The widespread prevalence of different serotypes of fowl adenoviruses (FAdVs) has led to diverse vaccine demands, especially for subunit vaccines targeting FAdV-4 and FAdV-11, which cause hydropericardium-hepatitis syndrome (HHS) and inclusion body hepatitis (IBH), respectively. Although the Fiber protein is known to elicit robust immune protection, further exploration is needed to enhance the production of cross-neutralizing antibodies. This study utilized structural prediction and homology modeling techniques, employing domain-swapping strategy to integrate neutralizing epitope-containing amino acid sequences (274–451aa and 364–543aa) into the shaft domain of the Fiber protein. Two novel chimeric proteins were recombinantly expressed and developed into subunit vaccines: Fiber-C4/D11 and Fiber-D11/C4. Immunogenicity assessments revealed that the Fiber-C4/D11 vaccine group rapidly induced an antibody response against FAdV-11 within 7 days post-vaccination. By 28 days post-vaccination (dpv), the Fiber-C4/D11 vaccine group exhibited significantly higher levels of cross-neutralizing antibodies compared to the Fiber-D11/C4 group (p < 0.05). Challenge experiments demonstrated that both vaccines effectively alleviated clinical symptoms and prevented mortality in SPF chickens. Compared to Fiber-D11/C4, Fiber-C4/D11 significantly reduced body weight loss, liver lesions, viral titers in tissues, and viral shedding. Notably, no cross-neutralizing antibodies were detected following FAdV-4 or FAdV-11 infection, indicating a lack of natural cross-protection between the two serotypes. The chimeric vaccine addressed this gap, offering a promising multivalent approach to mitigate FAdV infections and advancing fowl adenoviral subunit vaccine strategies. Full article

Insect bite hypersensitivity (IBH) is a seasonally recurrent allergic dermatitis representing one of the most prevalent dermatological conditions in horses worldwide. This condition, driven by hypersensitivity to salivary allergens of Culicoides spp., causes substantial discomfort, welfare impairment, and potentially economic loss in equine populations. The pathogenesis of IBH is complex, involving genetic predisposition, epithelial barrier dysfunction, and a skewed T-helper 2 (Th2)-mediated immune response with elevated IgE production and eosinophilic inflammation. Advances in immunogenetics and molecular immunology have improved the understanding of the disease’s multifactorial nature. Research on immunotherapy and cytokine-targeted treatments is contributing to the development of more effective therapeutic options. This review synthesizes current knowledge on the immunopathogenesis and genetic determinants of IBH and discusses both conventional and emerging strategies for its clinical management. Full article

Fowl adenovirus can cause various diseases such as hepatitis-hydropericardium syndrome (HHS) and inclusion body hepatitis (IBH), which leads to huge economic losses for China’s poultry industry. In recent years, the prevalence of FAdV-4, FAdV-8a, and FAdV-8b has been on the rise. In this study, a trivalent fowl adenovirus inactivated vaccine against FAdV-4/8a/8b was generated and the immune effect was evaluated. The results showed that chickens immunized with the trivalent vaccine could effectively resist the challenge of virulent strains of FAdV-4, FAdV-8a, and FAdV-8b. All chickens in the immunized groups survived after virus injection, gained weight normally, and the lesions in tissues and organs (liver, spleen, kidney, etc.) were significantly decreased compared with the control group. The viral load in the liver of the immunized group was significantly lower than that of the challenge control group five days after the challenge (p < 0.01), and the cloacal excretion was significantly lower than that of the non-immune control group (p < 0.01). Moreover, until 16 weeks after immunization, the neutralizing antibody level against FAdV-4, FAdV-8a, and FAdV-8b remained above 8.27 log2. As a result, the trivalent vaccine generated in our study was an attractive candidate to prevent and reduce the spread of HHS and IBH in China’s poultry industry. Full article

Fowl adenoviruses (FAdVs) are important emerging pathogens affecting the poultry industry in Egypt as they are the primary etiology of inclusion body hepatitis–hydropericardium syndrome (IBH-HPS) associated with severe economic losses. This study aims to identify the circulating FAdVs from cases of IBH-HPS in 5 Egyptian provinces during the period from October 2020 through September 2022. Out of the 210 examined flocks, liver samples from 66 flocks were positive for FAdVs (31.4%) using conventional polymerase chain reaction targeting loop 1 of the major hexon gene, with varying rates of mortality (1% to 14%). In the positive samples detected during the study, the histopathological examination revealed pathognomonic lesions of FAdVs, including basophilic and eosinophilic intra-nuclear inclusion bodies (INIBs). The percentage of FAdV positivity increased with the flock age; from samples collected at ages 1 to10, 11 to 20, 21 to 30, and >30 days of age, 10% (5/50), 25.6% (11/43), 34.3% (23/67), and 54% (27/50) were found positive for FAdVs, respectively. Notably, the positivity percentages among the flocks reared in cages were higher than for those reared in the deep litter system of housing. The gene sequencing and phylogenetic analysis of 19 strains revealed clustering into FAdV species D serotype 2/11, demonstrating that serotype 2/11 is most prevalent in the targeted Egyptian provinces during the period of the study. Several point mutations in the sequenced region among different strains were reported. These findings underscore the prevalence of FAdV and provide a basis for further research on circulating strains to develop effective control strategies. Full article

The chicken infectious anemia virus (CIAV) and fowl adenovirus serotype E8b (FAdV E8b) are pathogens that cause aplastic anemia and inclusion body hepatitis (IBH) in chickens, respectively. The co-infection of CIAV and FAdV E8b poses a significant threat to poultry health, potentially worsening clinical symptoms and increasing mortality rates. This study aimed to explore the combined pathogenic effects of FAdV E8b and CIAV co-infection on one-day-old specific pathogen-free (SPF) chickens. The results showed that co-infection led to significantly higher clinical scores and mortality rates compared to FAdV E8b infection alone. Additionally, there were different tissue distribution patterns for FAdV E8b between the single infection and co-infection groups, indicating potential changes in viral tropism. Biochemical analysis revealed elevated markers of liver and/or muscle damage in both the FAdV E8b infection group and the co-infection group, consistent with the viral infection process. These findings suggest that co-infection with FAdV E8b and CIAV can intensify clinical signs and mortality, and may potentially alter viral replication and tissue tropism in chickens. This study establishes a foundation for future investigations into the underlying mechanisms governing the interaction between CIAV and FAdV E8b during co-infection. Full article

In the United States (US), a longstanding solution to the unmet need for mental health treatment is integrated behavioral health care (IBH). Within a primary care model, problems are identified and treatment combines care for physical, mental and substance use disorders. Treatments are delivered through the collaboration of primary and behavioral health providers. According to US federal billing guidelines, in one integrated model, the Collaborative Care Model (CoCM), the psychiatric consultant must be a medical professional trained in psychiatry and capable of prescribing medications, i.e., either a psychiatrist, Psychiatric Mental Health Nurse Practitioner (PMHNP) or Physician Assistant. The development of integrated care has been slow for particular vulnerable populations, in part due to the lack of psychiatric consultants. PMHNPs are increasingly taking on the role of psychiatric consultants on CoCM teams and creating nurse-led IBH models for underserved populations. In this paper, eight such models are discussed along with implementation challenges and the strategies used to address them. Nurse leaders have the capacity to enhance and expand integrated care, particularly for underserved populations, through the optimal utilization of care teams, expanding measured outcomes, and developing measures for team-based effectiveness. Future directions are proposed that will accelerate this PMHNP-led expansion of IBH. Full article

Fowl adenovirus (FAdV) belongs to the Aviadenovirus genus within the Adenoviridae family. FAdVs are widely distributed and associated with various diseases in poultry, including adenoviral gizzard erosion (AGE), hepatitis-hydropericardium syndrome (HHS), and inclusion body hepatitis (IBH). In this study, we developed a multiplex conventional PCR for simultaneously detecting FAdV-4, -8b, and -11 by targeting the hexon gene. The multiplex PCR was optimized for primer concentrations and thermocycling conditions. The optimal primer concentration combination was set at 0.125 μM for FAdV-4, -8b, and 0.25 μM for FAdV-11. Under these conditions, the limit of detection (LOD) was 103 copies/μL of plasmid standards for FAdV-4, -8b, and -11. These results demonstrated that the developed multiplex PCR method exhibits high specificity and sensitivity, with no observed cross-reactivity among these serotypes or with other poultry viruses. Therefore, this multiplex PCR will be an effective tool for accurate serotyping of FAdV-4, -8b, and -11, enabling more precise identification and differentiation of these three serotypes. Eosinophils play a key role in allergic diseases such as insect bite hypersensitivity (IBH). Together with Th2 cells, they shape the course of inflammation in associated type I/IVb allergies. Therefore, a virus-like particle (VLP)-based vaccine targeting equine interleukin-5 (eIL-5), eIL-5-CuMV-TT, was developed to interfere with the IL-5 dependency of eosinophils by inducing the production of anti-self-IL-5 antibodies and alleviating clinical signs in IBH-affected horses. A previous study highlighted the presence of two eosinophil subsets, steady-state resident eosinophils (rEos) and inflammatory eosinophils (iEos), circulating in the blood of healthy and IBH-affected horses, distinguishable by the expression of integrin CD49f. Furthermore, eIL-5-CuMV-TT 1st year vaccination showed a significant decrease of total eosinophils and, in particular, iEos. Nevertheless, the very few remaining eosinophils still shared an iEos phenotype, reflected by bigger size and higher granularity. The aim of this study was to follow up on the phenotype of eosinophils in the 2nd year of vaccination of IBH-affected horses with eIL-5-CuMV-TT. Using flow cytometry analysis of the blood of healthy, IBH, IBH-placebo, and IBH-vaccinated horses, the percentage and count of cells were compared between groups with a focus on pair analysis of eosinophils in 1st and 2nd year vaccinated horses. Our data showed comparably low levels of iEos and a significant increase of rEos in 2nd year compared to 1st year vaccinated horses, suggesting a phenotypic shift toward a resident-like eosinophil population, primarily associated with the phenotype of healthy horses. The reduction of size, granularity, and expression of integrin CD49f in the 2nd year suggests a benefit of long-term treatment with the eIL-5-CuMV-TT vaccine. Full article

Zika virus (ZIKV), a mosquito-borne flavivirus, is a significant global health concern due to its association with neurodevelopmental disorders such as congenital Zika syndrome (CZS). This study aimed to compare the replication kinetics, viral persistence, cytopathogenic effects, and immune gene expression in human microglia cells (CHME-3) infected with an Asian lineage ZIKV (PRVABC59, referred to as ZIKV-PRV) and an African lineage ZIKV (IBH30656, referred to as ZIKV-IBH). We found that ZIKV-PRV replicated more efficiently and persisted longer while inducing lower levels of cell death and inflammatory gene activation compared with ZIKV-IBH. These findings suggest that the enhanced replication and persistence of ZIKV-PRV, along with its ability to evade innate immune responses, may underlie its increased neuropathogenic potential, especially in the context of CZS. In contrast, ZIKV-IBH, with its stronger immune gene activation and higher cytopathogenicity, may lead to more acute infections with faster viral clearance, thereby reducing the likelihood of chronic central nervous system (CNS) infection. This study provides crucial insights into the molecular and cellular mechanisms driving the differential pathogenicity of ZIKV lineages and highlights the need for further research to pinpoint the viral factors responsible for these distinct clinical outcomes. With a focus on education and teaching, we provide general background information on bioinspired optimization methods by comparing the concept of optimization and the search for an optimum in engineering and biology. We introduce both the principles of Darwinian evolution and the basic evolutionary optimization procedure of evolution strategies. We provide three educational modules in work sheets that can be used by teachers and students to improve their understanding of evolution strategies. The educational module “Optimization of a Milk Carton” shows that the material consumption in producing a milk carton can be minimized using an evolution strategy with a mutative step size control. The use of a standard dice and a pocket calculator enables new milk cartons to be generated, with the offspring having the lowest material consumption becoming the parent of the next generation. The other educational modules deal with the so-called brachistochrone problem. The module “Fastest and Shortest Marble Track” provides a construction plan for a marble track whereby students can experimentally compare the “path of shortest length” with the “path of shortest time”. The EvoBrach software, is used in the module “Various Marble Track Shapes” to compare the running times of a marble on a straight line, a parabola, and a brachistochrone. In conclusion, the introduction to the biomimetic method of evolution strategies and the educational modules should deepen the understanding of both optimization problems and biological evolution. Fowl aviadenovirus (FAdV) is a member of the Aviadenovirus genus within the Adenoviridae family. FAdVs are divided into five species based on genomic differences: Fowl aviadenovirus A to Fowl aviadenovirus E (FAdV-A to FAdV-E). They are classified into twelve serotypes (FAdV-1 to FAdV-8a and FAdV-8b to FAdV-11) through cross-neutralization tests. FAdVs are mainly associated with hepatitis hydropericardium syndrome (HHS), adenoviral gizzard erosion (AGE), and inclusion body hepatitis (IBH). The serotypes commonly involved in IBH are FAdV-2, FAdV-11, FAdV-8a, and FAdV-8b. IBH causes significant economic losses in the poultry industry, mainly due to high mortality, reduced productivity, and immunosuppression. This is the first case report on IBH in Chile caused—according to post-mortem findings, molecular analysis, sequencing, and phylogenetic analysis—by FAdV-11. Since the serotype had not previously been reported in Chile, continued monitoring of IBH cases is required to determine the serotype of the circulating FAdVs and adapt preventative vaccination programs. Brassinosteroids (BRs) play critical roles in plant growth by promoting cell elongation and division, leading to increased leaf inclination angles. BRASSINAZOLE RESISTANT 1 (BZR1) and BRI1-EMS-SUPPRESSOR 1 (BES1) act as transcription factors in the brassinosteroid signaling pathway and are involved in several physiological activities regulated by BRs. In this study, we identified and cloned BcBZR1 from the heitacai non-heading Chinese cabbage (NHCC) cultivar. The sequence analysis showed that the coding sequence length of BcBZR1 is 996 bp, encoding 331 amino acid residues. Subcellular localization assays showed that BcBZR1 is localized in the nucleus and cytoplasm and that BcBZR1 protein is transported to the nucleus after receiving BR signals. Compared with Col-0, the leaf inclination angle was smaller in BcBZR1-OX. The EBR treatment experiment indicated that BRs regulate the differential expression of paclobutrazol resistance1 (PRE1) and ILI1 binding bHLH1 (IBH1) in the adaxial and abaxial cells of the petiole through BZR1, thus regulating the leaf inclination angle. The bimolecular fluorescence complementation (BiFC) assay indicated that BcBZR1 interacts with C-repeat Binding Factor2 (BcCBF2) and CBF3. Taken together, our findings not only validate the function of BcBZR1 in leaf inclination angle distribution in non-heading Chinese cabbage, but also contribute to the mechanism of leaf inclination angle regulation in this species under cold stress. Thirty controls (C) and 30 IBH-affected (T) Lusitano horses were evaluated. T horses were included based on anamnesis and physical examination, supported by questionnaires. All horses were submitted to skin tests, Intrademal (IDT) and Skin Prick Tests (SPT), on the neck with 14 specific allergens, 13 recombinant proteins (r-proteins) from Culicoides nubeculosus (Cul n) and Culicoides obsoletus (Cul o) salivary glands and Culicoides nubeculosus Whole Body Extract (Cul n WBE). Addicionally, a cluster of six T and six C horses were also tested with Cul n 3 and Cul n 4 produced in insect cells and barley, as well as E. coli produced Cul o 3 and Cul o WBE. Allergen concentrations were 10 µg/mL for IDT and 100 µg/mL for SPT, and wheal diameters assessed at 20 min, 6 and 48 h. IDTs were considered positive when wheal diameter was ≥50% of the histamine wheal and SPT’s ≥ 0.9 cm. In vitro tests, allergen-specific serum IgE and sulfidoleukotriene (sLT) release assay were also carried out. Results showed that Cul n WBE, Cul n 7, 8, 9, Cul o1P and Cul o 2P were the best performing allergens for SPTs (p ≤ 0.0001) for the 1st allergen panel and Cul o WBE, Cul n 3 Bar and Cul n 4 Bac (p ≤ 0.05) for the 2nd, presenting a higher discriminatory diagnostic potential than IDTs, at a concentration of 100 µg/mL, with readings assessed at 20 min. Regarding in vitro tests overall, the sLT release assay performed best. Full article

Insect Bite Hypersensitivity (IBH, Queensland itch, sweet itch, equine summer eczema) is the most common pruritic disease of horses. It is most often caused by sensitivity to the saliva of Culicoides spp. of biting midges; however, it can also be caused by hypersensitivity to other insect species. The prevalence of IBH in horses is reported to be as high as 60% in some parts of the world. Due to the severe pruritus and effects of secondary self-trauma, IBH has animal welfare concerns, and there is currently no cure. Management of this condition is life-long, time consuming and costly. New grading systems to document disease severity are being validated, which will allow the comparison of clinical trial results of new and existing therapies. Management involves the minimisation of insect bites by use of stabling, fans, rugs and repellents. Symptomatic therapy involves the administration of systemic or topical corticosteroids, systemic antihistamines, and creams and sprays to promote skin healing and decrease inflammation. New immune-mediated therapeutics including vaccines, in addition to desensitisation procedures, show promise at controlling hypersensitivity reactions. This article will review aetiologic agents, pathophysiology, scoring systems and current and new therapies. Full article

Insect bite hypersensitivity (IBH) associated with Culicoides biting midges is a common allergic skin disease in horses, reducing the welfare of affected horses. This study investigated the effect of IBH on animal welfare and behaviour and assessed a new prophylactic insect repellent. In total, 30 horses were recruited for a prospective cross-over and case–control study. Clinical signs of IBH, inflammatory markers in skin biopsies and behavioural data (direct observations, motion index) were scored longitudinally during two consecutive summers. No differences were observed in the total number of itching behaviours or motion index between IBH-affected horses and controls, but higher numbers of itching behaviours were observed in the evening. IBH-affected horses showed both clinical and histopathological signs of inflammatory skin lesions, with even short periods of scratching being associated with moderate/severe inflammatory skin lesions. In order to improve the welfare of the IBH-affected horses, they should be stabled/given extra protection in the evening and even short-term exposure to Culicoides should be avoided. Preliminary results showed that the repellent tested can be used as a safe and non-toxic prophylactic to potentially reduce allergen exposure in horses with IBH, but further studies are needed to determine its efficacy.
