Background: Accidents caused by the Loxosceles laeta spider constitute a health problem in South America. Envenomation can lead to severe systemic manifestations, eventually compromising the patient’s life. Most regional health authorities consider antivenom administration the basis of effective treatment in the most serious cases. The availability of spider venom is the primary bottleneck for antivenom production. Herein, we present a novel biotechnological approach, based on the expression of recombinant versions of the most relevant toxin in loxoscelism, sphingomyelinase D (SphD), in insect larvae (Spodoptera frugiperda). Methods: We produced two versions of SphD: one conserving its biological activities (wtSphD) and a second alternative that was designed to be genetically detoxified (dSphD). Two horses were subjected to three consecutive hyperimmunization cycles with dSphD. The horses’ plasma was extracted at the end of each cycle and used to produce Active Pharmaceutical Ingredients (APIs) of antivenoms at a pilot scale. Results: Dermonecrotic activity of wtSphD was completely neutralized with the sera obtained from one horse and partially with that of the other. In contrast, the APIs derived in both cases completely neutralized wtSphD dermonecrotic activity. Direct hemolysis of human red blood cells by wtSphD was also neutralized by sera and APIs. Conclusions: These results show venom replacement or complementation potential by recombinant dSphD produced in this novel platform. Full article

Yellow fever remains a major public health threat in endemic and re-emerging regions of Africa and South America, with recent outbreaks highlighting persistent gaps in prevention and surveillance. Pregnant women represent a particularly vulnerable population, yet the epidemiology, clinical impact, and preventive strategies for yellow fever in pregnancy are insufficiently characterized. Physiological and immunological changes during gestation may influence host responses to infection; however, current evidence does not demonstrate increased susceptibility to or severity of yellow fever during pregnancy. Adverse materno-fetal outcomes, including miscarriage, stillbirth, preterm birth, and, in rare cases, perinatal transmission, have been reported but remain poorly characterized. Diagnostic challenges, overlapping clinical presentations with other arboviral and hepatic diseases, and limited access to specialized care further complicate clinical management in many endemic settings. This perspective provides a comprehensive overview of yellow fever in pregnancy during the 2024–2026 outbreak in the Americas, including a risk-stratification framework for prevention. We summarize current evidence on epidemiology, pathophysiology, diagnosis, and supportive care, and examine prevention strategies with particular emphasis on vaccination. Accumulated observational evidence and substantial real-world experience have not demonstrated an increased risk of serious adverse events and generally support the effectiveness of yellow fever vaccination during pregnancy when administered with appropriate clinical judgment. In high-risk settings, the benefits of maternal immunization clearly outweigh theoretical concerns, supporting a flexible, risk-based approach, despite relatively limited evidence. We also discuss national and international policies, post-pregnancy booster recommendations, and the importance of integrating vaccination assessment into antenatal care. Finally, we highlight critical knowledge gaps and research priorities, including the need for prospective registries and strengthened pharmacovigilance. Coordinated clinical and public health strategies are essential to protect maternal and neonatal health and to reduce the burden of yellow fever in endemic and re-emerging settings. Full article

This work presents an alternating sequential model predictive control (ASMPC) scheme applied to multimodular matrix converters. The proposed strategy alternately evaluates two control objectives: load current tracking and input reactive power minimization. The algorithm was implemented in MATLAB/Simulink on an architecture composed of two direct matrix converters in a multimodular configuration. The influence of parameter N2 on system performance was analyzed under step changes in reference current of 30 A and 60 A. To this end, performance metrics such as THD and MSE were used, along with a descriptive statistical analysis including the mean, standard deviation, mean absolute deviation (MAD), and coefficient of variation (CV). Simulation results show stable performance for variations in N2, with an input current THD of 8.10% and load THD reduced to 1.00%, demonstrating improved harmonic performance compared with classical weighted MPC approaches. Although diazotrophy is compatible with low-carbon agriculture and aligned with sustainability goals, its benefits could be expanded by better leveraging associative plant–diazotroph partnerships. Host control, however, remains underemphasized despite increasing resolution of microbial determinants of colonization. Understanding how plants tune permissiveness across fluctuating mineral N landscapes is therefore central to explaining when microbial presence translates into measurable diazotrophic function and plant N gain. Here, we propose an N-mediated host gatekeeping framework that organizes existing evidence into four licensing windows: (i) spatial positioning of permissive sites, (ii) N-sensitive transcriptional thresholding, (iii) local immune tuning at the contact interface, and (iv) carbon energy arbitration sustaining fixation and N transfer. Our model predicts that moderate, spatially heterogeneous mineral N biases the root interface toward permissive states in which microdomain colonization can translate into measurable biological nitrogen fixation, whereas at either extreme one or more windows tend to close. In crops, soil heterogeneity and genotype-linked root functional traits act as filters shaping when functional engagement becomes possible. By reframing N as both a resource and a signal acting through host arbitration, this model clarifies how permissiveness can be tuned to better realize diazotrophic potential and support plant N gain under rational mineral N management. Non-carious cervical lesions (NCCLs) present restorative challenges due to substrate sclerosis and marginal stress concentration, making the adhesive strategy a key determinant of long-term performance. This systematic review and frequentist random-effects network meta-analysis, conducted in accordance with PRISMA 2020 and the PRISMA-NMA extension and prospectively registered in PROSPERO, compared restorative strategies defined by etching approach, adhesive category, and restorative material, with marginal adaptation and retention loss as the primary outcomes. PubMed, Web of Science, Cochrane Library, Embase, and Scopus were searched without restrictions (25 January 2026), supplemented by alternative retrieval methods. Randomized clinical trials in permanent teeth evaluating at least two etching-based strategies (etch-and-rinse, self-etch, selective-etch, and/or resin-modified glass ionomer cement (RMGI)) were included. Risk of bias was assessed using RoB 2 and certainty of evidence with CINeMA. Seventy-four trials were eligible. Connected networks were established for marginal adaptation (57 studies; 28 interventions; 6798 patients; 1772 events) and retention loss (61 studies; 33 interventions; 7338 patients; 584 events). Selective-etch with a universal adhesive and nanocomposite reduced marginal adaptation failure compared with RMGI, whereas certain self-etch and non-universal combinations increased risk. For retention loss, selective-etch and etch-and-rinse protocols combined with universal adhesives and nanocomposites showed lower failure rates, while some self-etch or non-universal adhesive strategies performed less favorably. Overall confidence was predominantly high, with downgrading mainly due to imprecision and heterogeneity. Strategies incorporating selective enamel etching or etch-and-rinse approaches combined with universal adhesives and nanocomposites demonstrated the most consistent clinical advantages. Full article

Drug-resistant tuberculosis (DR-TB) represents a major public health threat, particularly in the prisons of Latin America and the Caribbean, where rates are up to 40 times higher than those observed in the general population. These facilities act as community amplifiers due to overcrowding, poor ventilation, diagnostic delays, and treatment discontinuity. This study offers a critical reflection on the magnitude, determinants, and implications of DR-TB in regional penitentiary contexts. A reflective analytical review was conducted in PubMed, Scopus, Web of Science, SciELO, and LILACS, complemented by WHO and PAHO reports, prioritising studies from 2019 to 2024. The findings reveal MDR-TB and pre-extensively drug-resistant (pre-XDR) outbreaks in Peru, Paraguay, and the Dominican Republic, as well as community transmission linked to prisons in Brazil and Colombia. Persistent gaps remain in systematic screening, drug susceptibility testing coverage, and post-release follow-up. Scientific production continues to be uneven and predominantly biomedical, with limited consideration of social and human rights determinants. DR-TB in prisons reflects the structural deficiencies of health and justice systems; its control requires intersectoral policies, genomic surveillance, and strategies that ensure early diagnosis, treatment continuity, and dignified detention conditions. The 2x2 Demonstrator, a prototype for the Deep Underground Neutrino Experiment (DUNE) liquid argon (LAr) Near Detector, was exposed to the Neutrinos from the Main Injector (NuMI) neutrino beam at Fermi National Accelerator Laboratory (Fermilab). This detector is a prototype of a new modular design for a liquid argon time-projection chamber (LArTPC), comprising a two-by-two array of four modules, each further segmented into two optically isolated LArTPCs. The 2x2 Demonstrator features a number of pioneering technologies, including a low-profile resistive field shell to establish drift fields, native 3D ionization pixelated imaging, and a high-coverage dielectric light readout system. The 2.4-tonne active mass detector is flanked upstream and downstream by supplemental solid-scintillator tracking planes, repurposed from the MINERvA experiment, which track ionizing particles exiting the argon volume. The antineutrino beam data collected by the detector over a 4.5 day period in 2024 include over 30,000 neutrino interactions in the LAr active volume—the first neutrino interactions reported by a DUNE detector prototype. During its physics-quality run, the 2x2 Demonstrator operated at a nominal drift field of 500 V/cm and maintained good LAr purity, with a stable electron lifetime of approximately 1.25 ms. This paper describes the detector and supporting systems, summarizes the installation and commissioning, and presents the initial validation of collected NuMI beam and off-beam self-triggers. In addition, it highlights observed interactions in the detector volume, including candidate muon antineutrino events. Full article

Urban and peri-urban river systems subjected to intensive agriculture are vulnerable to diffuse metal(loid) inputs, yet the integration of hydrological compartments, bioindicators, and human health risk remains poorly explored. This study investigated the seasonal dynamics, bioaccumulation patterns, and potential human health risks associated with metal(loid)s in the Santa Virgem River (Brazil–Paraguay border), using water from backwater zones and three plant groups (Apiaceae angiosperms, mosses, and the liverwort Dumortiera sp.). Water and plant samples were collected during five seasonal campaigns (2019–2020) and analyzed by ICP OES. Multivariate analysis (PCA) was applied, and biological accumulation coefficients (BAC) and chronic daily intake (CDI) were estimated for adults and children under different ingestion scenarios. Results showed that Mg, Fe, K, S, and P dominated water chemistry, while As, Cd, Cr, Cu, Pb, and Se were mostly below detection limits. PCA explained 77.6% of total variance, distinguishing agricultural and hydrological phases. Bryophytes exhibited markedly higher BAC values, particularly for Mn (up to 2.3 × 105) and Fe, compared with Apiaceae. CDI and hazard assessment indicated negligible non-carcinogenic risk for most elements (HQ < 1), except phosphorus, which dominated the Hazard Index due to its low reference dose. Overall, the results demonstrate that hydrodynamic conditions and plant functional traits jointly control metal(loid) dynamics, highlighting the value of multispecies biomonitoring in peri-urban river systems. Rapid agricultural expansion threatens water security in one of the world’s largest hydroelectric systems, the Itaipu dam, located on the Brazil–Paraguay border. Yet regional hydrological responses to land cover change and climate variability remain insufficiently characterized at management-relevant scales. The Upper Paraná River Basin (UPRB), which sustains agriculture, hydropower, and municipal water supply across both countries, exemplifies this challenge as accelerating cropland conversion raises concerns about long-term water availability. This study investigates hydrological transitions and their statistical associations with land cover changes in the Itaipu study region from 2002 to 2023. We integrate GRACE/GRACE-FO (Gravity Recovery and Climate Experiment Follow-On), Terrestrial Water Storage Anomalies (TWSAs), MODIS (Moderate Resolution Imaging Spectroradiometer) land cover, CHIRPS (Climate Hazards Group InfraRed Precipitation with Station data) precipitation, and LandScan population density using Pettitt’s breakpoint test and Mann–Kendall trend analysis to detect temporal breakpoints and quantify co-variability between hydrology and land surface dynamics. Together, these methods identify a significant basin-wide shift in TWSAs in mid-2009, with storage increases of 151.6 cm at Itaipu and 103.1 cm at Yguazú Reservoir. Over the study period, cropland expanded from 13.5% to 37.9% of total land cover, while savanna declined from 28.1% to 24.2%. After 2009, correlations between land cover and TWSAs strengthened substantially, particularly for wetlands (r = 0.88), croplands (r = 0.73), and savannas (r = −0.81; all p < 0.001), indicating strong coupling between landscape transformation and basin-scale storage variability. Principal Component Analysis shows land use change explains 39–41% of TWSA variance, exceeding hydroclimatic contributions. Granger causality analysis reveals bidirectional coupling between wetlands and water storage at Itaipu, while cropland and savanna dynamics exert predictive influence on downstream hydrology in the Yguazú basin. Water balance decomposition further indicates a post-2009 regime shift, with residual storage transitioning from −10.6 to +4.7 and 78% greater runoff generation per unit precipitation, consistent with reduced infiltration capacity. Together, these findings underscore intensifying land–water feedback and the need for adaptive watershed management under expanding agriculture and climate variability. Full article

We consider two steady-state heat conduction systems called, S and Sα, in a multidimensional bounded domain D for the Poisson equation with source energy g. In one system, we impose mixed boundary conditions (temperature b on the boundary Γ1, heat flux q on Γ2 and an adiabatic condition on Γ3). In the other system, the condition on Γ1 is replaced by a convective heat flux condition with coefficient α. For each of these systems, we consider three associated optimization problems (Pi) and (Piα), i=1,2,3, where the variable is the source energy g, the heat flux q and the environmental temperature b, respectively. In the particular case where D is a rectangle, the explicit continuous optimization variables and the corresponding state of the systems are known. In the present work, by using a finite difference scheme, we obtain the discrete systems (Sh) and (Sαh) and discrete optimization problems (Pih) and (Piαh), i=1,2,3, where h is the space step in the discretization. Explicit discrete solutions are found, and convergence and estimation errors results are proved when h goes to zero and when α goes to infinity. Moreover, some numerical simulations are provided in order to test theoretical results. Finally, we note that the use of a three-point finite-difference approximation for the Neumann or Robin boundary condition at the boundary improves the global order of convergence from O(h) to O(h2). Background: Psychiatrists operate at the interface of clinical care, legal frameworks, and governmental power, where external pressures and insufficient safeguards can potentially engender ethical vulnerabilities. Supranational instruments and wider professional standards notwithstanding, the extent to which national-level psychiatric associations articulate protections against torture and abusive practices in their ethical codes remains underexplored. Methods: A cross-sectional documentary audit was conducted of all 145 World Psychiatric Association (WPA) Member Societies, representing ≈250,000 psychiatrists globally. National-level psychiatric ethical codes were located via systematic web searches and examined for clauses specifically referencing torture or analogous abuses and for any associated enforcement procedures. Results: Only nineteen (13.1%) WPA Member Societies maintained publicly accessible ethical codes, with ten (6.9%) containing explicit provisions proscribing torture and associated abuses. These predominantly originated from high-income countries or jurisdictions with documented histories of human rights violations. Most codes invoked broad principles without directly addressing such abuses, and fewer than half delineated any enforcement mechanisms. Conclusions: Gaps persist in ethical governance and human-rights safeguards amongst WPA Member Societies. Although beneficence and non-maleficence provide moral foundations for psychiatric practice, generic commitments alone may prove inadequate under duress. Strengthening anti-torture prohibitions within national-level psychiatric codes could therefore help support ethical resilience and accountability in situations of institutional or political coercion. The Neoproterozoic Paraguay Belt (western Brazil), formed by the Brasiliano/Pan-African Orogenic Cycle during Western Gondwana amalgamation, hosts tens of gold occurrences and deposits. This review provides new insights into the metallogeny and tectonics of the Paraguay Belt Gold Province, based on previous and new data on stratigraphy, structural geology, metamorphism, hydrothermal alteration, and gold grades. The mineralizations correspond to turbidite-hosted orogenic gold systems, with quartz veins cutting metasedimentary rocks with minor metavolcanics. A six-phase tectonic–metallogenic evolution model is proposed. The early stages correspond to glaciolacustrine deposition in a rift that evolved into a passive margin and then into a foreland basin, with glaciomarine sediments. Late sedimentation corresponds to glacial-to-post-glacial shallow marine units in the foreland. The orogeny progressed with cratonic collisions, resulting in three deformation (two compressional and one extensional) and three metamorphic (regional, dynamic, and contact) phases. The Au mineralization results from metamorphic fluids that transported metals from the metasedimentary pile and deposited them in reactive rocks (rich in magnetite or organic carbon). Gold occurs in sulfide-rich (pyrite and galena) veins and hydrothermal alteration zones. The metallotects and structural controls highlighted here are useful tools for prospecting gold in the Paraguay Belt and similar geological terranes. Full article

West Nile virus (WNV) is a mosquito-borne flavivirus with one of the widest global distributions. Since its discovery in Uganda in 1937, it has become a major zoonotic pathogen, and after its introduction into the United States in 1999, it spread rapidly across the Americas, becoming the leading cause of neuroinvasive arboviral disease. Its expansion illustrates a remarkable ecological adaptability, further intensified by climate change. In Latin America and the Caribbean, WNV circulation has been consistently documented in birds, horses, and mosquitoes; however, confirmed human cases remain disproportionately scarce compared with North America and Europe. Reports include sporadic human cases in Brazil (>100 since 2014), Mexico (~13), Argentina (2006–2007), Puerto Rico (2007), Nicaragua, and Haiti, while animal and vector evidence extends to Guatemala, El Salvador, Belize, Costa Rica, Bolivia, Paraguay, Colombia, Venezuela, Cuba, and Ecuador. This paradox likely reflects structural limitations within regional health systems, including underdiagnosis, restricted diagnostic capacity, and significant surveillance gaps, particularly in contexts where mild febrile syndromes may be misclassified as dengue, Zika, or Chikungunya. The regional risk of emergence is further amplified by climatic variability, ecological change, and intensifying human–wildlife interactions. Experiences from Europe highlight the importance of early detection, transfusion safety, and integrated surveillance within a One Health framework. Strengthening preparedness in Latin America will require investments in diagnostic infrastructure, implementation of standardized seroepidemiological surveys, development of predictive models tailored to local ecological contexts, and robust intersectoral collaboration. Background/Objectives: As the prevalence of depression and the use of antidepressants have risen steadily in the last decade, new treatment options are needed. Aloysia gratissima var. gratissima ethanol extract has previously shown antidepressant-like activity, and the present study was conducted to identify the active fraction and clarify the possible mechanisms of action. Methods: Tail suspension (TST) and forced swimming (FST) behavioral tests were performed, and possible mechanisms of action were elucidated using serotonergic, dopaminergic, adrenergic, and GABAergic system antagonists. UPLC-DAD-MS analyses were performed to identify compounds in active fractions, and molecular docking studies were carried out to determine the binding affinities of these compounds to serotonergic and dopaminergic receptors (5-HT1A, 5-HT2A, 5-HT3, and D2R). Results: Ethyl acetate and butanol fractions were found to decrease immobility time in FST. The reduction in immobility time during the FST caused by the ethyl acetate fraction was reversed by pretreating mice with WAY100635 (5-HT1A antagonist), ketanserin (a 5-HT2A antagonist, ondansetron (5-HT3 antagonist), or haloperidol (D2 antagonist). UPLC-DAD-MS analysis revealed a similar composition for the ethyl acetate and butanol fractions of A. gratissima var. gratissima. Pharmacokinetic predictions suggest that only a few of the identified compounds have the potential to permeate the blood–brain barrier, and molecular docking simulations showed that compounds such as 13-oxooctadecadienoic acid, ferulic acid, and coumaric acid have binding affinities to the druggable site of serotonergic and dopaminergic receptors. Conclusions: These results suggest that the Agg ethyl acetate fraction possesses antidepressant-like activities, altering dopaminergic and serotonergic system functions. Computational simulations also suggest that some of the identified compounds have binding affinities to the 5-HT1A, 5-HT2A, 5-HT3, and D2R receptors. Full article
