Low-cost ocean monitoring systems are increasingly needed to address data gaps in coastal environments, particularly in regions where traditional research infrastructure is limited. This paper presents the design, development, and field deployment of a biophysical ocean buoy (BOB)—a compact, solar-powered autonomous buoy system capable of measuring sea surface temperature, salinity (via electrical conductivity), total dissolved solids, pH, and GPS position. The system features real-time data transmission via the Iridium satellite, local data logging, and modular sensor integration. The BOB was deployed for three missions in the Seto Inland Sea, Japan, ranging from 26–56 h in duration. The system successfully recorded high-resolution environmental data, revealing coastal gradients, diurnal heating cycles, and tidal current reversals. Over 95% of the measurements were successfully recovered, and the Iridium communications exceeded 90% reliability. The temperature and salinity data captured fine-scale variations consistent with freshwater plume interactions and tidal forcing. With a total system cost under USD 2000 and minimal deployment requirements, the BOB offers a scalable solution for distributed ocean monitoring. Its performance suggests strong potential for use in aquaculture monitoring, coastal hazard detection, and climate change research, especially in data-sparse regions. This work contributes to the growing field of democratized ocean observation, combining affordability with operational reliability. Full article

Dysbiosis of the gastrointestinal tract is the most common cause of disease in childhood and adulthood. The formation of the intestinal microbiome begins in utero, and composition modification during life depends mainly on various genetic, nutritional, and environmental factors. The main cause of intestinal dysbiosis is improper nutrition due to a short period of breastfeeding, insufficient intake of fresh fruits and vegetables, and/or consumption of a large amount of processed food. The benefits of a diet based on grains, legumes, fruits, and vegetables are reflected in reducing the risk of cancer, cardiovascular diseases, myocardial infarction, stroke, rheumatoid arthritis, high blood pressure, asthma, allergies, and kidney stones. Anaerobic fermentation of fibers produces short-chain fatty acids (SCFA) that have an anti-inflammatory role and great importance in shaping the intestinal microbiota. Factors associated with high fiber in a plant-based diet promote increased insulin sensitivity. Insulin and insulin-like growth factor 1 (IGF-I) act as promoters of most normal and pre-neoplastic tissues. Conclusion: A plant-based diet high in fiber prevents disease by creating metabolites in the gut that reduce oxidative stress. Full article

Fresh highland barley is difficult to store, leading to a lack of commercial products. To address these problems, the research investigated the effect of different heat treatments (steaming <SFB>, microwaving <MFB>, baking <BFB>, and cooking <CFB>) on the quality of fresh highland barley, and used pretreated fresh highland barley as material, combined with the milk tea market, to design and optimize the preparation process of fresh highland barley tapioca pearl and milk tea BOBA. The results showed that the different heat treatments reduced the content of ash and starch significantly, and SFB and MFB decreased the digestibility of fresh highland barley (P < 0.05). In particular, SFB had a significantly higher overall score for fresh barley than the other treatments, with the highest sensory evaluation for aroma, elasticity, and the overall taste of the grain, and the eGI value was the lowest (58.64). The optimal preparation process of fresh highland barley tapioca pearl and milk tea BOBA was designed and optimized by the L9(34) orthogonal test. The optimal tapioca pearl formula contained the following: apioca starch content of 36%, cooking time of 2.5 min, and erythritol stevia content of 1.5%. The optimal milk tea BOBA formula contained the following: sodium alginate content of 1.3%, erythritol stevia content of 0.6%, and calcium lactate content of 2.2%. This not only improves the comprehensive utilization rate of fresh highland barley, but also provides the accessory food, ensuring a lower eGI and increasing the healthiness and diversity of milk tea. In light of expected climate change, it is important to seek nature-based solutions that can contribute to the protection of our planet as well as to help overcome the emerging adverse changes. In an agricultural context, increasing plant resistance to abiotic stress seems to be crucial. Therefore, the scope of the presented research was focused on the application of botanical extracts that exerted positive effects on model plants growing under controlled laboratory conditions, as well as plants subjected to sorbitol-induced osmotic stress. Foliar spraying increased the length and fresh mass of the shoots (e.g., extracts from Taraxacum officinale, Trifolium pratense, and Pisum sativum) and the roots (e.g., Solidago gigantea, Hypericum perforatum, and Pisum sativum) of cabbage seedlings grown under stressful conditions, as well as their content of photosynthetic pigments (Pisum sativum, Lens culinaris, and Hypericum perforatum) along with total phenolic compounds (Hypericum perforatum, Taraxacum officinale, and Urtica dioica). The antioxidant activity of the shoots measured with the use of DDPH (Pisum sativum, Taraxacum officinale, Urtica dioica, and Hypericum perforatum), ABTS (Trifolium pratense, Symphytum officinale, Valeriana officinalis, Pisum sativum, and Lens culinaris), and FRAP (Symphytum officinale, Valeriana officinalis, Urtica dioica, Hypericum perforatum, and Taraxacum officinale) assays was also enhanced in plants exposed to osmotic stress. Based on these findings, the most promising formulation based on Symphytum officinale was selected and subjected to transcriptomic analysis. The modification of the expression of the following genes was noted: Bol029651 (glutathione S-transferase), Bol027348 (chlorophyll A-B binding protein), Bol015841 (S-adenosylmethionine-dependent methyltransferases), Bol009860 (chlorophyll A-B binding protein), Bol022819 (GDSL lipase/esterase), Bol036512 (heat shock protein 70 family), Bol005916 (DnaJ Chaperone), Bol028754 (pre-mRNA splicing Prp18-interacting factor), Bol009568 (heat shock protein Hsp90 family), Bol039362 (gibberellin regulated protein), Bol007693 (B-box-type zinc finger), Bol034610 (RmlC-like cupin domain superfamily), Bol019811 (myb_SHAQKYF: myb-like DNA-binding domain, SHAQKYF class), Bol028965 (DA1-like Protein). Gene Ontology functional analysis indicated that the application of the extract led to a decrease in the expression of many genes related to the response to stress and photosynthetic systems, which may confirm a reduction in the level of oxidative stress in plants treated with biostimulants. The conducted studies showed that the use of innovative plant-based products exerted positive effects on crops and can be used to supplement current cultivation practices. Full article

Beta-ketothiolases are involved in the beta-oxidation of fatty acids and the metabolism of hormones, benzenoids, and hydroxybutyrate. The expression of bacterial beta-ketothiolase in flax (Linum usitatissimum L.) results in an increase in endogenous beta-ketothiolase mRNA levels and beta-hydroxybutyrate content. In the present work, the effect of overexpression of beta-ketothiolase on retting and stem and fibre composition of flax plants is presented. The content of the components was evaluated by high-performance liquid chromatography, gas chromatography–mass spectrometry, Fourier-transform infrared spectroscopy, and biochemical methods. Changes in the stem cell walls, especially in the lower lignin and pectin content, resulted in more efficient retting. The overexpression of beta-ketothiolase reduced the fatty acid and carotenoid contents in flax and affected the distribution of phenolic compounds between free and cell wall-bound components. The obtained fibres were characterized by a slightly lower content of phenolic compounds and changes in the composition of the cell wall. Based on the IR analysis, we concluded that the production of hydroxybutyrate reduced the cellulose crystallinity and led to the formation of shorter but more flexible cellulose chains, while not changing the content of the cell wall components. We speculate that the changes in chemical composition of the stems and fibres are the result of the regulatory properties of hydroxybutyrate. This provides us with a novel way to influence metabolic composition in agriculturally important crops. Full article

Nighttime light (NTL) is a critical indicator of urban vitality and development. Using NTL as a representation of urban vitality and development, the study explores how different fresh-made beverage shops, namely boba and coffee shops, proxy various facets of urban vitality and development in four megacities in China. Existing studies mostly discuss urban vitality as a broad concept and seldom investigate the diverse urban vitality and development represented by different indicators. This study selects Beijing, Shanghai, Guangzhou, and Shenzhen as case study regions and explores (1) their urban vitality pattern represented by NTL. (2) the heterogeneous spatial distribution of boba and coffee shops; (3) how boba and coffee shops represent urban vitality differently; and (4) how boba and coffee shops portray the economy and population growth aspect of urban development differently. We acquired NTL data from remote sensing images to measure urban vitality and development. Cross-sectionally, the majority of urban vitality and development represented by NTL concentrates in urban centers. The distribution of coffee shops assimilates the spatial pattern of urban vitality represented by NTL while boba shops have a greater spatial extent in metropolitan fringes. Longitudinally, from 2012 to 2020, the global and local bivariate Moran’s I analysis between NTL and beverage shops shows that the coffee shops capture urban vitality and development better than boba shops in Beijing, while the pattern is reversed in Guangzhou and Shenzhen. Examining the evolving spatial dynamics between beverage shops’ growth and urban development using bivariate Moran’s I and Getis–Ord Gi/Mann–Kendall emerging hot spot analysis, we found that the locations with the most intense economic growth have seen the most spatial expansion of coffee shops. In contrast, those with the fastest population growth have seen the greatest spatial development of boba businesses. These results indicate that coffee shops represent the economic aspect of urban vitality while boba shops emphasize the population growth aspects. By examining the dynamic spatio–temporal relationship between small beverage shops and urban vitality and development represented by NTL data, this study broadens the usage of remote sensing data in urban studies and expands on previous research and offers insights for urban planners and geographers to reference when choosing indicators of urban vitality and growth. Faba beans (Vicia faba L.) are a high-protein legume crop that can be widely cultivated in most climates in Europe. The amino acid composition of the faba bean protein is also beneficial for monogastric animals since it contains a great deal of lysine, an amino acid that is deficient in cereals. Two genotypes of faba beans were cultivated at three sowing densities (45, 60 and 75 seeds per m2) during three growing seasons (years 2013–2015). The aim of the research was to assess accumulation of nutrients (N, P, K, Ca and Mg) of two faba beans in seeds and straw under different sowing densities. A field experiment was conducted in 2013–2015 at fields of Wroclaw University of Environmental and Life Sciences in southwestern Poland from which plant material was used for chemical analysis in terms of determining the accumulation of the following elements—N, P, Mg, K and Ca—in the seeds and straw. The results showed that the genotypes of the faba bean varied greatly in accumulation of nutrients in various sowing densities and in following years of research. However, the average accumulation of nitrogen, calcium and magnesium in seeds for three years (2013–2015) was significantly higher in the Bobas cultivar under 60 seeds per m2 as well as in both tested cultivars using 75 seeds per m2. Accumulation of potassium and phosphorus in seeds was highest in both analyzed cultivars using 60 and 75 seeds per m2 (average for 2013–2015). Considering accumulation of elements in the straw of faba bean, it is shown that the concentration of potassium and calcium was the highest in both tested cultivars under the effect of sowing density at 60 and 75 seeds per m2 while the concentration of nitrogen, phosphorus and magnesium did not differ significantly as an effect of the interaction of cultivar and sowing date (average for 2013–2015). Weather conditions played an important role in accumulation of nutrients of both faba bean genotypes in the years of research (2013–2015). Full article

Patients with co-morbidities like cirrhosis are at risk of worse outcome from COVID-19 infection. Given limited prior studies, we evaluated outcomes associated with COVID-19 infection in alcoholic and non-alcoholic steatohepatitis cirrhotic (CC+) versus cirrhotic without COVID-19 (CC−). We performed retrospective analysis of 822,604 patients including 28,610 COVID-19 patients from the National Inpatient Sample database with alcoholic and NASH cirrhosis enrolled between 1 January 2020 to 31 December 2020, with univariate and multivariate regression analyses. Primary outcome was mortality and secondary outcomes was mechanical ventilation, vasopressor use, length of stay, hospitalization expense and predictors of mortality. In-hospital mortality was three time higher in the CC+ group compared to those in the CC− group(18.6% vs. 5.96%, p < 0.001, adjusted odds ratio (OR)3.39 (95% 3.08–3.74 CI). Hospitalization was more likely for underrepresented racial and ethnic groups with COVID-19 and cirrhosis. CC+ group had over twice the rates of mechanical ventilation (19.92% vs. 9.07%, adjusted OR 2.71 2.71 (95% 2.51–2.93 CI)),1.7 times likelihood of receiving vasopressors (4.12% vs. 2.45%, p < 0.001, adjusted OR 1.71 (95% CI 1.46–2.01). COVID-19 is associated with increased mortality in patients with alcoholic and NASH cirrhosis, and patients with alcoholic cirrhosis and COVID-19 have a slightly higher mortality compared to NASH cirrhosis. “Boba balls” or pearls have recently gained popularity for beverages or food toppings. “Boba balls” could be developed into functional foods by the encapsulation of bioactive compounds. In this study, gelatin/sodium alginate composite “Boba balls” enriched with pomegranate peel extract (PPE) at different concentrations (0, 1, 2, and 3%) were prepared. They were characterized in terms of physical, rheological, textural, morphological, and sensory properties, as well as in vitro digestion, bio-accessibility, and release kinetic of PPE. Adding PPE improved the “Boba” mix’s viscoelasticity and decreased the “Boba balls”’ hardness. The increasing PPE ratio significantly (p < 0.05) increased the antioxidant capacity and total phenolic content. The addition of PPE preserved the spherical shape of the “Boba balls”, and as the PPE ratio increased, new junction zones were observed in SEM images. The in vitro digestibility of PPE was significantly (p < 0.05) improved by preserving PPE from the mouth and gastric medium, and “Boba balls” showed the highest release and bio-accessibility in the intestinal medium. Consequently, PPE as a by-product could be successfully used at 2% concentration for enhancing the functionality and bio-accessibility of “Boba balls” without affecting sensory properties. Fusarium culmorum is a ubiquitous soil pathogen with a wide host range. In flax (Linum ussitatissimum), it causes foot and root rot and accumulation of mycotoxins in flax products. Fungal infections lead to huge losses in the flax industry. Moreover, due to mycotoxin accumulation, flax products constitute a potential threat to the consumers. We discovered that the defense against this pathogen in flax is based on early oxidative burst among others. In flax plants infected with F. culmorum, the most affected genes are connected with ROS production and processing, callose synthesis and ABA production. We hypothesize that ABA triggers defense mechanism in flax and is a significant player in a successful response to infection. Full article

Diet bioactive components, in the concept of nutrigenetics and nutrigenomics, consist of food constituents, which can transfer information from the external environment and influence gene expression in the cell and thus the function of the whole organism. It is crucial to regard food not only as the source of energy and basic nutriments, crucial for living and organism development, but also as the factor influencing health/disease, biochemical mechanisms, and activation of biochemical pathways. Bioactive components of the diet regulate gene expression through changes in the chromatin structure (including DNA methylation and histone modification), non-coding RNA, activation of transcription factors by signalling cascades, or direct ligand binding to the nuclear receptors. Analysis of interactions between diet components and human genome structure and gene activity is a modern approach that will help to better understand these relations and will allow designing dietary guidances, which can help maintain good health. Sowing depth and density are factors which affect development and yielding due to their influence on plants’ competition for water, light and nutrients. The aim of the present research was to evaluate the effect of sowing depth (5 and 8 cm) and density (45, 60, 75 seeds m2) on the development and yield of two morphotypes of faba bean. Higher plants were found at 8 cm sowing depth in 2011 and 2013, in turn, plant density before harvest, at 5 cm in 2011 and 8 cm in 2013. The greatest seed yield was observed while sowing shallower in 2011 (4.50 t ha−1) and 2012 (6.62 t ha−1), and deeper in 2013 (3.53 t ha−1). Cultivar played in important role in shaping plant height and seed yield in all years of the experiment, as well as in straw yield in 2012. Bobas characterized by the greatest seed productivity in all years of experiment compared to Granit, as well as quantity of straw yield in 2011 and 2012 (3.51 and 4.88 t ha−1, respectively) and therefore can be recommended for cultivation. Taking into account plant density before harvest in 2011–2013 as well as straw yield in 2011 and 2013, the most favorable sowing density is 75 seeds per m2. Full article

Boba milk tea is very popular around the world. The “boba” balls in milk tea are usually made of tapioca. Reports on calcium alginate ball encapsulation in fruit-flavoured drinks have rarely been seen. The preparation method for this kind ball was studied. The “boba” balls were obtained by membrane formation on the interface through the addition of calcium chloride fluids into a sodium alginate solution. The operation conditions were studied, including drop height, flow velocity, sodium alginate and calcium chloride solution concentration. The diameter, mechanical strength, loading ratio and encapsulation rate of the “boba” balls are discussed. The optimized preparation conditions were as follows: the diameter of adding tube was 8 mm, the drop height was 25 cm, the drop flow rate was 60 mL/min, 1.0% sodium alginate, 1.0% calcium chloride. The prepared “boba” balls were stored at different temperatures. No microorganisms were detected in 90 days, and the sensory quality decreased with storage time. Shelf life was predicted using the Arrhenius equation; when the storage temperature was less than 10 °C, it could be stored for more than 1 year. This preparation technology of “boba” balls has potential for application by milk tea ingredient companies or relevant beverage manufacturing factories. Full article

Catecholamines are biogenic aromatic amines common among both animals and plants. In animals, they are synthesized via tyrosine hydroxylation, while both hydroxylation or decarboxylation of tyrosine are possible in plants, depending on the species, though no tyrosine hydroxylase—a counterpart of the animal enzyme—has been identified yet. It is known that in potato plants, it is the decarboxylation of tyrosine that leads to catecholamine production. In this paper, we present the effects of the induction of an alternative route of catecholamine production by introducing the tyrosine hydroxylase gene from rat. We demonstrate that an animal system can be used by the plant. However, it does not function to synthesize catecholamines. Instead, it leads to elevated reactive oxygen species content and a constant stress condition in the plant, which responds with elevated antioxidant levels and improved resistance to infection. Most losses in flax (Linum usitatissimum L.) crops are caused by fungal infections. The new epigenetic approach to improve plant resistance requires broadening the knowledge about the influence of pathogenic and non-pathogenic Fusarium oxysporum strains on changes in the profile of DNA methylation. Two contrasting effects on the levels of methylation in flax have been detected for both types of Fusarium strain infection: Genome-wide hypermethylation and hypomethylation of resistance-related genes (β-1,3-glucanase and chitinase). Despite the differences in methylation profile, the expression of these genes increased. Plants pretreated with the non-pathogenic strain memorize the hypomethylation pattern and then react more efficiently upon pathogen infection. The peak of demethylation correlates with the alteration in gene expression induced by the non-pathogenic strain. In the case of pathogen infection, the expression peak lags behind the gene demethylation. Dynamic changes in tetramer methylation induced by both pathogenic and non-pathogenic Fusarium strains are dependent on the ratio between the level of methyltransferase and demethylase gene expression. Infection with both Fusarium strains suppressed methyltransferase expression and increased the demethylase (demeter) transcript level. The obtained results provide important new information about changes in methylation profile and thus expression regulation of pathogenesis-related genes in the flax plant response to stressors.
