Captive reptiles bred for economic and medicinal purposes are often susceptible to intestinal parasites that undermine their health and overall farm productivity. In many regions, their economic value largely derives from medicinal use; accordingly, they are described as reptiles with medicinally driven economic value. In this study, we applied high-throughput sequencing to characterize gut eukaryotic communities and assess parasite prevalence in tokay geckos (Gekko gecko) and Chinese blue-tailed skinks (Plestiodon chinensis). Across both host species, six pathogenic genera were identified: Cryptosporidium (P. chinensis: 57.1%, 12/21; G. gecko: 0%), Eimeria (P. chinensis: 14.3%, 3/21; G. gecko: 0%), Oswaldofilaria (P. chinensis: 0%, G. gecko: 4.8%, 1/21), Strongyloides (P. chinensis: 14.3%, 3/21; G. gecko: 19.1%, 4/21), Spironucleus (P. chinensis: 14.3%, 3/21; G. gecko: 9.5%, 2/21), Spauligodon (P. chinensis: 0%, G. gecko: 14.3%, 3/21). Among these, Cryptosporidium exhibited a striking host-specific pattern, occurring exclusively in Chinese blue-tailed skinks (Fisher’s exact test, p = 5.32 × 10−5). Principal component analysis (PCA) demonstrated clear host-associated segregation along PC1, which accounted for 58.2% of total variance. Furthermore, co-occurrence network analysis revealed significant positive associations (all p < 0.01) between parasites and other eukaryotes, particularly fungi and protozoa. Collectively, these findings underscore the value of high-throughput sequencing for parasite surveillance in reptile farming and point to the importance of integrated diagnostic approaches and enhanced biosecurity measures for maintaining animal health. The gut microbiota plays a crucial role in the health and well-being of primates. This study applied 16S rRNA high-throughput sequencing technology, aiming to investigate the differences in gut microbiota composition and function between captive ring-tailed lemurs (Lemur catta) in different zoos across the north–south demarcation line in China. Results revealed significant differences in gut microbiota composition between northern and southern groups. Alpha diversity indices were higher in the southern group (p < 0.05), while beta diversity analysis showed distinct clustering based on geographic location (p < 0.001). Bacteroidetes were more abundant in the northern group (49.22% vs. 28.44%), while Firmicutes predominated in the southern group (59.10% vs. 32.78%). Functional prediction analysis indicated higher levels of membrane transport and lipid metabolism pathways in the southern group, suggesting differences in nutrient absorption and energy metabolism. These findings suggest that geographic location and associated environmental factors significantly influence the gut microbiota of captive ring-tailed lemurs, even under similar dietary and husbandry conditions. Our study provides insights into the impact of geographic location on gut microbiota in captive primates, highlighting the importance of considering regional factors in zoo animal management and informing future strategies for optimizing the care and conservation of captive primates across different geographic regions. Since its discovery in 2019, SARS-CoV-2 has continued to be detected in both humans and animals worldwide. Currently there is limited research focusing on serological surveillance of wildlife under human care. Here we tested 230 serum samples of 134 animals from two zoological institutions collected between 2015 and 2024. To assess prior exposure and antibody responses from natural infection or vaccination, we used three serological assays: a nucleocapsid protein-based ELISA (N-ELISA), a surrogate virus neutralization test (sVNT) for spike (S) protein and a neutralization assay with S-pseudotyped viral particles. Among the 114 samples collected from 58 animals at Fort Wayne Zoo in Indiana, 37 samples from 20 vaccinated animals were sVNT-positive, and 2 of the positive animals had 2 samples prior to vaccination that tested positive by N-ELISA. Of the 116 samples from 76 animals at Brookfield Zoo in Illinois, 20 samples of 20 animals were sVNT-positive, and 19 of the positive animals had been vaccinated. Among these 20 sVNT-positive samples, only one sample from a South American Tapir was positive from prior to vaccination and 1 sample from a sloth bear was also positive by N-ELISA, marking the first documented cases of SARS-CoV-2 exposure in both species. Neutralization assays with S-pseudotyped virus revealed that some of the sVNT-positive samples have strong activity against the WH1-S pseudovirus but showed significantly reduced neutralization against the Omicron LP.8.1-S pseudovirus. These results underscore the need for updated vaccines tailored to emerging variants. Overall, our findings highlight the importance of continued serological surveillance across multiple species to detect new SARS-CoV-2 exposures and monitor vaccine-induced immunity in captive animal populations. The water deer, although an internationally endangered species, is designated as a nuisance wild animal in South Korea and occupies a unique ecological niche. Studying the gut microbiome of this species is crucial for understanding its ecology. We amplified 16S rRNA DNA and compared the gut microbiomes of wild water deer from three regions with those of captive water deer from one region. Our results showed that the gut microbiome diversity of water deer did not differ significantly across regions in the wild but decreased significantly when raised in captivity. The similar microbiomes of water deer living in different regions are believed to be due to dietary diversity rather than dietary homogeneity. Furthermore, the monotony of the food supply appears to lead to significant variation in captive environments. From a conservation biology and biorestoration perspective, we suggest the importance of conserving the gut environments of animals conserved and restored outside their native habitats. Full article

Awareness regarding the welfare of captive baboons is rising. Consequently, the best possible medical care is offered to injured animals. To this purpose, knowledge of the species-specific anatomy is a prerequisite. However, detailed anatomical reference works on this species, such as overviews or atlases, are sparse. The existing anatomical literature is scattered in often outdated works or elaborates on a specific detail. Veterinarians responsible for the medical care of captive baboons, therefore, habitually rely on human anatomical atlases. As overviews of the baboon joint morphology are particularly sparse, this first study in a series of three aims to provide a comprehensive overview of the arthrology of the thoracic limb of the hamadryas baboon (Papio hamadryas). The several synovial joints present in the shoulder region, elbow region, and hand are included. Not only the typical connective tissue elements that form the joints but also the associated muscle tendons are depicted. The osseous structures to which these components attach are identified as well. Standard veterinary terminology is used, complemented by human anatomical nomenclature where the former falls short. High-resolution color photographs support the text, allowing this work to serve not only as a dissection guide for veterinary and academic use but also as a baseline for clinical medical care and future research in primate morphology and biomechanics. Although long neglected, sound is now an increasing topic of interest in zoo and aquarium science. Research has examined the impact of sounds, from various sources, on zoo-housed species, noting that the influence of sound is varied and very context specific. The zoo’s sound environment is influenced by the animals, the built environment, vegetation, climatic, temporal and seasonal factors, equipment use, husbandry practices, and human presence. Different sounds can dominate an enclosure at certain times. This article discusses a workshop involving 12 zoo professionals, held in March 2020, that explored how sound is considered or overlooked in zoo animal management. Although insights are based on a small group, limiting generalisability, the findings highlight areas where further understanding is required and should encourage research extension to other groups of stakeholders. Delegates emphasised that the auditory needs of animals are often underappreciated and that the influence of sound depends on the species involved and how the sound may be perceived. Delegates highlighted the importance of species- and individual-specific approaches, predictability, and how animals have (any) control over sounds experienced in their enclosure. Routine operational sounds, such as closing gates or doors, may inadvertently stress animals, suggesting the need to consider sound in enclosure design and husbandry schedules. Outputs also stated that sound, when carefully managed, can act as enrichment through (for example) structured auditory cues or naturalistic sounds if ecologically relevant. Overall, our findings support integrating sound measurement into broader welfare assessment frameworks and enclosure planning, and they identify practical applications including sound mitigation, enrichment strategies, and staff training. Future research should include a wider range of species (especially understudied taxa), consider the experiences of a wider stakeholder demographic, and evaluate how sound is management in complex or high-traffic areas of the zoo. Baboons’ management requires chemical restraint. Three intramuscular sedative protocols in captive hamadryas baboons (Papio hamadryas) undergoing health-check and male vasectomy were compared. Animals were assigned to TZD_G (n = 17; tiletamine/zolazepam 3 mg/kg + dexmedetomidine 20 μg/kg), KDM_G (n = 23; ketamine 6 mg/kg + dexmedetomidine 30 μg/kg + methadone 0.2 mg/kg), or MDM_G (n = 9; midazolam 2 mg/kg + dexmedetomidine 60 μg/kg + methadone 0.2 mg/kg). Propofol was titrated intravenously for anaesthetic induction and maintenance. Sedation time and quality and cardiopulmonary parameters were recorded. Atipamezole (TZD_G 0.2 mg/kg, KDM_G 0.3 mg/kg, MDM_G 0.6 mg/kg) and flumazenil (MDM_G 0.02 mg/kg) were administered intramuscularly post-procedure. Recovery time and quality were recorded. Data were reported as median (interquartile range) or regression coefficient (B). Sedation was deepest in TZD_G (20, 20–20; KDM_G 20, 19–20; MDM_G 19, 15–20; p = 0.017). MDM_G had a significantly higher heart rate (B = 10.27, p = 0.001), respiratory rate (B = 9.09, p < 0.001), and lower end-tidal carbon dioxide (B = −3.00, p = 0.03) than TZD_G, while KDM_G had a lower respiratory rate than TZD_G (B = −3.67, p = 0.02) and a higher temperature (B = 1.66 p = 0.001). TZD_G showed the longest recovery (minutes: 19, 11.5–30; KDM_G: 6, 4–12; MDM_G: 4, 2.5–5; p < 0.001), while MDM_G the best recovery (0, 0–0; TZD_G: 9, 6–12; KDM_G: 0, 0–6; p < 0.001). TZD produced deepest sedation but bad recovery; KDM offered optimal sedation and recovery, and cardiopulmonary stability; MDM provided lighter sedation and excellent recovery. Full article

Non-human primates (NHPs) are considered important models for the study of reproductive diseases, due to their anatomical and physiological similarities to humans. However, studies on spontaneous lesions of the reproductive system in NHPs housed in zoos remain limited compared to those in laboratory animals. In this study, we report a retrospective analysis of female reproductive pathologies in 103 necropsied non-human primates from Italian zoos between 2007 and 2024. Only adult, intact, non-pregnant females with macroscopically visible reproductive lesions were included. Histopathological examination revealed reproductive tract lesions in 15 individuals (14.6%), including both non-neoplastic (cystic endometrial hyperplasia, adenomyosis, endometriosis) and neoplastic (leiomyomas, cervical and ovarian adenocarcinomas, and metastatic tumors) conditions. Leiomyoma was the most common tumor, particularly in the genus Macaca, while rare malignant neoplasms and metastatic lesions were identified in the great apes and in the New World species. The results suggest an age-related degenerative component and highlight interspecific differences in the distribution of lesions, probably related to the reproductive physiology of the various species. These results underline the importance of systematic post-mortem surveillance to improve the management of reproductive health of these captive populations and provide comparative insights with humans. Assessing the well-being of captive brown bears is essential for responsible species management and meeting European and national animal care standards outside their natural habitats. The present study aims to assess the welfare of captive brown bears in Bulgaria, considering environmental parameters, husbandry procedures, and the psychological and physical condition of the animals. The study was conducted on 28 brown bears in 11 zoos in Bulgaria and 19 bears in Bear Sanctuary Belitsa. A specific questionnaire and interviews were conducted with zoo managers and keepers. The welfare of the bears was evaluated based on their living conditions, husbandry, and physical and psychological well-being. The Nature and Animal Protection Center in Dobrich received the highest rating for the welfare of two brown bears among the zoos in the study. The study identified annual veterinary check-ups and enrichment programmes as areas needing improvement. Full article

Background/Objectives: Toxoplasma gondii is a major cause of zoonotic infections in both humans and animals, resulting in significant mortality in susceptible species, such as New World primates and marsupials. Toxoplasmosis is particularly concerning in zoos and wildlife reserves, where outbreaks threaten conservation efforts for endangered species. In the absence of a commercially available vaccine against toxoplasmosis for humans and captive wild animals, current prevention strategies are limited to restricting the access of cats to enclosures, controlling rodent populations, and maintaining strict food hygiene. Recent research has shown promising results with an intranasal vaccine (VXN-Toxo) composed of maltodextrin nanoparticles conjugated with a purified, inactivated T. gondii parasite. This experimental vaccine does not pose a risk of causing disease and offers advantages such as better stability compared with live pathogen-based vaccines. Methods: This study presents a large-scale evaluation of the effect of VXN-Toxo administered to captive wildlife across 20 zoos in Europe and the Americas between 2017 and 2025. Seven hundred and eighty-four animals, representing over 58 species (including primates, marsupials, rodents, and felids), were vaccinated without any adverse events reported. Results: Retrospective mortality data from 20 participating zoological institutions revealed an overall 96.7% reduction—and, in many cases, a complete elimination—of toxoplasmosis-associated deaths post vaccination. Conclusions: These results demonstrate, for the first time, consistent and broad-spectrum protection against T. gondii of different strains in a wide array of captive wildlife species. This universal vaccine represents a promising tool for toxoplasmosis prevention in zoological collections, with significant implications for animal health and conservation strategies. Full article

Wildlife rescue centres face considerable challenges in promoting animal welfare and enhancing the care and housing conditions of animals under professional supervision. These challenges are further compounded by the diversity of species admitted, each with distinct specific needs. In Colombia and other Latin American countries, primates are among the most frequently rescued and behaviourally complex mammalian taxa, requiring particular attention. In response, this study aimed to assess the content validity of proposed animal welfare indicators for Cebus albifrons through a Delphi consultation process and to develop two species-specific assessment protocols: a daily-use tool for keepers and a comprehensive protocol for professional audits. A panel of 23 experts in primate care and rehabilitation participated in two consultation rounds to evaluate and prioritise the indicators based on their content validity, perceived reliability, and practicality. Indicators were classified as either animal-based (direct measures) or resource- and management-based (indirect measures). After each round, experts received summarised feedback to refine their responses and facilitate consensus building. Of the 39 initially proposed indicators, 28 were validated for inclusion in the extended protocol and 10 selected for the daily-use checklist. Among these, 20 indicators in the extended protocol and 6 in the daily protocol were resource- or management-based—such as adequate food provision, physical enrichment, and habitat dimensions—highlighting their practical applicability and relevance in identifying welfare issues and risk factors. Although these indirect indicators were more numerous, the top-ranked indicators in both protocols were animal-based, including signs of pain, affiliative behaviours, and abnormal repetitive behaviours. These are essential for accurately reflecting the animals’ welfare state and are therefore critical components of welfare assessment in captive non-human primates. This study demonstrates that welfare assessment tools can be effectively tailored to the specific needs of wildlife rescue centres, providing a robust foundation for enhancing welfare practices. These protocols not only offer practical approaches for assessing welfare but also underscore the importance of embedding animal welfare as a priority alongside conservation efforts. Future research should aim to refine these tools further, assess their implementation, and evaluate inter- and intra-observer reliability to ensure consistency across different settings. Full article

Extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae pose a significant public health risk. As zoos grow in popularity, exotic animals come into closer contact with humans, making them potential reservoirs of ESBLs. However, data on ESBL presence in Mexican zoos remains limited. For this reason, this study aimed to isolate and assess the antimicrobial susceptibility of Enterobacteriaceae that colonize avian species in a petting zoo and to identify any ESBL-producing isolates. Cloacal swabs were collected from 34 healthy birds at Miguel Angel de Quevedo Zoo, Veracruz, Mexico. Samples were analyzed microbiologically and molecularly to detect ESBL-encoding genes. A total of seventeen E. coli and one K. pneumoniae strains were isolated from cloacal swabs of bird species, and multidrug resistance (MDR) was found. The most frequently detected genes were blaCTX-M-1 (16/18) and blaTEM-1 (12/18). The detection of multidrug-resistant (MDR) strains carrying blaCTX-M-1, blaTEM-1, and blaSHV genes highlights the potential role of birds as reservoirs and disseminators of antimicrobial resistance (AMR) in urban environments. To the best of our knowledge, this is the first study conducted in Mexico. In conclusion, MDR ESBL-producing bacteria were found in the fecal microbiota of bird species at a petting zoo in Mexico. The limitations of this study emphasize the need for a One Health approach to analyze the genome-wide isolates and epidemiology of antimicrobial-resistant bacteria in captive zoo animals in Mexico. This would support targeted surveillance efforts and help reduce the emergence and spread of resistant bacteria among zoo animals and visitors. Full article

Oxytocin (OT) has emerged as a central focus in stress regulation and animal welfare research. While commonly measured in blood, saliva, and urine, its presence in feces remains unexplored. Fecal OT (fOT) could be highly valuable for wildlife studies, where direct handling is impractical. This study explored OT detection in feces using lions (Panthera leo) as a model species. The objectives included developing a reliable fOT extraction protocol, describing baseline fOT patterns in captive lions, and evaluating its relationship with fecal glucocorticoid metabolites (fGM) under non-stressful conditions. Fecal samples from 16 lions in stable social groups across five Spanish zoos were collected over two to six weeks. A methanol-based extraction and commercial enzyme immunoassay were successfully validated for fOT quantification. Concentrations ranged from 3.00 to 296.64 ng OT/g of feces, with notable intra- and inter-individual variability. Age, sex, and contraceptive status had no significant influence on fOT (p > 0.05), but concentrations differed significantly between zoos (p < 0.001). No consistent relationship was found between fOT and fGM levels under baseline conditions. This study is the first to demonstrate OT detection in feces. The validated methodology provides a foundation for investigating fOT as a biomarker of stress and welfare in wild and domestic species. Full article

The welfare of decapod crustaceans held in captivity, whether in aquariums, holding facilities, seafood restaurants, supermarkets, or fishmongers, is attracting growing attention and concern. This review focuses on three species that are commonly found in European commercial environments: the European spiny lobster (Palinurus elephas), the European lobster (Homarus gammarus), and the American lobster (Homarus americanus). We examine the biology and behavioural ecology of these decapods, emphasising their natural habits and environmental requirements. Based on current scientific knowledge, we propose conditions for their confinement that better align with their biological and behavioural needs. These crustaceans are commonly subjected to stressful captive environments characterised by overcrowding, claw immobilisation through banding, an absence of shelter or hiding spaces, and continuous exposure to direct lighting. Such practices contrast starkly with the animals’ innate behaviours and natural habitats, likely compromising their physical health and psychological welfare. It is thereby recommended that spiny lobsters and lobsters be kept separated and stocked at an appropriate density, that each animal should be provided with a shelter and have enough space to turn around, that true lobsters should never have their claws tied, that no direct light should be placed above the aquaria, and that water quality should be routinely monitored. By exploring the welfare challenges associated with confining these species, this review aims to promote a more ethical and humane approach to their care in commercial settings, and to encourage practices that enhance their quality of life in captivity. Humane euthanasia is an endpoint for production animals succumbing to disease or trauma. Euthanasia performed with barbiturates or other anesthetic/sedative drugs observes zero withdrawal time, and drug residues may remain in tissues. Carcasses may be submitted for rendering, and rendered products can be used to manufacture pet foods. The purpose of this study was to determine the concentration of two drugs, xylazine and ketamine, that may be used during the euthanasia process of food animals and to determine the fate of these drugs following a simulated rendering process using a commercial autoclave. Twelve cattle were administered xylazine or xylazine and ketamine prior to euthanasia via penetrating captive bolt, and samples of muscle, fat, liver, and kidney were collected. The tissue samples were analyzed by LC-MS/MS, both raw and following rendering. The parent compounds xylazine and ketamine were detected in all tissues, both before and after rendering. The highest concentrations were found in rendered kidney for both drugs, and the lowest in rendered and raw fat for xylazine and ketamine, respectively. Full article
