Round 1

Reviewer 1 Report

Kindly find attached the comments of the manuscript entitled "Evaluation of compounds from Balanites aegyptiaca against Squalene epoxidase of Micropsorum gypseum – In vitro and in silico studies". The authors tried to discover antifungal compounds from the phytochemicals of epicarp of Balanites aegyptiaca. They screened different extracts and fractions, then used LC-MS to elucidate the chemical structure of the constituents of the active fraction. They used in silico techniques to explain the activity of the identified compounds.

Also, add the following comments in the manuscript,

1.     The authors ran the dynamic simulation for upto 10ns, if possible please extend the time period (50ns). Because 10ns is very less time to say about the stability of the complex.

2.     Calculate the binding-free energy values of the compounds.

3.     Please Increase the quality of the figures.

4.     Please increase the quality of the language.

5.     Supplementary figure (Supp.Fig.1) and tables (Supp.Table 1,2 and 3) missing

6.     Below references are recommended to be also commented on where you discuss in the appropriate place. https://doi.org/10.1007/s11356-023-26412-8; https://doi.org/10.2174/1574885513666180402134054; https://doi.org/10.1016/j.mtcomm.2023.105652

7.     Reconsider, typo errors, spaces between words and punctuation especially before and after transition words.

The title of manuscript is remarkable. English language has good quality. There are some explainations that are needed about the section "Materials and methods" and "Results"

1. About section "Plant species collection and identification " in line 64

+ Please mention the name of the scientists that has performed the identification of the class of plant

+ Please write Geographical coordinates of the redion that you have collected the plant

+ Please mention that based on which scientific study you performed extracting with diverse chemical solvents of the plant.

2. Please explain that how you selected the selected amount of each concentration of the extract of the plant

3. About Figure 1

+ Please explain in detail what do various colors (black, green, yellow, gray and blue) stands for?

+ Please define the abbreviations of the figure in the title of this figure

4. Figure 3 have not enough quality to be in the manuscript because it contains some total fague information. Please ommit it from manuscript or replace it with other figure.

5. About the section "Molecular Dynamics Simulation" in line 282 in Page 12

+ Why you have not inserted the average amount of RMSD and RMSF as well as the standard deviation for both mentioned parameter?

+ Please write the upper limit of the proper

amount of RMSD and RMSF that show a stable bind

6. Please check and adjust the "Reference list" based on the regulations of reference list of journal. (Titles, doi, the name of journal and ... )
