Special Issue Information

Dear Colleagues,

The development of new vectors, technologies, and delivery systems for nucleic acid transfer has led to a promising generation of gene therapy with less toxicity and side effects, and improved efficiency compared to those obtained in the “first” generation.

RNA has demonstrated strong potential as a candidate for nucleic acid-based therapeutics, and the ability to tackle previously "undruggable" targets. With the introduction of omics technologies, a deep understanding of RNA biology has been achieved, boosting the development of RNA therapeutics. Positive clinical trials results with RNA-based drugs have proven to be a promising strategy with a strong curative potential for the treatment of diverse diseases.

The application of RNA therapies for the treatment of pulmonary diseases is rapidly increasing due to the unique advantage of direct delivery of nucleic acids into the lung. Although there are still many obstacles to overcome to achieve safe clinical application, this strategy offers the possibility of treatment for a wide variety of respiratory system-related pathologies, including inflammatory lung diseases, fibrosis, and lung cancer.

This Special Issue will discuss the latest research findings on RNA-based gene therapies for pulmonary diseases with the aim of providing new insights into their future development.

We invite researchers to contribute original articles and reviews on the newly identified RNA-based therapies for the treatment of pulmonary diseases.

In this up-to-date study, we first aimed to highlight the genetic and non-genetic factors associated with respiratory distress syndrome (RDS) while also focusing on the genomic aspect of this condition. Secondly, we discuss the treatment options and the progressing therapies based on RNAs or gene therapy. To fulfill this, our study commences with lung organogenesis, a highly orchestrated procedure guided by an intricate network of conserved signaling pathways that ultimately oversee the processes of patterning, growth, and differentiation. Then, our review focuses on the molecular mechanisms contributing to both normal and abnormal lung growth and development and underscores the connections between genetic and non-genetic factors linked to neonatal RDS, with a particular emphasis on the genomic aspects of this condition and their implications for treatment choices and the advancing therapeutic approaches centered around RNAs or gene therapy.
