Round 1

Reviewer 1 Report

Comments and Suggestions for Authors Paeonia ostii ‘Fengdan’ is a traditional medicinal plant and woody oilseed plant with a considerable agronomic potential. The research on BBM gene related to somatic embryogenesis will promote its resources conservation and breeding advancement. The subject of this paper is appropriate for publication in forests. And the work represents a new, original, and interesting contribution. The structure and organization of the paper appear to be sound in general, but some aspects should be clarified and/or more thoroughly explained and justified (see below).  Abstract section Line 30 Since the function of PoBBM was not verified, “PoBBM is a potential key gene” is suggested.  Introduction section Line 36 The English name of Paeonia ostii is different from one of P. suffruticosa.  Materials and methods section Line 185 Sampling period is different the one of Line 350. Meanwhile, whether do the cotyledon growth stage of mature embryo belong to the somatic embryogenesis development process?  Results section

Paragraphs (line 427-452) are little correlation with your content, are suggested to be excised.

Comments and Suggestions for Authors

The paper “Cloning, Characterization, and Expression Pattern Analysis of the BBM Gene in Tree Peony (Paeonia ostii)‘’ aimed to a screen BBM gene, named PoBBM,  clone, and identify from the third-generation full-length transcriptome data of peony.

The paper is prepared professionally. It includes a well-crafted abstract and an exhaustive introduction that justifies the research undertaken. The introduction points to the deficiencies in the literature on the subject. The aim is clearly defined. Modern analytical methods were used in the research. The discussion of the results is well prepared. The conclusions are well-defined. The illustrative material is appropriate.

In my opinion, the manuscript after corrections, will be suitable for publication in a journal.

Detailed comments:

Abstract: Should include some more numeric data obtained from the study

Do not use abbreviations when use first time.

Abstract should be rewritten based on international rules.

Introduction - The introduction is enough in my opinion. Introduction needs some minor changes

Line 44-45 Molecular breeding offers an effective strategy to overcome the 44 limitations of conventional breeding and reduce the breeding timeline.

Please add some references. I suggested below ones

Line 105-107

The experimental material selected for this study was the cultivated variety ‘Feng- 105 dan’, known for its ornamental, medicinal, and edible properties, with stable genetic traits 106 developed by prolonged cultivation and evolution....

Please add more information (pedigree etc.) of plant material

Line 131-132

RNA integrity and quality were assessed by 1% agarose gel electrophoresis....Does 1% agar worked well??????

What is the relationship The peony BBM iand the grape BBM???? Explain more please
