Their Combination with Anthropometric and Metabolic Components and Microvascular Complications in Latent Autoimmune Diabetes in Adults

Aims: Our study aimed to investigate the relationship between three autoantibodies and their combination with anthropometric and metabolic components and microvascular complications in patients with latent autoimmune diabetes in adults (LADA). Methods: Our study included 189 LADA patients divided into four subgroups according to the autoantibodies present: glutamic acid decarboxylase autoantibodies (GADA) only; zinc transporter-8 autoantibodies (ZnT8A)+GADA; insulinoma-associated-2 autoantibodies (IA-2)+GADA; and ZnT8+IA-2+GADA. Results: Compared to GADA positivity only, patients with ZnT8+GADA positivity and ZnT8+IA-2+GADA positivity had a shorter diabetes duration and lower body mass index (BMI); patients with ZnT8+GADA positivity were younger and showed an increase in glomerular filtration rate, while those with ZnT8+IA-2+GADA positivity had lower C-peptide and lower insulin resistance measured with HOMA2-IR. In a multiple regression analysis, ZnT8 positivity was associated with lower BMI (p = 0.0024), female sex (p = 0.0005), and shorter duration of disease (p = 0.0034), while IA-2 positivity was associated with lower C-peptide levels (p = 0.0034) and shorter diabetes duration (p = 0.02). No association between antibody positivity and microvascular complications of diabetes, including retinopathy, neuropathy, and microalbuminuria, as well as with variables of glucose control and β-cell function were found. Conclusion: The results of our study suggest that ZnT8 and IA-2 autoantibodies are present in a significant number of LADA patients and associated with clinical and metabolic characteristics resembling classic type 1 diabetes. Due to increased LADA prevalence, earlier identification of patients requiring frequent monitoring with the earlier intensification of insulin therapy might be of special clinical interest. The Single-Stage Grid-Connected Solar Photovoltaic (SSGC-SPV) topology has recently gained significant attention, as it offers promising advantages in terms of reducing overall losses and installation costs. We provide a comprehensive overview of the system components, which include the photovoltaic generator, the inverter, the Incremental Conductance Maximum Power Point Tracking (IC-MPPT) algorithm, and the PI regulator for DC bus voltage control. Moreover, this study presents detailed system configurations and control schemes for two types of inverters: 2L3PVSI and 3L3PNPC. In order to perform a comparative study between the two structures, we subjected them to the same irradiation profile using the same grid configuration. The Photovoltaic Array (PVA) irradiance is increased instantaneously, in 0.2 s, from 400 W/m2 to 800 W/m2, is kept at 800 W/m2 for 0.2 s, is then gradually decreased from 800 W/m2 to 200 W/m2 in 0.2 s, is then kept at 200 W/m2 for 0.2 s, and is then finally increased to 1000 W/m2 for 0.2 s. We explain the operational principles of these inverters and describe the various switching states involved in generating output voltages. To achieve effective control, we adopt the Finite Set–Model Predictive Control (FS-MPC) algorithm, due to the benefits of excellent dynamic responsiveness and precise current tracking abilities. This algorithm aims to minimise the cost function, while taking into account the dynamic behaviour of both the PV system and the inverter, including any associated delays. To evaluate the performance of the FS-MPC controller, we compare its application in the three-level inverter configuration with the two-level inverter setup. The DC bus voltage is maintained at 615 V using the PI controller. The objective is to achieve a Total Harmonic Distortion (THD) below 5%, with reference to the IEEE standards. The 2L3PVSI inverter is above the threshold at an irradiance of 200 W/m2. The 3L3PNPC inverter offers a great THD percentage, meaning improved quality of the power returned to the grid. Full article

We conducted a study on the dominant height growth of clonal teak (Tectona grandis Linn F.) plantations in the Brazilian Amazon to assess their potential and its agreement with volumetric production. We employed two approaches, ADA (algebraic difference) and GADA (generalized algebraic difference), and analyzed data from 58 permanent plots collected over a 10 year period. To classify the sites, we developed equations and evaluated their accuracy using various criteria, including correlation coefficient, mean square of residual, Akaike’s criterion, distribution of residuals, and validation through equivalence testing (TOST). We also assessed the biological realism of the constructed curves. We used cluster evaluation and dendrogram comparison to assess the agreement between site index and volumetric production for each approach. The Lundqvist–Korf baseline models (M1–ADA and M4–GADA) proved to be accurate and realistic in estimating dominant height in both approaches. Our findings indicate that the approaches utilizing dynamic equations and generating polymorphic curves effectively represent the sites and indicate the volumetric production of the plantations, with 98.3% of agreement rate. Based on our results, we recommend the use of ADA and GADA approaches for estimating the dominant height of clonal teak plantations in the Eastern Brazilian Amazon. Full article
