Background: Gallic acid (GA) is a dietary polyphenol widely found in walnuts, tea leaves, and grapes, and it is recognized for its potent antioxidant and anti-inflammatory properties. Chronic sleep deprivation (CSD) is known to disrupt redox balance, promote neuroinflammation, and impair cognition, while effective nutritional strategies to mitigate these effects remain scarce. This study was designed to evaluate the protective potential of GA against CSD-induced cognitive deficits in mice and to elucidate the underlying mechanisms. Methods: Seventy-two male ICR mice were randomly allocated to six groups, including control, CSD model, Ginkgo biloba extract, and GA at three doses (50, 100, and 200 mg/kg). After 28 days of treatment, cognitive performance was assessed using the open field test (OFT), novel object recognition (NOR), step-through passive avoidance (ST), and Morris water maze (MWM). Redox status and inflammatory mediators were determined by ELISA, while the hippocampal expression of proteins related to antioxidant defense and NF-κB signaling was analyzed by Western blotting. Results: GA supplementation improved exploratory activity, recognition memory, and spatial learning in the CSD mice. Biochemical evaluation revealed that total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) activity were restored, while malondialdehyde (MDA) levels, an indicator of lipid peroxidation, were reduced. These changes were accompanied by decreased circulating concentrations of interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α). At the molecular level, GA enhanced the expression of Nrf2, HO-1, and NQO1, while inhibiting p-p65, iNOS, and COX2 in the hippocampus. Conclusions: These findings demonstrate that GA alleviates CSD-induced cognitive deficits through the activation of the Nrf2/HO-1 antioxidant pathway and inhibition of NF-κB–mediated inflammatory responses. Thus, GA may represent a promising nutraceutical candidate for maintaining cognitive health under chronic sleep loss. Full article

Chilean hop (Humulus lupulus L.) ecotypes are an under-explored resource with high bioactive potential, offering promising applications in food preservation and health promotion. This study aimed to characterize the chemical composition and evaluate the antioxidant, antimicrobial, and cytotoxic properties of methanolic extracts from three native ecotypes—Ranco, La Unión, and Valdivia—to identify their potential as sources of multifunctional bioactive compounds. Each ecotype exhibited a distinct composition of bioactive compounds; Valdivia stood out for its pronounced levels of α- and β-acids and xanthohumol. Antioxidant capacity, assessed by DPPH, FRAP, and ABTS, was strong across extracts, with Valdivia showing the highest values in all the tests carried out. The extracts inhibited multidrug-resistant clinical isolates, notably Enterococcus faecalis and Pseudomonas aeruginosa, and showed dose-dependent cytotoxic effects in H1299 and MCF-7 cell lines, with the La Unión extract particularly active against H1299. Overall, these findings position Chilean hop ecotypes as promising sources of natural antioxidants and antimicrobial agents for functional food and nutraceutical applications. Full article

The Atlantic blue crab (Callinectes sapidus) is an opportunistic invasive species in the Mediterranean that is negatively affecting biodiversity, fisheries, and tourism. In Italy, it is appreciated for its good meat quality, but the processing yield is low (21.87 ± 2.38%), generating a significant amount of by-products (72.45 ± 4.08%), which are underutilized. Valorizing this biomass is in line with circular economy principles and can improve both environmental and economic sustainability. This study aimed to valorize Atlantic blue crab by-products (BCBP), producing protein hydrolysates and assessing their in vitro bioactivities, in order to plan applications in animal food and related sectors. BCBP hydrolysates were obtained by enzymatic hydrolysis using Alcalase and Protamex enzymes. The treatment with Alcalase resulted in a higher degree of hydrolysis (DH = 23% in 205 min) compared to Protamex (DH = 14% in 175 min). Antioxidant activity of the hydrolisates was evaluated through DPPH, ABTS, reducing power and FRAP assays, as well as in vitro test in fibroblasts (HS-68). At 10 mg/mL, hydrolysates from both enzymes exhibited the maximum radical scavenging activity in DPPH and ABTS assays. In HS-68 cells, 0.5 mg/mL hydrolysates protected against H2O2-induced oxidative stress, showing a cell viability comparable to cells treated with 0.5 mM N-acetyl cysteine (NAC), as an antioxidant. Statistical analyses were performed using one-way ANOVA followed by Student–Newman–Keuls (SNK) or Games–Howell post hoc tests, with significance set at p < 0.05. Overall, both enzymes efficiently hydrolyzed BCBP proteins, generating hydrolysates with significant antioxidant activity and cytoprotective effects. These results demonstrate the potential to produce high-quality bioactive compounds from BCBPs, suitable for food, nutraceutical, and health applications. Scaling up this valorization process represents a viable strategy to improve sustainability and add economic value to the management of this invasive species, turning a problem in a resource. Cannabidiol (CBD) and its derivatives show interesting therapeutic potential, including antioxidant, anti-inflammatory, and anticancer properties; however, their clinical translation remains a complex task due to physicochemical restrictions such as low water solubility, high lipophilicity, and instability under light, oxygen, and high temperatures. Polymeric encapsulation has emerged as a promising strategy to overcome these challenges, offering protection against environmental degradation, improved bioavailability, and controlled release. Natural and synthetic polymers, both biocompatible and biodegradable, provide versatile matrices for CBD delivery, enabling nanoparticle formation, targeted transport, and enhanced pharmacokinetics. This review highlights the structural characteristics of CBD, its interaction mechanisms with polymeric matrices such as hydrogels, electrospun nanofibers, biodegradable microparticles, thin films, and lipid-polymer hybrid systems, and the principal encapsulation techniques, such as emulsion solvent evaporation, electrospinning, and supercritical fluid technologies, that facilitate stability and scalability. Furthermore, material characterization approaches, including microscopy, thermal, and degradation analyses, are discussed as tools for optimizing encapsulation systems. While notable advances have been made, key challenges remain in achieving reproducible large-scale production, ensuring regulatory compliance, and designing smart polymeric carriers personalized for specific therapeutic contexts. By addressing these gaps, polymer-based encapsulation may unlock new opportunities for CBD in pharmaceutical, nutraceutical, and therapeutic applications, providing a guide for future innovation and translation into effective patient-centered products. The parallel global increase in obesity and Alzheimer’s disease (AD) underscores an urgent public health challenge, with converging evidence indicating that metabolic dysfunction strongly contributes to neurodegeneration. Obesity is now recognized not only as a systemic metabolic condition but also as a modifiable risk factor for AD, acting through mechanisms such as chronic low-grade inflammation, insulin resistance, and adipose tissue dysfunction. Among the molecular mediators at this interface, adipokines have emerged as pivotal regulators linking metabolic imbalance to cognitive decline. Adipokines are hormone-like proteins secreted by adipose tissue, including adiponectin, leptin, and resistin, that regulate metabolism, inflammation and can influence brain function. Resistin, frequently elevated in obesity, promotes neuroinflammation, disrupts insulin signaling, and accelerates β-amyloid (Aβ) deposition and tau pathology. Conversely, adiponectin enhances insulin sensitivity, suppresses oxidative stress, and supports mitochondrial and endothelial function, thereby exerting neuroprotective actions. The imbalance between resistin and adiponectin may shift the central nervous system toward a pro-inflammatory and metabolically compromised state that predisposes to neurodegeneration. Beyond their mechanistic relevance, adipokines hold translational promise as biomarkers for early risk stratification and therapeutic monitoring. Importantly, natural compounds, including polyphenols, alkaloids, and terpenoids, have shown the capacity to modulate adipokine signaling, restore metabolic homeostasis, and attenuate AD-related pathology in preclinical models. This positions adipokines not only as pathogenic mediators but also as therapeutic targets at the intersection of diabetes, obesity, and dementia. By integrating mechanistic, clinical, and pharmacological evidence, this review emphasizes adipokine signaling as a novel axis for intervention and highlights natural compound-based strategies as emerging therapeutic approaches in obesity-associated AD. Beyond nutraceuticals, antidiabetic agents also modulate adipokines and AD-relevant pathways. GLP-1 receptor agonists, metformin, and thiazolidinediones tend to increase adiponectin and reduce inflammatory tone, while SGLT2 and DPP-4 inhibitors exert systemic anti-inflammatory and hemodynamic benefits with emerging but still limited cognitive evidence. Together, these drug classes offer mechanistically grounded strategies to target the adipokine–inflammation–metabolism axis in obesity-associated AD. Full article

Background: Pomegranate peel (Punica granatum L.) is a rich source of phenols, particularly ellagitannins, highlighting punicalagin, a bioactive compound with recognized antioxidant potential. However, efficient recovery and purification methods are required to enable its application in food and health-related products. This study aimed to obtain a partially purified fraction of punicalagin from pomegranate peel using optimized extraction and purification strategies. Methods: A Taguchi L9 (3)3 experimental design was employed to optimize ultrasound-assisted extraction, evaluating extraction time (10, 20, 30 min), ethanol concentration (20, 40, 80%), and solid-to-solvent ratio (1:12, 1:14, 1:16). Total polyphenol content was quantified using the Folin–Ciocalteu method. Extracts obtained under optimized conditions were concentrated by rotary evaporation and subjected to semipurification using flash chromatography with Amberlite XAD-16 resin. Subsequently, the fractions were lyophilized and analyzed by HPLC/ESI/MS. Results: The Statistica software determined the optimal conditions for polyphenol extraction (20 min, 40% ethanol, 1:12), with the signal-to-noise (S/N) ratio reaching 88.43 ± 0.66, surpassing the predicted value of 77.42. Flash chromatography yielded four fractions, and HPLC/ESI/MS analysis revealed the presence of ellagitannins in all of them, with fraction number 2 showing the highest relative abundance of punicalagin (89.25%). Conclusions: The combination of ultrasound-assisted extraction and flash chromatography proved effective for obtaining punicalagin-rich fractions from pomegranate peel, supporting its potential for nutraceutical applications. Macroalgal polysaccharides represent a diverse group of structurally complex biopolymers with significant potential in biomedicine and functional food applications. This review provides a comprehensive examination of their structural features, biological activities, and molecular targets, with an emphasis on precision applications. Key polysaccharides such as alginates, carrageenans, fucoidans, ulvans, and laminarans are highlighted, focusing on their unique chemical backbones, degrees of sulfation, and branching patterns that underlie their bioactivity. Special attention is given to their roles in modulating inflammation, oxidative stress, apoptosis, gut microbiota, and metabolic pathways. Comparative assessment of extraction strategies, structure–function relationships, and bioactivity data highlights the importance of tailoring polysaccharide processing methods to preserve bioefficacy. Emerging insights from computational modelling and receptor-binding studies reveal promising interactions with immune and apoptotic signalling cascades, suggesting new therapeutic opportunities. Finally, the review outlines challenges related to standardisation, scalability, and regulatory approval, while proposing avenues for future research toward clinical translation and industrial innovation. By integrating structural biology, pharmacology, and nutraceutical sciences, this work underscores the potential of macroalgal polysaccharides as precision agents in health-promoting formulations and next-generation functional foods. Physicochemical and Functional Characterization of Pearl Millet-Based Probiotic Beverage for Antiaging Potential in Caenorhabditis elegans

Probiotics like Lactobacillus sp. are extensively studied for their beneficial host interactions, including the gut–brain axis, anti-inflammatory effects, immune system interactions, restoration of gut dysbiosis, and anti-aging effects. In the current study, pearl millet was fermented with Lactobacillus plantarum strains DHCU 70 and MCC 5231, which enhanced the nutritional, bioactive, and functional properties of derived probiotic beverages. Compared to unfermented controls, fermented beverages exhibited increased protein content and vitamins B1, B2, and B3, with decreased carbohydrate and dietary fiber levels. The probiotics have maintained viability exceeding 12 log CFU/mL and showed resistance to harsh gastrointestinal conditions. Fermentation increased total phenolic content from 13.38 ± 0.40 mg GAE/100 g to 42.10 ± 2.65 mg GAE/100 g (LPDB) and 47.76 ± 1.37 mg GAE/100 g (LPMB) and total flavonoid content from 13.01 ± 1.18 mg QE/100 g to 23.12 ± 2.73 mg QE/100 g and 24.21 ± 0.98 mg QE/100 g, respectively. Antioxidant assays showed DPPH radical scavenging improved by 37%, ferrous ion chelation rose from 71.69 ± 0.09 mg TE/100 g to 91.45 ± 0.006 mg TE/100 g, ABTS scavenging increased from 71.62 mg TE/100 g to 82.51 ± 0.04 mg TE/100 g (LPDB) and 89.74 ± 0.04 mg TE/100 g (LPMB) and superoxide radical inhibition rose from 51.40 ± 0.98% to 81.77 ± 0.03% (LPDB) and 79.92 ± 0.02% (LPMB). In the in vivo model, Caenorhabditis elegans, fermented beverage treatments significantly improved health-span parameters like head-swing frequency (13.51% increase), body bend frequency (8.41% increase), pharyngeal pumping (8.15% increase) with reduced lipofuscin accumulation and intracellular reactive oxygen species while median lifespan extended beyond 24 days versus 14–16 days in controls (p < 0.05). Gompertz mortality modeling revealed a significant decrease in the aging rate parameter, indicating systemic mitigation of stress-induced physiological decline. These combined nutritional, bioactive, and in vivo longevity results underscore the potential of L. plantarum-fermented pearl millet beverages as functional nutraceuticals that target oxidative stress and promote healthy aging. Pet humanization, particularly in dogs and cats, has transformed animal healthcare and highlighted the importance of nutrition in promoting human–pet social interaction, pet psychophysical well-being and, possibly, longevity. Nutraceuticals, such as omega-3 fatty acids, prebiotics, probiotics, plant extracts and dietary supplements, are endowed with antioxidant, anti-inflammatory, immune-modulating, cognitive-enhancing and gut-microbiota balancing properties. These effects have been shown to contribute to the possible prevention and management of bone and skin diseases, as well as gastrointestinal and behavioral disturbs. Moreover, the human–animal bond has been shown to play a pivotal role in reducing stress, improving sociability, and modulating pets’ emotional and physiological states. Evidence also suggests that nutrition and social interactions can influence the gut–brain axis, impacting the behavior, cognition, and resilience to stress-related disorders. Besides underlining the value of nutraceutical integration into pet nutrition strategies and offering a comprehensive, evidence-based perspective on their potential in improving animal welfare, literature reports about drawbacks of the use/misuse of such substances have been reported. The effect of varying sodium alginate (SA) concentrations (1%, 2%, and 3%; SA1–SA3) on the encapsulation of tangerine (Citrus reticulata Blanco ‘Cho Khun’) peel extract (TPE, 0.5% w/v) into hydrogel beads was evaluated. Overall, the results showed that increasing SA concentration significantly altered bead characteristics: lightness (L*) decreased from 56.35 to 45.57, red-green axis (a*) shifted negatively from −1.32 to −6.87, and yellow-blue axis (b*) increased from −17.81 to 6.41. Moisture content (97.85% to 93.16%) and water activity (0.96 to 0.93) declined with higher SA, while hardness increased (4.12 to 5.17 g). ζ-potential values shifted from −29.10 mV (SA1) to −39.10 mV (SA3), confirming enhanced electrostatic stabilization. FTIR spectra revealed characteristic alginate functional groups, and morphological analysis showed smoother, denser beads at higher SA concentrations. Phenolic (47.86–48.51 mg GAE g−1 DW) and flavonoid (34.02–36.68 mg QE g−1 DW) contents were well-retained, supporting antioxidant activities (DPPH 70.34–72.54%; ABTS 65.66–66.91%). Antimicrobial tests demonstrated > 4-log reductions against E. coli and P. aeruginosa. Sensory evaluation revealed that higher SA concentrations improved texture and taste. Overall, SA encapsulation, particularly at 3%, effectively stabilized TPE, preserving its functional properties for potential food and nutraceutical applications. Full article

The dairy sector produces considerable amounts of nutrient-rich effluents, which are frequently undervalued as simple by-products or waste. In particular, Second Cheese Whey (SCW), also known as scotta, exhausted whey, or deproteinized whey, represents the liquid fraction from ricotta cheese production. Despite its abundance and high organic and saline content, SCW is often improperly discharged into terrestrial and aquatic ecosystems, causing both environmental impact and resource waste. The available purification methods are expensive for dairy companies, and, at best, SCW is reused as feed or fertilizer. In recent years, increasing awareness of sustainability and circular economy principles has increased interest in the valorization of SCW. Biological treatment of SCW using microalgae represents an attractive strategy, as it simultaneously reduces the organic load and converts waste into algal biomass. This biomass can be further valorized as a source of proteins, pigments, and bioactive compounds with industrial relevance, supporting applications in food, nutraceuticals, biofuels, and cosmetics. This review, starting from analyzing the characteristics, production volumes, and environmental issues associated with SCW, focused on the potential of microalgae application for their valorization. In addition, the broader regulatory and sustainability aspects related to biomass utilization and treated SCW are considered, highlighting both the promises and limitations of microalgae-based strategies by integrating technological prospects with policy considerations. Lepidium meyenii Walp. (black maca, BM) is a traditional Andean crop increasingly studied for its bioactive potential. This work characterized the phytochemical profile and evaluated the antioxidant, antinociceptive, and neuroprotective properties of a lyophilized aqueous extract of BM hypocotyls. UHPLC-ESI-QTOF-MS/MS identified twelve major compounds, including macamides, imidazole alkaloids, sterols, and fatty acid amides. BM showed a moderate total phenolic content but strong electron transfer-based antioxidant activity in CUPRAC and FRAP assays, together with moderate radical scavenging capacity in ABTS and DPPH systems. In ovariectomized rats, BM significantly reduced brain malondialdehyde levels, mitigated oxidative stress, and improved spatial learning during acquisition in the Morris water maze, confirming its neuroprotective effect. Antinociceptive assays (hot plate, cold plate, and tail immersion) further revealed a rapid but transient increase in nociceptive thresholds. This study provides experimental evidence supporting the analgesic effect of black maca. Molecular docking highlighted lepidiline B and campesterol as key metabolites with strong interactions with redox enzymes, the μ-opioid receptor, and the FAAH enzyme, supporting their role in the observed bioactivities. ADMET predictions indicated favorable oral bioavailability, CNS penetration, systemic clearance, and acceptable safety profiles. These results substantiate the role of black maca as a neuroprotective nutraceutical and highlight its promise as a novel source of rapidly acting natural analgesic compounds. Full article

Fucoidan B (FucB) is a sulfated polysaccharide with recognized biological activity. In this study, FucB was chemically modified through redox conjugation with gallic acid (GA) to obtain FucB-GA, aiming to enhance its antioxidant properties. Structural characterization using FTIR, NMR, and electrophoresis confirmed the successful covalent binding of GA to FucB without major structural degradation. The conjugation increased the phenolic content and reduced crystallinity, as shown by XRD and SEM, indicating greater amorphous character, which can favor biological applications. Thermogravimetric analysis demonstrated enhanced thermal stability in FucB-GA. Antioxidant activity was evaluated through various in vitro assays. FucB-GA showed superoxide radical scavenging activity of 91.96%, copper chelating capacity of 43.2%, antioxidant capacity of 37 mg AEE/g, and reducing power of 94.22%, significantly higher results than FucB, while no sample chelated iron. Under the conditions analyzed, gallic acid alone showed minimal or no activity in most assays. These results suggest that conjugation with GA increases the antioxidant potential of FucB, while also improving the activity and bioavailability of GA, likely due to the increase in electron-donating and metal-binding groups. Overall, the study supports the development of FucB-GA as a promising antioxidant compound for pharmaceutical or nutraceutical applications. Full article

Mychonastes homosphaera MHSC24 is a green microalga newly isolated from a brackish coastal site in Korea. This study represents the first indigenous record of this species in the country. It provides a comprehensive characterization of its morphological, molecular, physiological, and biochemical characteristics. This microalga was identified through morphological observations and multilocus phylogenetic analyses. Strain MHSC24 exhibited robust growth under mesophilic temperatures (15–27 °C), moderate light intensities (88–300 μmol photons m−2 s−1), and low salinity levels (0–10 PSU). Optimal growth was observed at 27 °C, 193 μmol photons m−2 s−1, and 0 PSU. Under standard cultivation, the strain exhibited high protein levels (~54% of dry weight, DW) and accumulated substantial amounts of canthaxanthin (5.59 mg g−1 DW), the predominant carotenoid in its pigment profile. Thus, MHSC24 is a promising candidate for sustainable protein- and carotenoid-based applications. Palmitic acid (11.95 mg g−1 DW) and galactose (2.07 mg g−1 DW) were the predominant fatty acid and monosaccharide, respectively. The physiological resilience, high protein yield, and substantial canthaxanthin accumulation of MHSC24 support its potential utilization in the functional food, feed, and nutraceutical sectors. Therefore, this study provides a basis for optimized cultivation strategies and industrial exploitation of indigenous Korean microalgae. Full article

The agro-industrial residue known as hemp cake, derived from non-psychoactive Cannabis sativa L., represents a sustainable alternative for the development of protein-rich ingredients. In Ecuador, particularly in Bolívar Province, this by-product has been underutilized. However, similar challenges in the valorization of hemp residues have also been reported in other regions, where they are often discarded or used as low-value animal feed. These issues are not exclusive to Bolívar, and since protein stability depends primarily on drying and storage rather than geographic relocation, the valorization strategies proposed in this study can be extrapolated to other production zones. Protein concentrates were extracted from freeze-dried flower cake (TL, freeze-dried hemp cake) and oven-dried flower cake (TS, oven-dried hemp cake) using isoelectric precipitation, yielding protein concentrates from freeze-dried cake (CPL) and oven-dried cake (CPS). Protein content was determined using the Dumas combustion method, the Bradford dye-binding method, and the bicinchoninic acid (BCA) method. Functional properties such as solubility, water absorption, oil absorption, foaming capacity, and foam stability were evaluated, together with total phenolic and flavonoid content and in vitro antioxidant and anti-inflammatory activity. Results demonstrated high protein values (up to 90.42%), remarkable functional properties, and strong bioactive potential, supporting hemp cake concentrates as sustainable alternatives for food, nutraceutical, and pharmaceutical applications Full article
