Cover Story: Membrane protein integrase (MPIase) is the first known Escherichia coli glycolipid essential for membrane protein insertion. In this review, we discuss the various membrane properties that affect membrane protein insertion and the contributions of MPIase to these properties. We also present our comprehensive analysis of the membrane insertion activity, physicochemical properties, and molecular interactions between MPIase and its substrates. This provides an understanding of the atomic-level molecular mechanisms. MPIase prevents substrate protein aggregation, attracts substrates near the membrane surface, and loosens the membrane core to facilitate protein insertion into the membrane. These findings reveal novel biological roles for glycolipids. View this paper
