The benefits of Saccharomyces cerevisiae, both native and modified, var. boulardii and other S.cerevisiae in therapeutic uses are explained in the review. This is a nice review and an important subject. I just have a couple of comments.

1 Line 71

The description for innate health benefits is inaccurate in figure 1, for instance, it repeated between “toxin proteolysis” and “inactivation of toxin”, as well as “interference with toxin binding” and “interference with toxicity mechanism”. Please revise this section to reflect properly.

2 Line 87

I'm not sure what the “target disorder” stands for. Perhaps it would be better to say it another way.

3 Line 149

It is advised to combine sections 2.1 and 2.2 into one. The contents of "2.2 innate non-probiotic benefits" also discuss the innate probiotic benefits.

4 Line 317

It is suggested that "biosensing and expression systems" should be used instead of "biosensing and signaling systems."

5 Line 152,388, 394,399,401,430,439,476

It should say "polysaccharides" instead of "oligosaccharides."

Comments and Suggestions for Authors

Well-written manuscript, but could be improved for better impact to get accepted. Section 2 speaks about the health benefits of Sb and Sc. Although the section is divided into innate probiotic and non-probiotic benefits, it needs further details and modifications regarding the level/type of evidence. The evidence needs to be clarified whether they are based on in vitro or in vivo (animal or human) studies. A table with a list of studies describing the study design, methodology, and main outcomes will add additional value to the manuscript.

An additional section related to any safety aspects related to native and engineered Saccharomyces yeasts is needed.
