Bovine viral diarrhea virus (BVDV) is a globally significant pathogen affecting both domestic livestock and wildlife, including white-tailed deer (WTD; Odocoileus virginianus). While experimental infections have demonstrated WTD susceptibility to BVDV, natural infections and associated reproductive outcomes remain scarcely documented. Here, we report the first confirmed case of naturally occurring BVDV-1 infection associated with fetal mummification in farmed WTD in Florida. A two-year-old doe experienced a stillbirth involving two mummified fetuses, which were submitted for necropsy and laboratory diagnostics. Gross findings included diarrhea and underdeveloped eyes in the fetuses, along with small white nodules indicative of amnion nodosum. While not harmful, this condition suggests underlying fetal compromise or intrauterine stress. Virus isolation using Vero E6 and bovine turbinate cell lines, along with a reverse transcription PCR (RT-PCR) assay specifically developed in this study, confirmed the presence of BVDV-1 (Pestivirus bovis) RNA in both maternal and fetal samples, suggesting vertical transmission. Sanger sequencing of RT-PCR amplicons further verified the virus species as BVDV-1. Differential diagnostics for other pathogens, including bluetongue virus, epizootic hemorrhagic disease virus, Mycobacterium spp., and Toxoplasma gondii, were negative. These findings underscore the importance of using biosecurity measures and including BVDV in the differential diagnosis of abortions to reduce the risk of BVDV transmission and potential outbreaks on deer farms, particularly those close to cattle operations. The molecular tools developed in this study provide a robust framework for improved detection and monitoring of BVDV in both wildlife and livestock populations. Full article

This study aimed to determine whether the fetuses experienced hypoxic distress or intra-partum death due to compromised oxygen supply by quantitatively analyzing the lung structures of deceased captive South China Tiger (Panthera tigris amoyensis, SCT) cubs. Furthermore, it investigated the etiology and mechanisms of asphyxia in the deceased cubs and explored potential factors contributing to the high mortality rate of SCT cubs. The research focused on three deceased SCT cubs, using three deceased neonatal Amur Tigers (P. t. altaica, AT) with no inbreeding background as controls. Histological analysis of the cubs’ lungs was conducted using hematoxylin–eosin (H&E) and special staining techniques. The possible causes of death in SCT cubs were evaluated by comparing pulmonary pathological changes and morphological parameters, including alveolar space (P(ASP)) values, mean alveolar linear intercept (Lm), and mean thickness of alveolar septa (Tas). Our research reveals that hypoxia is one of the most significant factors contributing to the mortality of tiger cubs. Additionally, we hypothesize that inbreeding depression may lead to abnormal lung development (e.g., thickening of lung tissue) in SCT cubs. This study lays the foundation for comprehensive investigations into the mortality of highly inbred SCT cubs. Full article

This study evaluated the performance of gestating and lactating sows supplemented with tributyrin from gestation until the end of lactation. Additionally, it assessed the effects of this supplementation on pig growth performance during the nursery phase. A total of 148 commercial-line sows (DB90 Brasil; parity 1–4; initial weight: 201.7 ± 12.2 kg) were used during gestation and lactation, along with 180 weaned pigs. A randomized block design was utilized with two dietary treatments during gestation and lactation, and a 2 × 2 factorial arrangement during the nursery phase. Data on sow body composition and productivity during gestation and lactation, as well as piglet performance during nursery, were collected. Treatment effects and interactions were analyzed using ANOVA with the MIXED procedure of SAS Version 9.0 (SAS Inst. Inc., Cary, NC, USA). Sows fed tributyrin tended to weigh 3.05 kg more (p = 0.053) and had 6.51% less backfat accumulation prepartum (p = 0.099). Supplementation reduced stillborns by 35.47% (p = 0.032) and increased litter weight at birth by 10.14% (p = 0.018). Additionally, there was a trend toward an 18.73% reduction in weight variability (p = 0.053) with more piglets weighing ≥1.4 kg. Sow and litter performance during lactation were not affected. Piglets from sows supplemented with tributyrin showed a trend for higher feed intake immediately post-weaning (p = 0.056). Tributyrin supplementation during gestation reduced stillborn rates and increased litter weight at birth. Full article

Background: Umbilical artery thrombosis (UAT) masquerading as a single umbilical artery (SUA) is a rare but critical diagnostic challenge in prenatal care. Case Presentation: We described a case of a 22-year-old primigravida with an uneventful obstetric history who presented with reduced fetal movements at 22 weeks of gestation. Ultrasound showed no gross fetal structural anomalies while umbilical artery Doppler flow imaging revealed an isolated SUA. The patient again presented with diminished fetal movement at 24 weeks gestation, and a diagnosis of intrauterine demise was confirmed ultrasonographically. She was then induced and delivered a macerated stillborn female fetus. Placental examination revealed three umbilical vessels with an occlusive thrombus seen within the umbilical artery consistent with UAT, a finding previously mistaken for SUA. Conclusions: This case underscores the diagnostic difficulties of UAT radiologically, especially when there was no prior documented evidence of two umbilical arteries. Identification of at-risk fetuses would allow for close monitoring or effective interventions to be implemented as early as possible to avert preventable fetal loss. Full article

Objective: To investigate the prevalence, risk factors, and complications associated with delivering macrosomic babies. Methods: Singleton term pregnancies (12,045) were studied. Macrosomia was categorized using the following two definitions: birthweight > 4 kg and birthweight ≥ 90th percentile (3.7 kg). Regression models were developed to identify significant risk factors for macrosomia such as maternal age, parity, pre-pregnancy body mass index, gestational weight gain, and hyperglycemia. Other models were constructed to identify the independent effect of macrosomia on outcomes such as shoulder dystocia, emergency cesarean section, stillbirth, and low APGAR scores. Results: The 50th centile birth weight of Saudi term infants is 3.18 kg; the 90th and 95th centiles were 3.70 and 3.91 kg, respectively. The prevalence of macrosomia (>4 kg) was 3.4%. The likelihood of macrosomia was associated with maternal age >40 years for newborns >4 kg, OR = 1.88, 95% CI (1.02–3.48), and maternal age < 18 for newborns ≥90th centile, OR = 5.23, 95% CI, (1.05–26.06). Regardless of the classification of macrosomia, it was associated with gestational age ≥41 weeks, parity > 4, pre-pregnancy BMI > 30, and maternal hyperglycemia. Macrosomia, using either definition, was associated with increased risk of shoulder dystocia, OR = 11.45, 95% CI (4.12–31.82) and OR = 9.65, 95% CI (3.89–23.94), and emergency CS, OR = 2.03, 95% CI (1.36–3.08) and OR = 1.77, 95% CI (1.34–1.52), for birthweight > 4 kg and ≥90th centile, respectively. Furthermore, newborns whose weights >4 kg were at greater risk to be stillborn, OR = 4.24, 95% CI (1.18–15.20), and to have low APGAR scores at birth, OR = 3.69, 95% CI (1.25–10.98). Conclusion: The risk of macrosomia among Saudi women significantly increases with maternal age, parity, gestational age, hyperglycemia, and pre-pregnancy obesity. Regardless of the definition used, delivering a macrosomic baby was associated with risks of shoulder dystocia and emergency cesarean section. Newborns (>4 kg) were at greater risk of stillbirth and low APGAR scores. Full article

This study evaluated the impact of precision feeding and bump feeding strategies during gestation on the reproductive performance of sows monitored over three cycles. Four treatments were compared: two constant-concentration feeding strategies (0.53% standardized ileal digestible lysine content; SID Lys) with the feed supply remaining constant (flat feeding; FF) or variable (bump feeding; BF) and two precision feeding strategies based on the InraPorc model considering performance by parity (precision feeding per parity; PFP) or the weight of each sow at breeding (precision feeding by individual; PFI). Sows were followed over three gestation and lactation cycles. In the first cycle (n = 502), the birth-to-weaning piglet mortality for PFP (8.7%) and PFI (10.3%) was lower than for BF (13.8%), with FF (11.3%) being intermediate (p = 0.001). No differences were observed in litter performance during the second cycle (n = 340). During the third cycle (n = 274), the stillborn rate was lower for PFP (6.2%) than for BF (9.1%) and FF (10.4%), with PFI (7.0%) being intermediate (p = 0.01). The BF strategy did not significantly improve sow or litter performance during lactation. Meanwhile, precision feeding could reduce nitrogen (10–13%) and total phosphorus intake (6–9%) with PFP and PFI strategies. Also, the results showed that it could even reduce piglet mortality during lactation. Full article

The aim of this study was to determine the optimal SID Arg: Lys ratio for maximizing the reproductive performance, immunity and biochemical parameters of sows during gestation, the colostrum composition, and the performance of their offspring. A total of 174 multiparous sows were randomly allocated to five treatment groups varying in dietary SID Arg: Lys ratios (0.91, 1.02, 1.14, 1.25 and 1.38) through modification of the levels of Arg or alanine supplementation (the total level of nitrogen was the same in all treatments). The results showed that increasing the dietary SID Arg: Lys ratio increased the number of piglets born alive (p < 0.05, linear and quadratic). The number of stillborn piglets, the birth weight variation of born alive piglets, the birth interval (p < 0.05, linear and quadratic) and the number of mummies (p < 0.05, quadratic) reduced with increasing the dietary SID Arg: Lys ratio. Broken-line regression analysis indicated that the optimal SID Arg: Lys ratio requirement for gestating sows to maximize the number of piglets born alive was 1.25. The content of non-fat solid, total solid, protein, and energy in colostrum increased linearly and quadratically (p < 0.05) with increasing dietary SID Arg: Lys ratio. Additionally, when increasing the dietary SID Arg: Lys ratio, the concentration of IgA (p < 0.05, quadratic) and IgM (p < 0.05, linear and quadratic) of plasma increased at day 90 of gestation; IgG (p < 0.05, linear and quadratic) concentration increased at day 110 of gestation of sows. The dietary SID Arg: Lys ratio had an increasing effect (p < 0.05, linear and quadratic) on plasma insulin levels at day 90 of gestation. Furthermore, there were increases in plasma concentrations of nitric oxide and ornithine at day 110 of gestation, Arg at day 90 and 110 of gestation (p < 0.05, linear and quadratic) and total protein at day 110 of gestation (p < 0.05, linear) with increasing dietary SID Arg: Lys ratio. The results of our study indicated that increases in the dietary SID Arg: Lys ratio during gestation resulted in an increase in the number of piglets born alive, a decrease in birth intervals, and an improvement in immunity and colostrum composition. The optimal SID Arg: Lys ratio for gestating sows to maximize the number of piglets born alive was 1.25. Full article

Oropouche virus (OROV) is an emerging arbovirus endemic in Latin America and the Caribbean that causes Oropouche fever, a febrile illness that clinically resembles some other arboviral infections. It is currently spreading through Brazil and surrounding countries, where, from 1 January to 1 August 2024, more than 8000 cases have been identified in Bolivia, Brazil, Columbia, and Peru and for the first time in Cuba. Travelers with Oropouche fever have been identified in the United States and Europe. A significant occurrence during this epidemic has been the report of pregnant women infected with OROV who have had miscarriages and stillborn fetuses with placental, umbilical blood and fetal somatic organ samples that were RT-PCR positive for OROV and negative for other arboviruses. In addition, there have been four cases of newborn infants having microcephaly, in which the cerebrospinal fluid tested positive for IgM antibodies to OROV and negative for other arboviruses. This communication examines the biology, epidemiology, and clinical features of OROV, summarizes the 2023–2024 Oropouche virus epidemic, and describes the reported cases of vertical transmission and congenital infection, fetal death, and microcephaly in pregnant women with Oropouche fever, addresses experimental animal infections and potential placental pathology findings of OROV, and reviews other bunyavirus agents that can cause vertical transmission. Recommendations are made for pregnant women travelling to the regions affected by the epidemic. Full article

Preterm births comprise all pregnancies coming to an end before the gestational age of 37 weeks and remain the leading cause of death in children under 5 years old despite efforts to reduce their occurrence. We aim to analyze all morbidity and mortality data to understand causes and risk factors, helping in prevention efforts. This study includes 140 cases collected during 2018–2022. Demographic, maternal, and thanatogenetic data were statistically analyzed. We observed an upward slope of stillborn babies. In the case of live-born premature, the average survival was 301.76 h. The multivariate analysis noted that extremely low birth weight (HR = 5.141) and very low birth weight (HR = 4.177) are risk factors involved in mortality. Increased parity was associated with premature births with low and very low birth weight (p = 0.019). We observed that a mother’s age of over 30 years is predictable for the development of pregnancy-induced hypertension. Cerebral and pulmonary hemorrhages were the most common intermediate morbid conditions, with prematurity and plurivisceral hemorrhages serving as their root causes. We have identified that anthropometric measurements have a high predictability on malformed babies. The identified associations indicate a shared mechanism for certain lesion processes, which can help optimize resources for predicting and preventing preterm neonatal issues. Full article

Background: Intrauterine fetal death and perinatal death represent one of the most relevant medical scientific problems since, in many cases, even after extensive investigation, the causes remain unknown. The considerable increase in medical legal litigation in the obstetrical field that has witnessed in recent years, especially in cases of stillborn births, has simultaneously involved the figure of the forensic pathologist in scientific research aimed at clarifying the pathophysiological processes underlying stillbirth. Methods: our study aims to analyze cases of sudden intrauterine unexplained death syndrome (SIUD) to evaluate the role of oxidative stress in the complex pathogenetic process of stillbirth. In particular, the immunohistochemical expression of specific oxidative stress markers (NOX2, NT, iNOS, 8-HODG, IL-6) was evaluated in tissue samples of placentas of SIUDs belonging to the extensive case series (20 cases), collected from autopsy cases of the University of Ferrara and Politecnica delle Marche between 2017 and 2023. Results: The study demonstrated the involvement of oxidative stress in intrauterine fetal deaths in the placenta of the cases examined. In SIUD, the most expressed oxidative stress markers were NOX2 and 8-HODG. Conclusions: The study contributes to investigating the role of oxidative stress in modulating different pathways in unexplained intrauterine fetal death (SIUD) tissues. Full article

It has been shown that vertical transmission of the SARS-CoV-2 strain is relatively rare, and there is still limited information on the specific impact of maternal SARS-CoV-2 infection on vertical transmission. The current study focuses on a transcriptomics analysis aimed at examining differences in gene expression between placentas from mother–newborn pairs affected by COVID-19 and those from unaffected controls. Additionally, it investigates the in situ expression of molecules involved in placental inflammation. The Papa Giovanni XXIII Hospital in Bergamo, Italy, has recorded three instances of intrauterine transmission of SARS-CoV-2. The first two cases occurred early in the pandemic and involved pregnant women in their third trimester who were diagnosed with SARS-CoV-2. The third case involved an asymptomatic woman in her second trimester with a twin pregnancy, who unfortunately delivered two stillborn fetuses due to the premature rupture of membranes. Transcriptomic analysis revealed significant differences in gene expression between the placentae of COVID-19-affected mother/newborn pairs and two matched controls. The infected and control placentae were matched for gestational age. According to the Benjamani–Hochberg method, 305 genes met the criterion of an adjusted p-value of less than 0.05, and 219 genes met the criterion of less than 0.01. Up-regulated genes involved in cell signaling (e.g., CCL20, C3, MARCO) and immune response (e.g., LILRA3, CXCL10, CD48, CD86, IL1RN, IL-18R1) suggest their potential role in the inflammatory response to SARS-CoV-2. RNAscope® technology, coupled with image analysis, was utilized to quantify the surface area covered by SARS-CoV-2, ACE2, IL-1β, IL-6, IL-8, IL-10, and TNF-α on both the maternal and fetal sides of the placenta. A non-statistically significant gradient for SARS-CoV-2 was observed, with a higher surface coverage on the fetal side (2.42 ± 3.71%) compared to the maternal side (0.74 ± 1.19%) of the placenta. Although not statistically significant, the surface area covered by ACE2 mRNA was higher on the maternal side (0.02 ± 0.04%) compared to the fetal side (0.01 ± 0.01%) of the placenta. IL-6 and IL-8 were more prevalent on the fetal side (0.03 ± 0.04% and 0.06 ± 0.08%, respectively) compared to the maternal side (0.02 ± 0.01% and 0.02 ± 0.02%, respectively). The mean surface areas of IL-1β and IL-10 were found to be equal on both the fetal (0.04 ± 0.04% and 0.01 ± 0.01%, respectively) and maternal sides of the placenta (0.04 ± 0.05% and 0.01 ± 0.01%, respectively). The mean surface area of TNF-α was found to be equal on both the fetal and maternal sides of the placenta (0.02 ± 0.02% and 0.02 ± 0.02%, respectively). On the maternal side, ACE-2 and all examined interleukins, but not TNF-α, exhibited an inverse mRNA amount compared to SARS-CoV-2. On the fetal side, ACE-2, IL-6 and IL-8 were inversely correlated with SARS-CoV-2 (r = −0.3, r = −0.1 and r = −0.4, respectively), while IL-1β and IL-10 showed positive correlations (r = 0.9, p = 0.005 and r = 0.5, respectively). TNF-α exhibited a positive correlation with SARS-CoV-2 on both maternal (r = 0.4) and fetal sides (r = 0.9) of the placenta. Further research is needed to evaluate the correlation between cell signaling and immune response genes in the placenta and the vertical transmission of SARS-CoV-2. Nonetheless, the current study extends our comprehension of the molecular and immunological factors involved in SARS-CoV-2 placental infection underlying maternal–fetal transmission. Full article

Mpox (monkeypox) is a neglected tropical disease that has received increased attention since the multi-nation outbreak that began in 2022. The virus is endemic in West and Central Africa, where the Democratic Republic of the Congo (DRC) is the most affected country. Clade I monkeypox virus (MPXV) infection is endemic in the DRC and has an overall case fatality rate of 10.6% among children and adults. A study conducted in Sankuru Province, DRC, from 2007 to 2011 demonstrated that 75% of pregnant women with mpox had miscarriages or stillbirth. Further analysis of a stillborn fetus showed that MPXV could infect both the placenta and fetus, causing congenital infection. No additional cases of Clade I MPXV in pregnant women were reported until a new outbreak occurred in South Kivu Province during 2023 and 2024. Eight pregnant women having Clade I MPXV infection were identified, of whom four had either miscarriages or stillbirth, representing a 50% fetal mortality rate. These reports confirm previous data from the DRC that indicate the capability of Clade I MPXV to affect the fetus, causing congenital infection and fetal loss in a high percentage of cases. In this article, we review both past and new data from the DRC on the effects of Clade I MPXV during pregnancy and discuss the association of mpox with fetal loss. Full article

The primary aim of this study was to investigate the effect of the pelvic dimensions of Holstein cows on the traction forces during parturition. Additionally, the relationship between calf measurements and traction forces was explored. For this purpose, a modified in vitro biomechanical model simulating obstetric tractions was used. For the requirements of the experiment, six bone pelvises of deceased Holstein cows were collected based on their estimated pelvic inlet area (EPA) and prepared. Additionally, six stillborn calves were collected based on their body weight (BW). The parameters of the pelvic inlet and cavity were measured using computed tomography (CT). Using the simulator, every calf was pulled in a random order through all pelvises, realizing a total of 36 obstetrical tractions, and the required forces were documented with appropriate software. In each extraction, three peaks of forces were recorded, with the first peak occurring at the entrance of the elbows into the maternal pelvic cavity, the second peak at the entrance of the thorax, and the third at the entrance of the calf’s pelvis. Logistic regression revealed an exponential relationship between pelvic parameters and traction forces for the entrance of the elbows and the pelvis, with the recorded forces being higher in the two smallest pelvises and stabilizing at a lower level thereafter, while for the entrance of the thorax, the correlations were either exponential or linear. The adjusted coefficients of determination (r2) were generally above the threshold of 0.5 for the entrance of the elbows and pelvis and lower (0.3–0.4) regarding the thorax and were statistically significant (p < 0.05) in all cases. Regarding the relationships between the calf dimensions and the required traction forces, the types of correlations were primarily linear and of lower magnitude. The combination of pelvic and calf parameters in a multivariate model resulted in an r2 of 0.72 for the entrance of the elbows using the pelvic diagonal and calf’s body weight, an r2 of 0.62 using the pelvic area and calf’s thoracic circumference, and an r2 of 0.75 using the pelvic diagonal and calf’s fetlock joint width. In conclusion, under the conditions of the present experimentation, the applied traction forces were mainly influenced by the pelvic dimensions in an exponential manner, whereas the calf body measurements showed a weaker effect. Based on these findings, critical cut-off points exist, different for every pelvic parameter, below which a significant increase in the required traction forces is expected. Full article

Previously, we found that a greater dissimilarity in swine leukocyte antigen (SLA) class I and class II alleles between mating partners resulted in increased farrowing rates in a highly inbred population of Microminipigs (MMPs). In this follow-up study, we have analyzed the effects of dissimilarity in SLA alleles between mating partners for seven different reproductive traits, including litter size and the number of stillborn and live or dead weaned piglets. We determined the relationships among reproductive traits within each mating event and the amino acid distances of SLA alleles as markers of diversity between mating partners. Our results indicate that mating partners with greater amino acid pairwise genetic distances in the SLA-1 class I gene or DQB1 class II gene alleles were associated with significantly larger litter sizes and higher numbers of live piglets at birth and weaning. Also, partners with greater pairwise distances in the SLA-2 class I gene alleles exhibited fewer pre-weaning deaths. These findings suggest that the dissimilarity in SLA class I and class II alleles between mating partners may affect not only farrowing rates but also other key reproductive traits such as litter size and improved piglet survival rates. Consequently, SLA alleles could serve as valuable genetic markers for selecting mating partners in breeding programs and for conducting epistatic studies on various reproductive traits in MMPs. Babesia gibsoni is a parasitic protozoan transmitted through tick bites and can cause severe disease in dogs. It can also be transmitted through direct contact with infected blood during dog fights, blood transfusions, and from dam to offspring during the perinatal period, resulting in stillborn or dead newborn puppies. This study aimed to determine the incidence of infection, the viability of newborn puppies, and the degree of B. gibsoni transmission from infected dam to offspring during pregnancy and lactation. Using PCR-based molecular methods, B. gibsoni infection in a pregnant American Pit Bull Terrier and her newborn puppies was confirmed. The incidence of B. gibsoni infection in the litter reached 75%. Out of eight puppies, six were infected with B. gibsoni, and one died. A therapeutic protocol comprising Malarone®, azithromycin, and artesunate was administered to a lactating B. gibsoni-positive bitch. By day 77 after birth, three out of five positive puppies showed negative PCR tests for B. gibsoni, indicating successful treatment through breast milk during nursing. In the two remaining positive puppies, therapy was started and parasitemia was successfully eliminated. Full article
